Protective effect of riluzole on the damage of rat retina induced by acute intraocular hypertension

Abstract: Objective To investigate the protective effect of riluzole on the damage of rats' retinal ganglion cells induced by acute intraocular hypertension. Methods A total of 120 SD rats were randomly divided into control group, high intraocular pressure group, and riluzole intervention group, 40 in each group. Acute ocular hypertention rats model, which were established using the anterior chamber perfusion way, were injected intraperitoneally with riluzole. After 12, 24, 48 and 72 h of reperfusion, the expression of Caspase-3 protein and the levels of apoptosis in each layer of rats' retinal cells were detected by immunohistochemistry and terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL). The structural changes of the rats' retina in each group were observed by HE staining. Results Compared with the high intraocular pressure group, the thickening and structural disorder in riluzole intervention group were relieved, and the expression of Caspase-3 and the apoptosis cells at different time points in riluzole intervention group decreased (P< 0.05). Conclusion Riluzole can reduce the apoptosis of retinal cells in rats with damaged retina caused by acute ocular hypertention and has significant protective effects to retina.
