A new method of inducing adipose-derived stem cells into cartilage tissue

Abstract: Objective To rexplore a new method of inducing adipose-derived stem cells (ADSCs) into cartilage tissue. Methods ADSCs were isolated from rat adipose tissues and characterized by cell-surface markers CD90 and CD44. The passage-6 cells were seeded in alginate beads and induced into chondrocytes in medium containing TGF-β1. After 2 weeks of culture, the cells in alginate beads were released and induced for another 2 weeks. After the induced tissues were digested by type Ⅱ collagenase, the cells were harvested and identified, and expression of type Ⅱ collagen protein was detected by cell immunohistochemistry. Results Adipose-derived stem cells exhibited a fibroblast-like morphology and were positive for typical MSC-related markers CD44 and CD90. The expression of typeⅡcollagen protein in cells of induced tissues showed that it was a new method of chondrogenic engineering to induce adipose-derived stem cells released from alginate. Conclusion Chondrogenic differentiation of cells released from ADSCs-alginate beads after 2 weeks of chondrogenic inducing is a new method of cartilage tissue engineering.
