    Lymphoma is a kind of tumor that comes from lymphatic hematopoietic. According to the previous points of views, lymphoma is a kind of malignant tumor which proliferates monoclonally. The concept of “benign lymphoma” does not exist at present. In the clinical practice of recent years, we have found that some monoclonal lymphoid tissue lesions show benign biological behavior and their malignant potential is extremely limited and almost does not develop, so it presents a challenge to the concept of lymphoma and its diagnostic standard.     Expression changes and mechanism of Nod-like receptor protein 3 inflammasome in the cerebral microvascular endothelial cells in type 2 diabetes mellitus

    Objective To investigate the expression changes and mechanism of Nod-like receptor protein 3(NLRP3)inflammasome in cerebral microvascular endothelial cells of type2 diabetes mellitus. Methods A total of 3-month old Wistar rats(n=5)and spontaneous type 2 diabetic rats(GK, n=5)were enrolled. The expression of NLRP3 in the brain tissues was observed with immunohistochemical method. After cerebral microvascular endothelial cells(CMECs)were cultivated in vitro, they were divided into 5 groups: normal control group, high glucose group 1(HG1 group, glucose concentration=10 mmol/L), high glucose group 2(HG2 group, glucose concentration=20 mmol/L), high 山 东 大 学 学 报 (医 学 版)55卷3期 -李帅,等.Nod样受体蛋白3炎性体在2型糖尿病脑微血管内皮细胞中的变化及变化机制 \=-glucose group 3(HG3 group, glucose concentration=30 mmol/L), and HG+NAC group. The protein expressions of thioredoxin interacting protein(TXNIP)and NLRP3 were detected with Western blotting. The relationship between NLRP3 and TXNIP was analyzed. The content of interleukin-1β(IL-1β)was evaluated with ELISA kits. The content of intracellular reactive oxygen species(ROS)was measured with flow cytometry. Results NLRP3 gathered in microvascular endothelial cells. Compared with Wistar rats, GK rats had increased strength and number of vascular with positive staining(P<0.05). Compared with normal control group, HG1, HG2 and HG3 groups had higher expressions of TXNIP and NLRP3(P<0.01), especially in the HG3 group, in which the level of IL-1β and ROS also increased (P<0.01). TXNIP and NLRP3 was positively correlated(r=0.993; P<0.05). Lompared with HG3 group, in the HG+NAC group, the level of intracellular ROS, TXNIP, NLRP3 and IL-1β decreased(P<0.01). Conclusion NLRP3 inflammasome is activated in type 2 diabetes mellitus by ROS-TXNIP pathway. It promotes the release of inflammatory factor IL-1β and plays an important role in the microvascular injury.
    Regulatory effect of lncRNA RP11-543N12.1 on the expression of CDH13 in Alzheimers disease cell model

    Objective To explore the regulatory function of long noncoding RNA(lncRNA)RP11-543N12.1 on the expression of CDH13 in Alzheimers disease(AD)cell model. Methods Firstly, differently expressed lncRNAs and mRNAs were screened through microarray, and upregulated expressions of both RP11-543N12.1 and CDH13 were verified by real-time PCR. Secondly, SH-SY5Y cells were transfected with RP11-543N12.1-siRNA, and then Aβ25-35 was added to the transfected cells. After 48 h, the survival rate was detected with MTT; RNA expressions of RP11-543N12.1 and CDH13 were detected with real-time PCR; protein expression of CDH13 was determined with Western blotting; morphological changes were observed with Hoechst33258 staining; cellular apoptosis rate was detected with flow cytometry. Results Compared with control SH-SY5Y cells, cells treated with Aβ25-35 showed inhibited proliferation(P<0.05), upregulated RNA and protein expressions of CDH13(both P<0.01), and increased cellular apoptosis rate(P<0.01). Apoptotic nuclei were also observed. In cells transfected with RP11-543N12.1-siRNA and treated with Aβ25-35, cellular survival rate, RNA and protein expressions, and cellular apoptosis rate were all close to normal levels. Nuclei apoptosis disappeared. Conclusion LncRNA RP11-543N12.1 can regulate expression of CDH13, and knock-down of RP11-543N12.1 can downregulate the expression of CDH13.
    FOXM1 regulates transcription of SYK in breast cancer cell line MCF-7

    Objective To explore the underlying mechanism of Forkhead box M1(FOXM1)regulating spleen tyrosine kinase(SYK)expression in breast cancer cell line MCF-7. Methods PCR and Western blotting assay were used to analyze the mRNA and protein levels of SYK in MCF-7, T47D, SKBR3 and MDA-MB-231 cells. The methylation status of SYK promoter CpG island was detected. FOXM1 was overexpressed in MCF-7 cells, and then mRNA and protein levels of SYK and FOXM1 were detected. Chromatin immunoprecipitation(ChIP)assay and luciferase reporter assay were adopted to examine whether FOXM1 directly regulated the promoter activity of SYK. Results MCF-7 cells expressed endogenous SYK while T47D, SK-BR3 and MDA-MB-231 cells did not. The rate of DNA methylation of SYK in MCF-7 cells was lower than that in control cells(P<0.05). Overexpression of FOXM1 decreased SYK expression in MCF-7 cells and silence of FOXM1 in SK-BR3 cells increased SYK expression. ChIP assay indicated that FOXM1 directly regulated the transcriptional level of SYK gene. Knockdown of FOXM1 increased the basal SYK promoter activity and overexpression of FOXM1 decreased luciferase activity of SYK in MCF-7 cells. Conclusion SYK is a target gene of FOXM1 in MCF-7, while FOXM1 is involved in the genesis and development of breast cancer by directly regulating SYK.
    Susceptibility of mesenchymal stem cells to enterovirus 71 and differential expression of miRNA

    Objective To explore the functions of microRNA(miRNA)in Enterovirus 71(EV71)infection in human mesenchymal stem cells(MSCs). Mehods The EV71 infection model was established by using umbilical cord derived MSC in vitro, then the expression level of miRNAs was characterized via microarray platform and the related signaling pathways were revealed through bioinformatics analysis. Results Totally 84 differentially expressed miRNAs were found, mainly involved in the signaling pathway of the MAPK, Wnt and etc. Conclusion MSCs have a clear susceptibility to EV71, revealing that the EV71 infection can be regulated by using MAPK and Wnt signaling pathways via miRNAs.

    Objective To investigate the effects of norcantharidin(NCTD)on the expression of Notch signal pathway in multiple myeloma U226 cells. Methods U226 cells were co-cultured with NCTD at various concentrations in vitro. The proliferation inhibition was measured with CCK-8 test. The cell apoptosis rate was analyzed with flow cytometry. The mRNA and protein expressions of Notch2, Hes1, CyclinD1 and P21 in U266 cells were detected with qRT-PCR and Western blotting. Results NCTD could inhibit the proliferation of U266 cells and induce the apoptosis of cells in a time- and concentration-dependent manner. The mRNA and protein expressions(Notch2, P21)elevated with increased concentration of NCTD, while the expression of Hes1 and CyclinD1 decreased. Conclusion NCTD can inhibit the proliferation and induce the apoptosis of U266 cells, which may be related to the up-regulation of Notch2 and P21, and the down-regulation of CyclinD1 and Hes1.
    Effects of hydrogen-rich saline for lumbar intervertebral disc degeneration in rabbits

    Objective To investigate the effects of hydrogen-rich saline for lumbar intervertebral disc degeneration(LIVDD)in rabbits by intraperitoneal injection. Methods A total of 36 New Zealand white rabbits were randomly divided into 3 groups: normal control group, model control group and model experimental group, with 12 rabbits in each group. The lumbar discs of all rabbits were revealed from the right posterior lateral muscle gap. No further procedures were performed in the normal control group. Models of LIVDD induced by puncture and aspiration were established in the other 2 groups. Rabbits in the model experimental group were intraperitoneally injected with 8 mL/kg hydrogen-rich saline, once every 3 days. Equivalent saline was injected in rabbits of the model control group in the same way, while no treatment was done to the normal control group. The treatment lasted for 8 weeks. X-ray and histopathological examinations were conducted at the 4th and 8th week respectively after treatment. The contents of tumor necrosis factor(TNF-α), interleukin-1(IL-1)and matrix metalloproteinases-3(MMP-3)in the intervertebral disc tissue were detected with ELISA method. Results Significant improvements in imaging and histopathological data were found in the model experimental group after 4 or 8 weeks of hydrogen-rich saline treatment. The contents of TNF-α, IL-1 and MMP-3 in the experimental group were significantly lower than those in the model control group(P<0.001), but higher than those in the normal control group(P<0.001). The contents of inflammatory mediators over time had statistical difference in the model control group(P<0.001). Conclusion The intraperitoneal injection of hydrogen-rich saline in the treatment of LIVDD in rabbits plays a certain periodic positive role.

    Objective To investigate whether the expression of Sost gene was post-transcriptionally regulated in primary osteoblasts and MC3T3-E1 murine pre-osteoblast cells. Methods Osteogenic differentiation was induced in MC3T3-E1 cells and primary calvarial osteoblasts were isolated from C57BL/6 mice. The mRNA and protein levels of Sost gene in these cells were evaluated respectively. The 3'UTR of mouse Sost gene was amplified and subcloned into the SacI restriction site of pMIR-REPORT miRNA expression reporter, and the resulted plasmid was named as pMIR-REPORT-SOST 3' untranslated regions(UTR). Calvarial osteoblasts, MC3T3-E1 cells and NIH3T3 cells were then transfected with pMIR-REPORT-SOST UTR and pMIR-REPORT, respectively. Results Sost mRNA levels in calvarial osteoblasts and MC3T3-E1 cells were higher than control groups(P<0.05)during osteogenic differentiation. In contrast, the protein levels of Sost gene had no significant difference(P>0.05)with control group in calvarial osteoblasts and MC3T3-E1 cells. Relative luciferase levels transfected with pMIR-REPORT-SOST UTR had dramatically decline with transfected with pMIR-REPORT, respectively(P<0.05). In contrast, relative luciferase levels had no statistically significant differences between cells transfected with pMIR-REPORT-SOST UTR and pMIR-REPORT in NIH3T3 cells(P>0.05). Conclusion pMIR-REPORT-SOST UTR was successfully generated, and the expression of Sost gene was post-transcriptionally regulated via the 3'UTR.
    Inhibition of human apical papilla cells on the activation of mononuclear cell THP-1 and its molecular mechanism

    Objective To investigate the effects of human apical papilla cells(HAPCs)on the mononuclear cell line THP-1 activation and the molecular mechanism in vitro. Methods HAPCs and THP-1 were co-cultured in Transwell system, and then the mRNA expressions of TNF-α, IL-12, IL-10 and TGF-β1 in THP-1 were detected with Real-time PCR. The expression of TNF-α in THP-1 3d after co-culture was determined with flow cytometry. The phosphorylation of Stat3 and Smad3 were tested with Western blotting. Results HAPCs inhibited the mRNA expressions of TNF-α and IL-12 in THP-1 and promoted the mRNA expressions of IL-10 and TGF-β1(P<0.05). The expression of TNF-α significantly decreased in THP-1 3 d after co-culture(P<0.05). HAPCs activated the Stat3 pathway in THP-1 after co-cultivation(P<0.05), but had no effect on the Smad3 pathway(P>0.05). Conclusion HAPCs may inhibit the activation of THP-1 through the Stat3 pathway.
    Animal studies of the impact of initial stability on the immediate implantation success rate

    Objective To observe the influence of initial stability on HA coating, soft tissue level and root form implant success rate, in order to provide theoretical basis for clinical surgery. Methods The planting sites in adult mongrel dogs bilateral mandibular posterior tooth areas were randomly divided into experimental group(ISQ<50)and control group(ISQ>65). The implant ISQ value at the time of the implanting, week 4, 8 and 12 were measured and recorded. Meanwhile, CT scan was performed on each animal. The rates of implant osseoiniegration under light-microscopy were computed to observe the impact of different values of ISQ on immediate implantation success rates. Results The initial ISQ values of immediate implant BLB explants ranged 28-49, with corresponding implantation torque values of 10-20 N. There was no significant difference in immediate implantation success rate between the two groups. After 3 months, no implants loosed or dropped. CT scan showed no significant difference in bone density around the implant and implant-bone binding rates between the two groups(P>0.05). Conclusion The initial ISQ values of the immediate implant BLB explants range 28-49 or 68-75, with corresponding implantation torque values being 10-20 N or >30 N. There is no significant difference in the success rates.
    Correlation between porphyromonas gingivalis FimA genotype V and ginival inflammatory in the orthodontic patients

    Objective To explore the correlation between the gene expression of FimA genotype V of Porphyromonas gingivais(P.g)and gingival inflammatory in orthodontic patients. Methods Totally 61 patients with healthy gingiva were randomly selected. Their gingival crevicular fluid(GCF)was collected before and 3 months after orthodontic treatment. The content of P.g and FimA genotype V in the GCF was detected with PCR technique. Results The detection rate of P.g and FimA genotype V were 26.23% vs 73.77%, and 9.84% vs 26.23% before and after orthodontic treatment, showing significant difference(P<0.01, P<0.05). Conclusion Three months after patients wore appliance, the detection rate of P.g and FimA genotype V were positively correlated to gingival inflammation index. The detection rate of FimA genotype V rose with increased gingival index. FimA genotype V can cause gingival inflammatory in orthodontic patients.
    Bioinformatics analysis and construction of an eukaryocyte vector of ROP19 protein in Toxoplasma gondii

    Objective To construct and express the eukaryotic expression vector of ROP19 protein of Toxoplasma gondii PRU strain. Methods The physical and chemical characteristics of ROP19 protein were analyzed, and the B cell epitopes and T cell epitopes of ROP19 were compared with SAG1 using bioinformatics. The recombinant eukaryotic expression plasmid pROP19 was constructed by the molecular cloning technology. After being identified by PCR, restriction enzyme cleavage and sequencing, pROP19 was transfected into the HEK293T cells. The cells from different groups(control, pEGFP-C1 and pROP19)were respectively detected with fluorescence microscope under blue laser. The ROP19 protein was detected by Western blotting. Results The ROP19 protein was mainly located in the membrane and had better antigenic index than SAG1. The plasmid was constructed successfully. Western blotting showed that the expressed proteins could be recognized by anti-STAg mouse sera. Conclusion The eukaryotic expression vector pROP19 is constructed and expressed in eukaryotic cells.
    Nucleoprotein gene analysis of measles virus in Guizhou Province in 2013

    Objective To analyze the nucleoprotein genes of measles virus in Guizhou Province in 2013. Methods The throat swab, urine and sera samples were collected. Virus RNA was extracted directly from clinical specimens or cultured positive product of Vero/SLAM cell separation. Fragments of 634 nucleotide acids at the carboxyl end of the nucleoprotein gene were amplified by RT-PCR and then the PCR products were sequenced and analyzed. Results A total of 41 strains of measles virus were detected from 74 suspected measles cases. The results of phylogenetic analysis showed that 39 strains of measles virus were identified as H1a sub-genotype of H1 genotype, and located in two obvious branches. Two strains of them were identified as measles vaccine strain(A genotype). The nucleotide and amino acid divergences between 39 strains of measles virus strains and H1 genotype reference strain MVi/Hunan CHN/0.93/7(AF045212)were 2.0%-2.9% and 2.0%-4.7%, respectively. The nucleotide and amino acid divergences between 39 strains of measles virus strains and WHO H1a sub-genotype reference strain(Chin9322)were 0.9%-1.8% and 0.7%-3.4%, respectively. In addition, the nucleotide and amino acid homology among 39 MV strains of H1a sub-genotype were 97.3%-100% and 96.6%-100%, respectively. The nucleotide and amino acid homology between 2 strains of measles vaccine strains and S191 were 99.8%-100% and 99.3%-100%, respectively. Conclusion H1a sub-genotype was the predominant genotype in Guizhou Province in 2013, and there existed different chain of transmission in one year.
    Immunogenicity of the eukaryotic expression vector pcDNA3.1(-)-P6-gBCTL-TBK-1

    Objective To investigate the immune effects of herpes simplex virus type 2(HSV-2 )DNA vaccine of pc(pcDNA3.1-)-P6-gBCTL-TBK-1 in BALB/c mice. Methods The eukaryotic expression plasmid of pc-P6-gBCTL-TBK-1 was constructed and transfected into HeLa cells, and the expressed protein was verified by Western blotting. The BALB/c mice were immunized by the plasmids through intramuscular injection for three times, in week 0, 2 and 4. In week 5, the levels of interferon(IFN-γ), interleukin-18(IL-18), interleukin-4(IL-4), interleukin-6(IL-6)and specific HSV-2gD-P6 IgG antibody were measured by ELISA. In week 8, spleen cells of vaccinated mice were separated and the proliferation of spleen lymphocytes were detected by MTT. The activity of cytotoxic T lymphocyte(CTL)was detected by lactic dehydrogenase(LDH)method. Results After being inoculated by the plasmid of pc-P6-gBCTL-TBK-1, the mice were induced to generate higher level of specific antibody, including IFN-γ, IL-18 and IL-4; the CTL kill cells activity and proliferation capability of splenic lymphocytes were improved. Conclusion The recombinant plasmid of pc-P6-gBCTL-TBK-1 has immunogenicity, which can induce the humoral and cellular immune responses, and TBK-1 may have good function of molecular chaperone.
    Epidemiological characteristics of COPD patients attacked by respiratory system diseases during the flu season in Jinan City

    Objective To investigate the epidemiological characteristics of COPD patients attacked by respiratory system diseases in Jinan City during flu season and provide a reference for the health intervention and management. Methods According to cross-sectional study design, 948 COPD patients( ≥ 40 years)were selected and diagnosed by general hospitals which are at least Rank Two in Shizhong District, Huaiyin District and Tianqiao District during January 1 to October 31, 2013, as research objects. Self-designed questionnaire was used to investigate the incidence of respiratory system diseases in COPD patients during November 1, 2013 to March 31, 2014. The characteristics on incidences of research objects were descriptively analyzed. Results During the influenza season, a total of 450 in 948 COPD patients suffered from respiratory system disease, and the attack rate was 47.47%(450/948). The top three diseases were bronchitis(45.58%), upper respiratory tract infection(22.33%)and influenza(10.35%). Twice or more times of incidences accounted for 24.58%(233/948). The peak incidence of respiratory disease in COPD patients appeared in December. Conclusion The attack rate of COPD patients suffering respiratory disease during the influenza season was high. COPD patients should be well treated to prevent disease during flu season.

    Objective To investigate the expression of CK14 and its significance in the occurrence, development and metastasis of non-small cell lung cancer(NSCLC). Methods A total of 110 NSCLC patients were involved in the study. The NSCLC cancerous tissues, the tissues adjacent to carcinoma and normal lung tissues of 110 patients and 12 patients were analyzed for CK14 expression by immunohistochemistry and Western blotting, respectively. The relationship between CK14 expression and clinical pathological parameters of NSCLC patients was analyzed by non-conditional Logistic regression analysis. Results The immunohistochemistry results showed that CK14 was overexpressed in NSCLC cancerous tissues which was significantly higher than tissues adjacent to carcinoma and normal lung tissues(P<0.001). In addition, the positive rate of CK14 was 68.18%. Its level was significantly correlated with sex, histopathologic type, differentiation of tumors, clinical stages and lymph node metastasis(P<0.05). However, no correlation was observed between CK14 level and age(P>0.05). Additionally, Western blotting result also showed that CK14 was overexpressed in NSCLC cancerous tissues compared with the tissues adjacent to carcinoma and normal lung tissues. The results of non-conditional Logistic regression analysis showed that poor differentation, staging Ⅱ and Ⅲ, and the positive expression of CK14 were important risk factors in lymph node metastasis of NSCLC(P<0.05). Conclusion CK14 plays a promotional role in the development and metastatic progression of NSCLC. CK14 expression would be a significant biomarker in evaluating the biological malignant behavior of NSCLC.

    Objective To explore the pathogenic mechanism of thioredoxin domain containing protein 5(TXNDC5)involvement in rheumatoid arthritis(RA). Methods Human rheumatoid arthritis synovial cells(RASFs)were separated from RA patients. After the TXNDC5 expression was inhibited by anti-TXNDC5 siRNA, the total RNA was pooled, and the expressions of related genes were detected with PCR Array. The mRNA and protein expressions of related genes were verified using RT-PCR and ELISA. Results PCR Array analyses demonstrated that IGF-1, PCK1, SLC2A4, IL1R and IGFBP1 expressions significantly changed after TXNDC5 expression was inhibited. RT-PCR and ELISA verification indicated that IGFBP1 expression was significantly enhanced in both mRNA and protein levels(both P<0.001), while expressions of IGF-1, PCK1, SLC2A4 and IL1R1 showed no significant difference(P=0.792 7, P=0.713 0, P=0.452 9, P=0.354 2). No difference was observed in IGFBP3 expressions(P=0.511 5). Conclusion In RASFs, TXNDC5 is involved in RA by regulating the expression of IGFBP1.
    Expression and role of Keap1 in renal cell carcinoma

    Objective To investigate the expression of Keap1 and evaluate its role in renal carcinoma. Methods The total RNA of tumor tissues and adjacent tissues were isolated from 32 cases of renal carcinoma treated during May 2014 to April 2015 by TRIzol. The expressions of Keap1 and NFE2L2 were detected by Q-PCR technology and immunohistochemical staining. The changes of proliferation and colony forming ability of ACHN after overexpressed Keap1 were detected by CCK-8 cell proliferation and clone formation assay. Results Q-PCR results showed that the expression of Keap1 mRNA in adjacent tissues in 75% patients was higher than that in tumor tissues(P=0.000), the expression of NFE2L2 mRNA in tumor tissues in 62% patients was higher than that in adjacent tissues(P=0.046). The immunohistochemical results showed that the expression of Keap1 protein in adjacent tissues(87.50%)was higher than that in tumor tissues(25.00%)(P=0.000), and the expression of NFE2L2 protein in tumor tissues(81.25%)was higher than that in adjacent tissues(46.88%)(P=0.004). Overexpression of Keap1 decreased proliferation and colony forming ability of renal cell carcinoma cell line ACHN. Conclusion The abnormal expression of Keap1 in tumor tissues indicates that the Keap1-NFE2L2 pathway may play an important role in renal carcinoma. Keap1 has a probable inhibiting effect on the progression of renal carcinoma.

    Objective To explore the role of cancerous inhibitor of protein phosphatase 2A(CIP2A)in the malignant transformation of breast ductal epithelium and its association with the prognosis of breast ductal cancer. Methods Immunohistochemistry staining was used to detect CIP2A protein in 322 paraffin-embedded breast samples including normal breast duct, usual ductal hyperplasia(UDH), atypical ductal hyperplasia(ADH), ductal carcinoma in situ(DCIS)and invasive ductal carcinoma(IDC). Chi-square test was used to assess the difference of CIP2A expressions, and the correlation between CIP2A expression and the clinicopathologic characteristics. Kaplan-Meier method and Cox proportional hazard regression model were adopted to analyze the survival rate of patients. There was significant difference between UDH and DCIS groups, and UDH and IDC groups(P=0.004 3, P=0.004 0). Furthermore, CIP2A expression was closely associated with high histological grade and Ki67 index in DCIS samples(both P<0.05). In IDC patients, CIP2A expression was significantly correlated with CerbB2(Her-2)positive expression, high Ki67 index, high TNM stage and lymph node metastasis(all P<0.05). Patients with positive CIP2A expression exhibited significantly shortened 5-year disease-free and overall survival rate. CIP2A could be an independent prognostic indicator for IDC. Conclusion CIP2A plays a key role in the transition from proliferative lesions to ductal carcinoma. It holds the potential to monitor the malignant transformation of breast duct epithelium, and to guide the prognosis and personal treatment of patients with breast cancer.

    Objective To investigate the changes of hypothalamic-pituitary-gland axis function and metabolic manifestations of children with Prader-Willi syndrome(PWS). Methods A total of 17 PWS children diagnosed during Sept. 2012 and June 2016 were selected as the PWS group, and 35 age-and-sex-matched healthy children served as the control group. The differences of pituitary-gland hormone levels and related metabolic indexes were analyzed with t-test and Mann-Whitney U test. The growth hormone stimulation test and the stimulation of GnRHa were performed in the PWS group. Results In the PWS group, the height-SDS was -0.47±1.20; the weight was higher than the 90th percentile of normal children of the same gender and age; the BMI was 32.7±7.4 kg/m2, which was higher than 97th percentile of normal children of the same gender and age. Compared with the control group, PWS children had lower serum levels of growth hormone, insulin-like growth factor1(IGF-1)and free thyroxine(P<0.05). The free thyroxine in 1 child was <11.5pmol/L. Results of growth hormone stimulation test showed growth hormone deficiency. After the stimulation of GnRHa, the peak of luteinizing hormone in 12 children was <2.8 mIU/mL. The levels of total cholesterol, low-density lipoprotein cholesterol, triglyceride, apolipoprotein B, glycosylated hemoglobin, HOMA-IR, insulin and C-peptide in the PWS group were higher than those in the control group(P<0.05).     Objective To detect the value of plasma lipoprotein-associated phospholipase A2(Lp-PLA2)and the contrast-enhanced untrasonography(CEU)degree of neovascularization of carotid artery plaque in diagnosis of acute cerebral infarction(ACI). Methods A total of 44 patients with ACI(ACI group)and 41 patients without ACI(non-ACI group)were selected. The enrolled patients underwent conventional and CEU. The development intensity of neovascularization was observed and classified. The plasma Lp-PLA2 levels were detected by double antibody sandwich immunochromatography assay. Results There was significant difference between ACI group and non-ACI group in the grade of neovascularization of carotid plaques(Z=-5.29, P<0.01). The grade of neovascularization in ACI group was mostly Ⅲ and Ⅳ(median Ⅲ)and that of non-ACI group was mostly Ⅰ and Ⅱ(median Ⅱ). The plasma Lp-PLA2 levels in ACI group were significantly higher than those in non ACI group(t=6.64, P<0.001). With cutoff value 199.76 μg/L, the sensitivity, specificity, and area under the curve(AUC)were 84%, 68%, and 0.84, respectively. Conclusion As a noninvasive method, CEU can be used to observe neovascularization of carotid plaque in real time, and make a quantitative analysis. The higher grade of neovascularization of carotid plaque and the higher levels of the plasma Lp-PLA2 may be valuable for clinical diagnosis of ACI.

    Objective To evaluate the anesthetic effect of intrathecal injection of low dose bupivacaine with sufentanil during procedure for prolapse and hemorrhoids(PPH). Methods Sixty patients who underwent selective operation of PPH were randomly divided into Group BS and Group B, with thirty in each group. Group BS received intrathecal injection of bupivacaine(6 mg)and sufentanil(2 μg), namely, 0.75% bupivacaine(0.8 mL)and 0.002% sufentanil(0.1 mL). Group B received intrathecal injection of bupivacaine(10 mg)only, namely, 0.5% bupivacaine(2 mL). Patients discomfort when using anastomosis instruments on hemorrhoids mucosa was evaluated by VAS score. The degree of motor nerve block of lower extremities immediately after surgery was evaluated by revised Bromage score. The level of anesthesia and the performer satisfaction were evaluated. Postoperative micturition situations were recorded. Results Compared with Group B, the VAS and immediate postoperative revised Bromage scores were significantly decreased(P<0.001), the level of anesthesia were lower(P<0.001), and the performer satisfaction were risen(P=0.002)in Group BS. There was no significant difference in the postoperative micturition condition between the two groups(P=0.250). Conclusion Intrathecal injection of low dose bupivacaine with sufentanil is superior to bupivacaine alone for PPH.

    Objective To provide reference for clinical diagnosis by analyzing the occurrence and clinical characteristics of supernumerary teeth. Methods Clinical and imaging data of 558 cases of supernumerary teeth were retrospectively analyzed, including patients age and gender, the location, number, morphology, impaction of the teeth, and the concomitant symptoms and effects on adjacent teeth. Results There were a total of 804 supernumerary teeth in the 588 patients, whose average age was 11.92 years. The ratio between male and female patients was 2.99∶1. There were 743 supernumerary teeth in the maxilla(92.14%), and 61 in the mandible(7.58%). Most supernumerary teeth(75.37%)grew in the lingual or palatal side of jaws. Most patients had one supernumerary tooth, and some had two. Altogether 627 supernumerary teeth(77.98%)were impaction, and 97(12.06%)were complicated with dentigerous cyst. Odontoma was observed in 9 patients(1.61%). Conclusion Supernumerary tooth mainly occurs in children in mixed dentition and anterior maxilla. Men are more vulnerable to this lesion than women. Most patients have one or two supernumerary teeth.
