Effect of tumor associated macrophages on the expression of c-Met in hepatocellular carcinoma cells

Abstract: Objective To explore the molecular mechanism of c-Met expression in hepatocellular carcinoma cells(HCC)influenced by tumor associated macrophages. Methods HCC cells were cocultured with macrophages derived from THP-1 cells. The expression of c-Met in HCC cells was detected at mRNA level by RT-PCR and protein level by flow cytometry. The migration of HCC cells was detected by Transwell cell migration test. Results The expression of c-Met in HCC cells was upregulated by the macrophages at both mRNA and protein levels. After HCC cells were cocultured with macrophages, the migration was enhanced in the presence of human recombinant protein hepatocyte growth factor(HGF). Conclusion Tumor associated macrophages promote the migration of HCC cells by upregulating the expression of c-Met in HCC cells.
