Morphological changes and expressions of cleaved Caspase-3, PARP-1 in cerebellar fastigial nucleus of rats with vascular dementia

Abstract: Objective To observe the morphological changes and expressions of cleaved Caspase-3 and PARP-1 in the cerebellar fastigial nucleus of rats with vascular dementia (VD). Methods Thirty-two healthy Sprague-Dawley (SD) rats were randomly divided into two groups, sham group(n=8) and VD group(n=24). Morris water maze test was performed to evaluate the behavior of rats in sham group at week 1 and VD group at three time points(week 1, 2 and 4) after injury. The morphological changes of cerebellar fastigial nucleus were examined by Nissl staining. The expressions of cleaved caspase-3 and PARP-1 in the cerebellar fastigial nucleus were detected by immunohistochemical method. Results Compared with sham group, water maze escape latencies of VD group at three time points increased significantly (all P<0.01). Compared with sham group, the number of neurons in the cerebellar fastigial nucleus significantly decreased in VD groups (P<0.01). Compared with sham group, the expressions of cleaved Caspase-3 and PARP-1 in VD group significantly increased in the cerebellar fastigial nucleus (P<0.01). Conclusion The cerebellar fastigial nucleus is damaged in VD rats, while the cleaved caspase-3 and PARP-1 play an important role in this process.
