    The effect of resistin-like molecule β on the vulnerability of atherosclerotic plaques in ApoE-/- mice

    Objective To explore the effect of resistin-like molecule β(RELMβ)on the vulnerability of atherosclerotic plaques in ApoE-/- mice. Methods A total of 60 male ApoE-/- mice(8 weeks old)were fed with a high-fat diet and a constrictive silastic tube was placed to induce the vulnerable plaques. All mice were divided into 3 groups after surgery(n=20 per group): control, si-NC and si-RELMβ. At the end of week 14, ApoE-/- mice were euthanized to collect the right common carotid arteries and blood from the left ventricle. The contents of total cholesterol, triglycerides, low-density lipoprotein cholesterol and high-density lipoprotein cholesterol in the blood samples were detected. The protein expressions of RELMβ in all mice were determined with Western blotting. The collagen and lipoid depositions in the plaques were determined with collagen sirius red staining and oil-red O staining, respectively. The contents of macrophages, smooth muscle cells(SMCs), interlenkin-1 beta(IL-1β)and interlenkin-6(IL-6)expressions in the plaques were detected with immunohistochemical methods. Results There were no significant differences in body weight and lipid levels among the 3 groups. Compared with the control group, the si-RELMβ group had significantly decreased RELMβ protein expression, lipid, macrophages, vulnerable index, and expressions of IL-1β and IL-6, increased contents of collagen and SMCs(P<0.05), while those in the si-NC group had no statistical differences. Conclusion RELMβ can increase atherosclerotic plaque instability, and its mechanism is involved with the elevated expressions of IL-1β and IL-6.
    Effects of oxygen with different concentrations on the expressions of S100β protein and neuron-specific enolase after cardiopulmonary resuscitation in rats

    Objective To investigate the effects of oxygen with different concentrations on the outcome of cardiopulmonary resuscitation(CPR)in rats. Methods Thirty-two SD rats were randomly divided into the control group and resuscitation group, which was subdivided into 21% oxygen concentration group, 50% oxygen concentration group and 100% oxygen concentration group, with 8 in each group. Anesthesia, tracheal intubation, and vascular puncturation were performed for the control group. Other groups were given 21%, 50% and 100% oxygen respectively. ELISA assay was used to detect the levels of of serum S100β protein and neuron-specific enolase(NES)at 0.5, 3, and 6 h after successful CPR. Results Compared with the control group, the level of serum S100β protein in resuscitation group was higher(P<0.05)and reached the peak at 3h time point, while the level of serum NSE rose stably with the increase of time(P<0.05). After a successful resuscitation, the risen levels of S100β protein and NSE were as follows: 21% oxygen concentration group > 100% oxygen concentration group > 50% oxygen concentration group(P<0.05). Conclusion Compared with 21% and 100% oxygen, 50% oxygen inhalation can decrease the levels of serum S100β protein and NSE after a successful CPR, minimize secondary brain injury and thus improve the brain functionality.
    Effect of Helicobacter pylori infection on NLRP3 inflammasome activation

    Objective To study the impact of Helicobacter pylori(H.pylori) infection on NLRP3 inflammasome associated proteins expression and activation in gastric epithelial cells and gastric cancer cells, and further discuss the role of NLRP3 inflammasome when gastric cells were infected by H.pylori. Methods The gastric epithelial cell line GES-1 and gastric cancer cell line MKN-45 were infected by H.pylori NCTC11639 at a multiplicity of infection(MOI)of 100 or 200. At 6 h or 12 h after infection, the cells were harvested and the total proteins were extracted. Then the expressions of NLRP3 and Caspase-1 were analyzed by Western blotting and the level of IL-1β secretion was detected by ELISA. Results The expressions of NLRP3 and Caspase-1 and the secretion of IL-1β were increased with the increase of NCTC11639 MOI in GES-1 cells(P<0.05). Although the expression of NLRP3 was increased, the expression of Caspase-1 and the secretion of IL-1β were decreased with the increase of NCTC11639 MOI in MKN-45 cells(P<0.05). In addition, the expressions of NLRP3 and Caspase-1 and the secretion of IL-1β were not significantly different in GES-1 or MKN-45 infected by H.pylori for 6 h and 12 h with the same MOI(P>0.05). Conclusion H.pylori infection promotes the expressions and activations of NLRP3, Caspase-1 and IL-1β in gastric epithelial cells. However, H.pylori infection facilitates the expression of NLRP3 but inhibits the expressions and activations of Caspase-1 and IL-1β in gastric cancer cells.

    Objective To explore the molecular mechanism of c-Met expression in hepatocellular carcinoma cells(HCC)influenced by tumor associated macrophages. Methods HCC cells were cocultured with macrophages derived from THP-1 cells. The expression of c-Met in HCC cells was detected at mRNA level by RT-PCR and protein level by flow cytometry. The migration of HCC cells was detected by Transwell cell migration test. Results The expression of c-Met in HCC cells was upregulated by the macrophages at both mRNA and protein levels. After HCC cells were cocultured with macrophages, the migration was enhanced in the presence of human recombinant protein hepatocyte growth factor(HGF). Conclusion Tumor associated macrophages promote the migration of HCC cells by upregulating the expression of c-Met in HCC cells.

    Objective To explore the effect of propofol on carbachol(CCh)-induced chloride secretion in rat ileum mucosa. Methods Cch-induced chloride secretion was measured by Ussing chamber technique. Results Propofol attenuated the CCh-stimulated increase in short-circuit current(Isc)of rat ileum mucosa in a dose-dependent manner(10-2-10-6mol/L). The propofol-evoked inhibitory effect was serosal side-dependent. Furthermore, both TTX, a Na+ channel blocker, and hexamethonium bromide, a nicotinic acetylcholine receptor antagonist, had no effect on propofol-evoked inhibitory effect. Conclusion Propofol inhibites CCh-induced intestinal secretion via basolateral mAChR pathway.
    CXXC4 promotes sensitivity to chemotherapy drugs in epithelial ovarian cancer cells

    Objective To explore the effect of CXXC4 on the proliferation and sensitivity to chemotherapy drugs in ovarian carcinoma cells. Methods Protein and mRNA levels of CXXC4 were detected in nomal ovarian epithelial cells and ovarian carcinoma cells by Western blotting and Real-time PCR, respectively. The recombinant plasmid pcDNA3-CXXC4 and siRNA-CXXC4 were transfected into SKOV3, Caov3 cells and IOSE80 cells via Lipo2000. CCK8 assay was used to evaluate the effect of CXXC4 on the proliferation and sensitivity to chemotherapy drugs. Results Compared with the normal ovarian epithelial cells, CXXC4 expression was lower in ovarian carcinoma cells. Overexpression of CXXC4 inhibited the proliferation capacity and increased the sensitivity to chemotherapy drugs in SKOV3 and Caov3 cells. Decreased CXXC4 expression could promote the proliferation capacity and the chemo-resistance to chemotherapy drugs of normal ovarian epithelial cells. Conclusion Overexpression of CXXC4 could inhibit the proliferation capacity and increase the sensitivity to chemotherapy drugs in ovarian carcinoma cells, suggesting it would be a potential biomarker to predict the efficacy or a molecular target to enhance the efficacy to chemotherapy in ovarian carcinoma cells.

    Objective To determine the crucial enzymes of glucose metabolism in rheumatoid arthritis(RA). Methods The glucose metabolism in synovial tissues of collagen-induced arthritis(CIA)was analyzed with Rat Glucose Metabolism RT2 ProfilerTM PCR Array. The result was verified using real time PCR, Western blotting and ELISA in synovial tissues and blood from patients with RA. Results The PCR array detected up-regulation of ENO1, HK2 and PGK1, and down-regulation of PCK1 and PDK4 in synovial tissues of CIA rats. Real time PCR and Western blotting detected increased expressions of ENO1 and PGK1 in synovial tissues of RA. ELISA detected a high level of PGK1 in blood of RA patients. The result identified the increased expression of ENO1 and PGK1 in CIA and RA synovial tissues, and elevated level of PGK1 in blood of the patients with RA. Conclusion Glycolytic activity is increased in RA, which may result in the abnormal expression of genes involved.

    Objective To observe the effects of different doses of bisphenol A(BPA)on the bone mineral densities(BMDs)of vertebrate and femoral bone in female F344 rats. Methods A total of 40 female F344 rats aged 4 weeks, bodyweight(53.90±4.23)g, were randomly divided into 5 groups: control group, BPA50 group[50 mg/(kg·d)], BPA200 group[200 mg/(kg·d)], BPA400 group[400 mg/(kg·d)] and estradiol benzoate(EB)group[1 mg/(kg·3 d)], with 8 rats in each group. BPA(diluted into corn oil)was given via gavage, EB was given via intraperitoneal injection, and the same volume of corn oil was given via gavage to the control group. The changes of mental state, body hair and bodyweight were recorded. The BMDs of vertebrate and femoral bone were measured after 13 weeks’ exposure by dual-energy X-ray absorptiometry(DEXA). Results The bodyweight of the BPA200 group was the lowest(P=0.003); the bodyweight of EB group was the highest(P=0.014); the bodyweight of the BPA400 group was significantly lower than that of the control group(P=0.015). Conclusion A certain doses of BPA can significantly reduce the bodyweight and the BMDs of vertebrate and femoral bone in female F344 rats.

    Objective To optimize the prescription of self-microemulsifying drug delivery system for quercetin(QT-SMEDDS)by central composite design-response surface method and prepare QT-SMEDDS. Methods Components of prescription was screened by measuring the solubility of quercetin in different secondary solvents, investigating the phase instability and plotting pseudo-ternary phase diagram. According to the balanced solubility and particle size of quercetin in different self-microemulsifying drug prescriptions, the optimal prescription was determined using central composite design-response surface methodology. The particle size and Zeta-potential were investigated in detail. Results The optimal prescription of quercetin in SMEEDS was composed of Labrafil M1944CS(oil), Cremophor EL(surfactant), and Transcutol HP(co-surfactant), with a weight ratio of 27.0%∶55.6%∶17.4%. The balanced solubility was 67.87 mg/g, the average particle size diluted 50 times with water was 25.26 nm, and Zeta-potential was -6.73 mV. Conclusion The optimized prescription of QT-SMEDDS is obtained and its quality is stable.

    Objective To study the chemical constituents from the aerial parts of Litsea glutinosa(Lour.)C.B. Rob. Methods The obtained compounds were isolated by different chromatographic methods, including silica gel, Sephadex LH-20 and high performance liquid chromatography, and then their structures were elucidated on the basis of spectroscopic analyses, particularly the nuclear magnetic resonance spectroscopy. Results Five compounds were isolated from the EtOAc partition of the 95% EtOH extract of the aerial parts of Litsea glutinosa, and their structures were identified as pubinernoid A(I), vomifoliol(II), atroside(III), 1,2-dihydro-6,8-dimethoxy-7-1-(3,5-dimethoxy-4-hydroxyphenyl)- N1,N2-bis-[2-(4-hydroxyphenyl)ethyl]-2,3-naphthalene dicarboxamide(IV), and daucosterol(V). Conclusion Compounds I-IV are isolated from Litsea glutinosa for the first time, and compounds I-III are obtained from the genus of Litsea for the first time.
    The value of multiphase dynamic reconstruction with CTA to the intimal flap motion of abdominal aortic dissection

    Objective To explore the value of low dose of multiphase dynamic reconstruction using retrospective ECG gated CTA to the intimal flap motion of abdominal aortic dissection. Methods Thirty patients with abdominal aortic dissection underwent with retrospective ECG gated double source CT(DSCT), and 20 datasets were reconstructed in 5% steps between 0 and 95% of the R-R interval in each case. The short axis diameter of the true lumen and abdominal aorta in the level of the root of celiac and superior mesenteric artery, left renal artery and right renal artery in each phase were measured, and the ratio of both(d-T/d-AA)was calculated. The ratio of the minimum and maximum as well as 70% R-R intervals was calculated and the three statistical difference for evaluating the intimal flap indirectly was compared. Results The scope of intimal flap was strong in the level of celiac and superior mesenteric artery, and intimal flap was significantly changed between 70% R-R, the maximum and the minimum. Intimal flap was significantly changed between the maximum and minimum in the level of right renal artery and left renal artery.     Objective To analyze the data of distal embolization(DE)occurred during interventional therapy(IT)of lower extremities artery occlusion, and summarize its rules and treatment. Methods One hundred and twenty six patients with lower extremities artery occlusion who were performed IT were included from October 2011 to May 2014 in Shanxi Provincial Peoples Hospital. DE occurred in six patients; three of whom received catheter-directed thrombolysis, two received thrombectomy and one received observation. Results The six patients had no serious consequence and acquired follow-up. During follow-up, DE didnt cause the adverse impact on the early, middle and later therapy effect. Conclusion Careful film reading before IT and careful manipulation during IT can reduce the incidence of DE. If DE happens, the correct and positive treatment can minimize its harmfulness according to the embolism location and clinical presentation.

    Objective To observe the efficacy of patients with intermediate carpal tunnel syndrome(CTS)treated with oral medicine, local sealing and surgery. Methods CTS patients were divided into 3 groups according to the principle of complete voluntary: oral drug treatment group(56 cases, 85 sides), locally closed treatment group(75 cases, 108 sides), surgical treatment group(68 cases, 96 sides). After CTS patients were treated with oral medicine, local sealing or operation, the characteristics of high-frequency ultrasound and electrophysiological changes were observed. Results The effective rate of surgery was higher than that of local sealing and oral medicine. There were significant differences in the cross-sectional area of the median nerve in pisiform bone and hamate hook, and the ratio of transverse diameter and anteroposterior diameter of the hamate hook between patients with poor and good therapeutic efficacy. Even patients with poor efficacy had improved electrophysiological changes. Conclusion Surgical treatment is the most effective treatment for CTS. Larger cross-sectional area of the median nerve in pisiform bone and hamate hook, and larger ratio of the transverse diameter and anteroposterior diameter of the hamate hook mean poorer prognosis. Patients with ineffective surgical treatment still show significant electrophysiological changes after treatment.
    Clinical application value of the new guide for patients with chronic obstructive pulmonary disease

    Objective To investigate the clinical significance of the clinical classification of the Spanish new clinical guidelines in chronic obstructive pulmonary disease(COPD)in 2013. Methods A total of 640 cases who had been diagnosed as COPD and hospitalized in the department of respiratory medicine of Anqing Municipal Hospital Affilicated to Anhui Medical University were collected from January 2009 to December 2014. According to the typing method of Spanish guidelines in 2013, the 640 cases were divided into four groups named A, B, C and D respectively. The pulmonary function, index of analysis of blood gas, morbidity of complication, the usage of antibiotics and hormone of each group were analyzed and compared. Results Pulmonary function forced expiratory volume in one second(FEV1)and forced expiratory volume in one second/forced vital capacity(FEV1%)in groups C and D were significantly lower than those in groups A and B(P<0.05). There was no significant difference in each group about the morbidity of diabetes and hypertension(P>0.05), while the incidence of coronary heart disease, arrhythmia and heart failure in groups C and D were significantly higher than those in groups A and B(P<0.05), and in group D higher than in group C(P<0.05). The usage time of antibiotics in groups A and B were significantly lower than that in groups C and D(P<0.05), and in group D was obviously higher than in group C(P<0.05). The use amount of hormone in group B was higher than that in groups A, C and D(P<0.05), and in groups C and D higher than in group A(P<0.05). In all groups, hospitalization time in group D was longest and shortest in group A.     Objective To evaluate the clinical significance of serum miR-375 in the diagnosis and postoperative monitoring of cervical cancer by analyzing its expression in cervical intraepithelial neoplasia(CIN)and cervical cancer. Methods A total of 178 subjects, including 64 patients with cervical cancer, 64 patients with CIN and 50 healthy controls, were enrolled in the study. The expressions of serum miR-375 and CA125 were measured, and their diagnostic significance was evaluated. The relationship between miR-375 and clinical parameters of cervical cancer was analyzed. Results The expression of serum miR-375 in patients with cervical cancer was significantly lower than that in patients with CIN and healthy controls(both P<0.05). The level of serum miR-375 in CIN patients was significantly lower than that in healthy controls(P<0.05). Receiver operating characteristic(ROC)curve analysis showed that the area under the ROC curve(AUC)for serum miR-375 was 0.782(95% CI: 0.714-0.840; sensitivity, 73.4%; specificity, 78.9%), which was significantly higher than that of CA125[0.677, 95%CI: 0.603-0.745], P<0.05. The expression of serum miR-375 was significantly up-regulated after surgery in the same subset of patients with cervical cancer(P<0.05). Moreover, serum miR-375 in patients with cervical cancer was significantly correlated with FIGO stages and lymphoid node metastasis(both P<0.05), while there was no significant association between serum miR-375 and 山 东 大 学 学 报 (医 学 版)54卷3期 -彭慧,等.血清循环miR-375检测对宫颈癌早期诊断及术后监测的意义 \=-age, histological type or cell differentiation(all P>0.05). Conclusion Serum miR-375 is conducive in the diagnosis of cervical cancer and CIN. It can serve as an index for the postoperative monitoring, progression and lymphoid node metastasis evaluation of cervical cancer.
    Effects of rosiglitazone and GW9662 on the expression of PPAR-γ in JEG-3 cells

    Objective To investigate the effects of peroxisome proliferator-activated receptor gamma(PPAR-γ)agonist rosiglitazone and antagonist GW9662 on PPAR-γ expression and invasion ability of JEG-3 cells. Methods Immunohistochemistry method was used to detect PPAR-γ expression and distribution in normal chorionic, hydatidiform mole and choriocarcinoma tissues. Then PPAR-γ expression in rosiglitazone, GW9662 and control groups were examined respectively by Transwell assay, immunocytochemistry, Western blotting and qPCR method. Results PPAR-γ was expressed and distributed in cell nucleus of normal chorionic, hydatidiform mole and choriocarcinoma tissues, and its expression was significant down-regulated in choriocarcinoma tissues. Invasion ability of JEG-3 cells treated by rosiglitazone was decreased, while that of GW9662 group was increased. There was a statistical significance between the two groups(P<0.05). The expression of PPAR-γ in rosiglitazone group was lower than that in GW9662 group(P<0.05). Conclusion The activity of PPAR-γ is modulated via a negative feedback regulation mechanism by rosiglitazone and GW9662, and can influence the invasion ability of choriocarcinoma cells.
    Relationship between mir-99a expression and prognosis of epithelial ovarian cancer

    Objective To investigate the mir-99a expression in epithelial ovarian cancer(EOC), and its relationship with the clinicopathological features and prognosis. Methods The mir-99a expression in 137 cases of EOC tissues was detected with qRT-PCR assay. The correlation between mir-99a expression and clinicopathological features of EOC were analyzed. Survival curves for patients with different mir-99a expressions were plotted using the Kaplan-Meier method and compared using the log-rank test. Results The mir-99a expression reduced significantly in EOC tissues compared with that in adjacent normal tissues(P<0.001). Low mir-99a expression was significantly correlated with higher histological grade(P=0.027), lymph node metastasis(P=0.001), peritoneal metastasis(P<0.001), and higher FIGO stage(P<0.001). Kaplan-Meier analysis with log-rank test revealed that low mir-99a expression was correlated with short survival time of patients with EOC(log-rank test, P=0.005). Multivariate analysis showed that mir-99a expression was an independent prognostic factor for the overall survival(HR: 2.368, 95%CI: 1.621-10.925, P=0.016). Conclusion The mir-99a expression is an independent prognostic factor of patients with EOC. It can serve as a diagnostic and prognostic marker in clinical practice.
    Comparative study of gemstone spectral CT and conventional ultrasound spectroscopy for the diagnosis of benign and malignant thyroid nodules

    Objective To explore the value of the gemstone spectral CT imaging in differential diagnosis of benign and malignant thyroid nodules,and compare its diagnostic value with ultrasonography. Methods Forty-four thyroid nodules were analyzed retrospectively. All patients underwent gemstone spectral CT scanning, arterial phase enhanced CT imaging and ultrasonography. According to the pathology results, nodules were divided into benign group and malignant group. Data of spectral CT imaging and ultrasonography were analyzed by independent sample t test or nonparametric test and multivariate Logistic regression analysis respectively. The optimal iodine concentration threshold to predict malignancy was obtained by receiver operating characteristic(ROC)curve, and the sensitivity and specificity were achieved. Results A total of 44 nodules were detected, including 23 benign and 21 malignant nodules. Between the two groups, there was difference of the average iodine concentration, standardized iodine concentration, and slope of spectral curve in non-contrast CT imaging(P<0.05), while there was no difference in the arterial phase(P>0.05). The optimal iodine concentration to predict malignancy was 0.32×100 μg/mL in non-enhanced phase with 100% sensitivity and 79.5%specificity. The nodules form, border, cystic situation between the two groups were statistically different in ultrasonography with multivariate logistic regression analysis(P<0.05). Conclusion Gemstone spectral CT imaging in non-contrast phase has potential value in diagnosis of benign and malignant thyroid nodules, and is better than that of ultrasonic in diagnosis of the nodules whose diameter great than or equal to 1 cm.
    Efficacy of rhombic subauricular flap to restore ear lobe

    Objective To investigate the efficacy of rhombic subauricular flap in the repair of earlobe defects. Methods A total of 7 cases of earlobe defects were collected during Sept. 2013 and July 2015. According to the size of the defect, full-thickness rhombic flap was designed. The flap was folded with the short diagonal as axis, and then the corner of the flap near the defect was sutured with the apex of the lower defect edge. All patients were followed up, the shape of reengineered lobe was observed, and patients’ satisfaction was recorded. Results All flaps survived, and incisions healed in the first stage. During the follow-up of 1 to 12 months(mean 7 months), the contour of the ear was smooth and natural, the shape and position of earlobe was fine, and no distortion or deformation occurred. All operations produced satisfactory results. Conclusion Repairing earlobe defects with rhombic subauricular flap is simple and effective, which can achieve the purpose of decorative repair and reconstruction, and accord with the aesthetic concept of modern plastic surgery.
