Role of glucagon-like peptide-1 on proinsulin mRNA levels in diabetic mouse

Abstract: Objective: To investigate the role of intravenous glucagon-like peptide-1 on blood glucose and on pancreas's proinsulin mRNA level in diabetic mice. Methods: Alloxan (45mg/kg) was infused by a tail vein in male Kunming mice to successfully make the diabetes model. Twenty diabetic mice were divided at random into a control group (normal sodium) and an experimental group[GLP-1(7-36)NH2]. Tail blood glucose was measured respectively before infusion of normal sodium[GLP-1(7-36)NH2] and at 5 and 10 minutes after infusion. Proinsulin mRNA levels were assessed by the semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR). Results: Blood glucose was not significantly different before intravenation between the control and the experiment groups(P>0.05); blood glucose concentrations at 5 and 10 minutes after intravenation were significantly decreased in the experiment group compared with the control group(P<0.05). The levels of proinsulin mRNA were significantly higher in the experiment group compared with the control group (P<0.05). Conclusion: Intravenous injection of GLP-1(7-36)NH2 can promote proinsulin gene transcripts, increase pancreas′s proinsulin mRNA level, stimulate insulin biosynthesis and secretion and quickly decrease blood glucose.
