Effects of ALA-PDT on Caspase-3 and Caspase-7 expressions in cutaneous squamous cell carcinoma A431 cells

Abstract: Objective To investigate the effect of aminolevulinic acid hydrochloride (ALA)-photodynamic therapy (PDT) on Caspase-3 and Caspase-7 expressions in cutaneous squamous cell carcinoma A431 cells. Methods A431 cells were cultured in vitro. The proliferation and apoptosis of A431 cells with different drug concentration, different incubation time, different light dose, as well as with drug and light alone, were assessed by MTT. Hacat was as control. The mRNA expressions of Caspase-3 and Caspase-7 were detected by Real-time PCR. The protein expressions of Caspase-3 and Caspase-7 in A431 cells and Hacat cells were detected by Western blotting. Results The mRNA and protein expressions of Caspase-3 and Caspase-7 were lower in A431 cells than in the Hacat cells with statistically significant difference (P<0.05). ALA-PDT inhibited the proliferation of A431 cells in a certain extent, and increased the mRNA and protein expressions of Caspase-3 and Caspase-7 (P<0.01). The inhibition on A431 cells could reach the maximum when the light dose was 80 J/cm2 with the same incubation time and concentration of ALA. Conclusion ALA-PDT can induce the expressions of Caspase-3 and Caspase-7 directly or indirectly, which induce the apoptosis of cutaneous squamous cell carcinoma A431 cells.
