    Effect of cisplatin on IL-6 expression in tumor mesenchymal stem cells and underlying mechanism of p38MAPK pathway

    Objective To investigate the effect of cisplatin(CDDP)on interleukin(IL)-6 expression in tumor mesenchymal stem cells(MSCs)and explore its possible mechanisms. Methods Lewis lung carcinoma(LLC)cells or MSCs were treated with CDDP, and their IL gene expression was detected by polymerase chain reaction array(PCR Array)and further verified by quantitative real-time polymerase chain reaction(qRT-PCR)and enzyme-linked immunosorbent assay(ELISA). After CDDP treatment, the activation of p38MAPK pathway in MSCs was analyzed by Western blotting. CDDP activated p38MAPK pathway was inhibited by the specific inhibitor, and IL-6 expression in MSCs was measured by ELISA. Results CDDP treatment showed no significant effect on the expression of IL-6 in LLC cells(P>0.05). However, IL gene expression was changed in MSCs treated with CDDP for 24 h, and IL-6 expression was increased most significantly. Further studies confirmed the increase of IL-6 mRNA and protein in MSCs after CDDP treatment for 3 h, 6 h and 24 h(P<0.05). Higher expressions of p38 and pp38 were detected in MSCs after CDDP treatment. Moreover, the ratio of pp38/p38 was significantly increased. After the p38MAPK pathway was inhibited by SB203580, the up-regulation of IL-6 caused by CDDP was partially reversed. Conclusion CDDP has no significant effect on IL-6 expression in LLC cells, but it significantly up-regulates the expression of IL-6 in MSCs. CDDP treatment activates the p38MAPK pathway in MSCs, and inhibiting this pathway resultes in down-regulation of IL-6. These results indicate that CDDP might regulate the expression of IL-6 in MSCs via the p38MAPK pathway.
    Role of endoplasmic reticulum stress in ozone-induced neurotoxicity in rats

    Objective To investigate the effects of ozone on the expression of endoplasmic reticulum stress associated protein in rats spinal cord neurons in vivo, and to explore the mechanism of endoplasmic reticulum stress in ozone-induced neurotoxicity. Methods A total of 24 healthy and clean SD rats were randomly divided into 3 groups, control group, high concentration group and low concentration group, with 8 rats in each group. The process of ozone entering the subarachnoid space through intervertebral disc and epidural treatment was simulated by injecting ozone into the built-in tube of rats. Rats in the control group were intrathecally catheterized without any medication, while rats in the 60 μg/mL ozone group and 30μg/mL ozone group were intrathecally administered 10 μL 60 μg/mL and 30 μg/mL oxygen-ozone respectively. Twenty-four hours after the injection, 3 rats in each group were sacrificed, and the lumbar spinal cords were collected to detect the protein expressions of GRP78, CHOP, Caspase-12 and Sig-1R with Western blotting. The gait scores of all rats were recorded before and after oxygen-ozone injection once a day for 12 days. The 60 μg/mL group had longer recovery time of hind limb motor function than the 30 μg/mL group. Compared with the control group, the experimental groups had significantly higher expressions of endoplasmic reticulum stress related protein GRP78, CHOP, Caspase-12 and Sig-1R(all P<0.05), and the 60μg/mL group showed higher expressions than the 30 μg/mL group(all P<0.05). Conclusion In vivo, both high and low concentration medical ozone can cause damage on rats’ motor function. The mechanism may be explained that a low concentration of medical ozone activates the apoptosis through CHOP pathway, while high concentration of medical ozone can activate apoptosis through both CHOP and Caspase-12 pathway.

    Objective To investigate the effects of Physalin B(PB)on proliferation, migration and apoptosis of A549 cells in non-small cell lung cancer(NSCLC), and preliminarily explore its mechanism. Methods The A549 cells were divided into different concentrations(2.5, 5, 10, 20, 40 μmol/L)of PB groups, control group and blank group. The inhibition rate of PB on A549 cells proliferation was detected by real-time cell analyzer(RTCA)and CCK-8 assay, cell migration was examined by transwell chamber assay, the apoptosis and cycle of PB on A549 cells were detected by flow cytometry and the effect of PB on the expression of related proteins was detected by Western blotting. Results PB with 2.5, 5, 10, 20 and 40 μmol/L significantly inhibited the proliferation of A549 cells in a dose-dependent manner. The cell migration ability of 10 and 20 μmol/L PB groups was significantly lower than that of control group. After treatment with 10 and 20 μmol/L PB, the apoptosis rate of A549 cells increased in a dose-dependent manner. Compared with control group, the proportion of G2/M phase cells increased and the proportion of G0/G1 phase cells decreased in 10 and 20 μmol/L PB groups. Compared with the control group, the expression level of Akt protein decreased while the expression levels of Caspase-9 and PTEN increased in 10 and 20 μmol/L PB groups. Conclusion PB can inhibit the proliferation of A549 cells, induce cell apoptosis and cell cycle arrest, which may be related to the regulation of cell cycle related protein expression.

    Objective To investigate the effect of resveratrol(Res)on the proliferation and cell cycle of human retinal capillary endothelium cells(HRCECs)in high glucose environment, and to explore its underlying mechanism. Methods HRCECs were divided into 5 groups with different treatment: Control group(N group)was treated with 5.5 mmol/L glucose; hypertonic group(M group)was treated with 5.5 mmol/L glucose and 19.5 mmol/L mannitol; high glucose control group(H group)was treated with 25 mmol/L glucose; experiment groups(R1-R4 groups)were treated with 25 mmol/L glucose and 25, 50, 100, 200 μmol/L Res. CCK-8 assay and flow cytometry were used to detect the effect of Res on the proliferation and cell cycle of HRCECs, respectively. Real-time PCR and Western blotting were used to detect the mRNA and protein levels of PCNA, Cyclin A1, CDK2 and P21CIP1, respectively. Compared with H group, the cell numbers in S phase were increased in R2 and R3 groups(both P<0.01). Compared with N group, the mRNA and protein expressions of Cyclin A1, CDK2 and PCNA were increased while those of P21CIP1 were decreased in H group(all P<0.01). Compared with H group, the expressions of Cyclin A1, CDK2 and PCNA in HRCRCs were decreased and those of P21CIP1 were increased in R2 and R3 groups(all P<0.01). Conclusion Res can inhibit the high glucose-induced proliferation by down-regulating the expressions of Cyclin A1, CDK2 and PCNA and up-regulating the expression of P21CIP1 in HRCECs.

    Objective To investigate the effects of sevoflurane preconditioning on silent information regulator of transcription 3(SIRT3)expression and the acetylation of H9C2 cardiomyocytes after hypoxia/reoxygenation. Methods Rat H9C2 cardiomyocytes were randomly divided into control group, hypoxia/reoxygenation group and sevoflurane pretreatment group. Cells without any treatment served as control group. Hypoxia/reoxygenation models were prepared in a hypoxia incubator, in which cells were exposed to 2 h hypoxia followed by 2 h reoxygenation. Cells in the sevoflurane pretreatment group were treated with 2.5% sevoflurane for 1 h before hypoxia/reoxygenation processing. Mitochondrial membrane potential was examined with JC-1 fluorescence probe. The acetylation of proteins, especially that of mitochondrial protein, SIRT3 expression, and mitophagy level were detected with Western blotting. Results Compared with the control group, the hypoxia/reoxygenation group had decreased expression of SIRT3(0.78±0.04 vs 1.04±0.06), increased acetylation level of proteins(1.72±0.06 vs 0.98±0.03)and mitochondrial protein(0.96±0.03 vs 0.45±0.03)(P<0.05), decreased cell survival [(48.2±0.4)% vs 100%], decreased mitochondrial membrane potential(1.72±0.14 vs 2.83±0.11), and increased mitophagy level(P<0.05). Compared with the hypoxia/reoxygenation group, the sevoflurane pretreatment group had increased expression of SIRT3(0.93±0.03 vs 0.78±0.04), decreased acetylation level of proteins(1.34±0.05 vs 1.72±0.06)and mitochondrial protein(0.65±0.04 vs 0.96±0.03)(P<0.05), increased cell survival [(65.80±1.53)% vs (48.20±0.40)%] and mitochondrial membrane potential(2.33±0.12 vs 1.72±0.14), and decreased mitophagy level(P<0.05). Conclusion Sevoflurane pretreatment is capable of elevating the expression of SIRT3 and downregulating the acetylation of H9C2 cells after hypoxia/reoxygenation injury, which might be the molecular mechanism underlying the protective effects of sevoflurane on cardiomyocytes.
    Establishment of sensitization model based on the activation of basophil in mice

    Objective To detect the expression of CD63 of basophil in the peripheral blood of sensitized BALB/c mice, and to establish the basophils activation test(BAT)for assessing the sensitization of low molecule weight compounds(LMWC). Methods The morphology and number of basophils in control BALB/c mice(control group)and ovalbumin(OVA)-sensitized BALB/c mice(OVA-sensitized group)were determined by flow cytometry(CD45 APClow/IgE FITChigh)and peripheral blood smears stained by Wright-Giemsa. CD45 APClow/IgE FITChigh was used as the flow scheme to identify mice basophils, and anti-IgE was used as a positive control to incubate with basophils in vitro to observe the expression of CD63 and establish the BAT of mice. In addition, the BALB/c mice was sensitized with penicillin G conjugated BSA(PG/BSA group), and the blood was stimulated by penicillin G-conjugated OVA(PG/OVA)in vitro to observe the expression of CD63, thereby detect the effectiveness of BAT. Results Flow cytometry and blood smears showed that the morphology and numbers of basophils in control group and OVA-sensitized 山 东 大 学 学 报 (医 学 版)57卷3期 -郭岩乳,等.基于小鼠嗜碱性粒细胞活化致敏性模型的建立 \=-group had no significant difference(P=0.655,P=0.667). Anti-IgE could stimulate the expression of CD63 on basophils. After the incubation with OVA in vitro, the CD63 expression in peripheral blood of the OVA-sensitized group was higher than the control group[(59.70±37.93)% vs (2.15±1.13)%, P=0.016]. After the incubation with PG/BSA in vitro, the CD63 expression in peripheral blood of the PG/BSA-sensitized group was higher than the control group[(62.74±41.05)% vs (2.73±1.40)%,P=0.019]. Conclusion Specific allergen in vitro can lead to increased CD63 expression in the sensitized mice, indicating the successful construction of BAT in mice, which is effective verified by PG and can be used to evaluate the sensitization of LMWC.

    Objective To study the chemical constituents of Chiloscyphus polyanthus var. rivularis. Methods The chemical constituents were isolated and identified on the basis of various chromatographic methods such as MCI gel, silica gel, Sephadex LH-20 column, and semi-preparation HPLC as well as a combination of their spectroscopic data, such as NMR and HRESIMS. Results Isolation of Chiloscyphus polyanthus var. rivularis afforded five compounds, including two new eudesmane type sesquiterpenoids, which were named (3S,7R,10S)-7-hydroxy-3-methoxy-eudesma-4E-ene(1)and (3S,7R,10S)-7-hydroxy-3-ethyoxyl-eudesma-4E-ene(2), one guaiane type sesquiterpenoid(guaianediol, 3), one labdane-type diterpenoid(nivenolide, 4), and one triterpenoid(betulin, 5). Conclusion Compounds 1 and 2 are previously undescribed and compounds 3-5 are isolated from the plants in genus Chiloscyphus for the first time.
    Effect of gene mutation on the clinical efficacy of decitabine for patients with myelodysplastic syndrome

    Objective To investigate the effect of gene mutation and related clinical indicators on the prognosis of patients with myelodysplastic syndromes(MDS). Methods Clinical data of 38 MDS patients were collected. Clinical specimens such as blood and genetic sequencing and karyotype analysis of bone marrow fluid were also collected. The response rate and survival rate of MDS patients treated with decitabine were statistically analyzed by single factor and multiple factors using SPSS software. The effects of clinical features and gene mutation on the prognosis of MDS patients treated with decitabine were retrospectively analyzed. Results Among the 38 cases, 92% of patients had at 山 东 大 学 学 报 (医 学 版)57卷3期 -窦春慧,等.骨髓增生异常综合征患者基因突变对地西他滨临床疗效的影响 \=-least one mutant gene and the detection rate of chromosome abnormality was 52.6%. Patients accepted monotherapy of decitabine or combined treatment with decitabine. The median number of treatment cycles was 4, and the total response rate was 63%(24/38). Fourteen patients were invalid. Multifactor analysis found that the response rate of patients with TET2 gene mutation was higher than those without mutation(82% vs 48%, Ps=0.043, Pm=0.034). The follow-up patients ended up with 12 deaths(32%). Median survival time of all patients was 27.6 months. Multifactor survival analysis found that chromosome karyotypes were good, intermediate and poor in 38 MDS patients and their survival time were 31.4, 17.6, 10.4 months, respectively. The risk stratification of IPSS were low risk, intermediate-1, intermediate-2, high risk and median survival time were 31.4, 27.6, 23.4, 11.4 months, respectively. Median survival time for patients with hemoglobin ≥100 g/L and <100 g/L were respective 31.4 and 19.4 months. Median survival time for patients with the number of cytopenias with 0-1 and greater than 2 were respective 31.4 and 17.6 months. The indicators mentioned above were statistically different(all P<0.05). Median survival time of patients with and without TP53 mutation were 16.4 and 31.4 months without statistical difference(P=0.097). Conclusion Patients with TET2 gene mutation have higher response rate to chemotherapy than those without mutation.
    Risk factors of recurrent atrial fibrillation after radiofrequency catheter ablation

    Objective To evaluate recurrence-related risk factors of atrial fibrillation(AF)after radiofrequency catheter 山 东 大 学 学 报 (医 学 版)57卷3期 -陈琳琳,等.预测心房颤动患者射频消融术后复发的危险因素 \=-ablation(RFCA). Methods A total of 270 patients who underwent RFCA were enrolled as the AF group, and 100 healthy subjects as the control group. Clinical baseline information and peripheral venous blood of the two groups were collected. Biochemical markers and miRNA expressions were detected. During the follow-up of 12 months, the patients were divided into the recurrence group and non-recurrence group, and further divided into paroxysmal AF and persistent AF subgroups. Univariate analysis and Logistic regression analysis were used to screen out relevant hazards after RFCA. The area under the receiver operating characteristic(ROC)curve(AUC)in the AF subgroups was analyzed to judge the accuracy of predictive factors. Results The univariate analysis showed statistical differences in the expressions of miRNA-21, miRNA-133, miRNA-150, miRNA-206 and miRNA-328 between the control group and AF group(P<0.05), and statistical differences in the AF type, electric conversion, duration of disease, CHADS2 scoring, left atrial diameter(LAD), left ventricular ejection fraction(LVEF), uric acid and miRNA-21 expression between the recurrence group and non-recurrence group(P<0.05). Logistic regression analysis revealed that LAD(OR=1.187, P=0.031)and miRNA-21 expression(OR=1.226, P=0.012)in the paroxysmal AF subgroup had statistical significance. LAD(OR=1.125, P=0.035), course of disease(OR=1.023, P=0.019), and miRNA-21 expression(OR=1.177, P=0.032)in the persistent AF subgroup had statistical significance. In the paroxysmal AF subgroup, LAD was used as the index to calculate AUC, and the result was 0.750. When miRNA-21 expression and LAD together served as the comprehensive index, AUC=0.844 was derived. AUC measured by the combination of LAD, course of disease and miRNA-21 expression in the persistent AF group also had reference value. Conclusion LAD and miRNA-21 expression are independent recurrence-related risk factors after RFCA in patients with paroxysmal AF, while LAD, miRNA-21 expression and duration of disease are independent recurrence-related risk factors for persistent AF.

    Expression and clinical significance of tripartite motif protein 26 in glioma tissues

    Objective To detect the expression of tripartite motif protein 26(TRIM26)in glioma tissues, to analyze the relationship of its expression and multiple pathological parameters, and to explore the effect on its overexpression in U87 and U251 for the proliferation of glioma cells. Methods A total of 32 cases of glioma tissues and 8 cases of normal brain tissues were collected. Real-time quantitative PCR and immunohistochemistry were used to detect the expression of TRIM26 in levels of mRNA and protein. The relationship between TRIM26 mRNA expression and clinical pathological parameters was analyzed. Effect on TRIM26 overexpression in U87 and U251 for proliferation of glioma cells was detected by MTT assay. Results Immunochemistry results showed that the percentage of TRIM26 positive cells was higher in glioma tissues than control tissues. Compared with the control group, the expression of TRIM26 in glioma tissues was significantly increased [3.51±0.92 vs 1.99±0.45, P<0.01]. The expression of TRIM26 in patients of Ⅲ-Ⅳ grade was markedly higher than in patients of Ⅰ-Ⅱ grade [3.75±0.84 vs 2.65±0.62, P<0.01]. TRIM26 expression was observably associated with stage(P=0.01), but not gender(P=0.72), age(P=0.72), and IDH1 mutation(P>0.999). Moreover, Kaplan-Meier analysis demonstrated that increasing of TRIM26 expression contributed to shorter overall survival [(16.29±1.82)vs(28.73±1.41)months, P<0.01] and recurrence-free survival time [(8.33±1.78)vs(15.95±1.83)months, P<0.01]. Overexpression of TRIM26 in U87 and U251 cell lines promoted the proliferation of glioma cells. Conclusion TRIM26 is highly expressed in glioma, and the overexpression of TRIM26 is associated with pathological grade and prognosis.

    Objective To explore the clinical efficacy of anal canal reduction by external decollement through tunnel and internal suspension in the treatment of circumferential mixed hemorrhoids with anal canal prolapse. Methods A total of 120 cases of circumferential mixed hemorrhoids with anal canal prolapse were randomly divided into 3 groups: patients in the treatment group were treated with anal canal reduction by external decollement through tunnel and internal suspension(n=40); patients in the conventional group received traditional operation(n=40); patients in the procedure for prolapsed hemorrhoids(PPH)group were managed with procedure for prolapsed hemorrhoids(n=40). The operation time, hospital stay, pain score within 3 days after operation, and incidence of postoperative complications(anal edema, hemorrhage, urine retention, rectal tenesmus)were compared. Three months after surgery, the incidence of infection, anal stenosis and fecal incontinence were followed up. Results The treatment group suffered less pain within 3 days after operation than the conventional group(P<0.05); the operation time and hospital stay of the treatment group ranked second among the three groups, and the differences were statistically significant(P<0.001). The treatment group had lower incidence of rectal tenesmus than the other two groups(P<0.05).     Objective To compare the effects of enteral and parenteral nutrition on serum alanine transaminase(ALT), serum creatinine(SCr)and complications in patients with early severe stroke. Methods A total of 60 patients with early severe stroke were randomly divided into two groups:30 patients in the parenteral group were given parenteral nutrition support, while another 30 patients in the enteral group were given early enteral nutrition support. The serum ALT and SCr before and after treatment, and incidence of complications were compared between the two groups. Results The serum ALT concentration significantly increased after treatment in both groups (P<0.01). The increase of ALT in the enteral group was significantly lower than that in the parenteral group (P<0.01). The serum SCr concentration showed comparable increase in both groups after treatment (P>0.05). The incidence of complications in the enteral group was 16.7%, which was significantly lower than 40.0% in the parenteral group (P<0.05). Conclusion Compared with parenteral nutrition support, enteral nutrition support has greater protective effects on liver function, causes fewer complications, and has better clinical effects in the treatment of early severe stroke.

    Objective To explore the incidence, curvature and anatomy of pulp floors of the distolingual roots of permanent mandibular first permanent molars in order to provide theoretical guidance and basis for root canal therapy. Methods The cone-beam computed tomography(CBCT)images of 1 164 permanent maxillary first molars with distolingual roots were measured. The incidence of distolingual roots, the distance between root canal orifices, DB-DL-ML angle and curvature of distolingual roots were determined. Results (1) The incidence of distolingual roots was 25.60%, and the incidence on the right side was higher than that on the left side(P<0.05). There was no significant difference in the incidence between male and female(P<0.05). (2) The incidence of three-rooted and two-rooted mandibular first permanent molars was 25.60% and 22.68%, respectively. There were significant differences between the two types in DL-DB distance and DB-DL-ML angle (P<0.05). (3) The curvatures of the distolingual root canal were greater in the bucco-lingual view(11.60±10.05)° than in the mesio-distal view(30.19±13.48)°(P<0.05). The distolingual root canal bending was mostly seen in the middle third of the root. Conclusion The incidence of distolingual roots of mandibular first molars is high, and the incidence on the right side is higher than that on the left side. The location of DL root canal orifices of three-rooted molars is farther from DB root canal orifice than that of two-rooted molars with four canals.     Objective To evaluate the diagnostic values of abnormal features of lung ultrasonography in childrens pneumonia. Methods The examination results of lung ultrasonography in 91 patients were analyzed retrospectively. The results of ultrasonography diagnosis and clinical diagnosis of childrens pneumonia were compared, and the diagnostic efficiency was calculated. The binary Logistic analysis and the receiver operating characteristic(ROC)curve were used to investigate the diagnostic values of these features. Results There were 56 cases with pneumonia and 35 cases without pneumonia. The main characteristics of childrens pneumonia were B line, A line abnormalities, pleural line abnormalities, lung sliding sign abnormalities, lung consolidation and pleural effusion. Lung consolidation was most correlated with pneumonia(P<0.001). The area under the ROC curve(AUC)of lung consolidation was 0.845(P<0.001). The sensitivity, specificity and accuracy of lung consolidation were 80.4%, 88.6%, and 83.5%, respectively. Conclusion Among all the abnormal features, including B line, A line abnormalities, pleural line abnormalities, lung sliding sign abnormalities, lung consolidation and pleural effusion, of lung ultrasonography in childrens pneumonia, lung consolidation is the most valuable feature with highest specificity and accuracy.
    Health-related quality of life and its influencing factors in type 2 diabetes patients in Shandong Province

    Objective To evaluate the health-related quality of life(HRQOL)in patients with type 2 diabetes mellitus(T2DM)in Shandong Province and to explore its influencing factors. Methods The European quality of life 5-dimensions(EQ-5D)was used to evaluate the HRQOL of T2DM patients. At the same time, the research investigated sociological characteristics of patients, complications of diabetes, and participation in exercise. Logistic regression model and Tobit regression model were used to analyze the factors affecting HRQOL in T2DM patients. Results Logistic regression results showed that male, high-age and participating sports patients has relatively higher quality of life, and the risk factors associated with diabetes quality of life included low income level, mental health status below the general level and diabetes duration(P<0.05). The results of the Tobit regression model analysis showed that age, family annual income level, mental health status, drinking status, participation in exercise, complications, and duration of diabetes had an impact on HRQOL in T2DM patients(P<0.05). Conclusion The influencing factors of HRQOL in T2DM patients include age, family annual income level, participation in exercise, complications, and duration of diabetes, and the effects of the influencing factors should be considered comprehensively.

    Objective To investigate the current situation of Traditional Chinese Medicine(TCM)service providing by rural doctors in Yantai City, Shandong Province, China and to provide support for the grass-roots medical and health system improving. Methods A multistage stratified random sampling method was used to select 3 sample counties in Yantai City of Shandong Province to conduct a questionnaire survey among 129 rural doctors, and a descriptive statistical analysis was conducted using SPSS 20.0 software. Results The survey found 108 rural doctors could provide TCM services, accounting for 83.7%. The number of appropriate TCM techniques mastered by 56.5% of the rural doctors is less than or equal to 3. The highest proportion of TCM services rural doctors providing was cupping, counting 71.3%, followed by massage(57.4%). Sixty-eight point five percent of rural doctors did not prescribe Chinese herbal medicine, 56.5% of village clinics were not equipped with Chinese herbal medicine slices, 34.3% were equipped with 50-100 kinds of Chinese herbal medicine slices, and 82.4% of rural doctors participated in the TCM health management project in public health services.     Objective To evaluate the effects of the compensation mechanism reform in public hospitals on the income structure and to provide references for policy making. Methods The quasi-experimental study design was a time series study analyzing the monthly income changes of a provincial comprehensive public hospital in Shandong Province. The time from Jan. 2014 to Jun. 2016 was taken as the pre-reform, while the period between Jun. 2016 and Dec. 2017 as the post-reform. Results The monthly trend changes of the percentage of income from drugs, and income from medical services before and after the reform was -0.124% and 0.146%, respectively, and the differences were statistically significant(both P<0.01). After the reform, the median proportion of income from laboratory tests, examinations and health materials increased to 7.96%, 9.91% and 25.57%, respectively(all P<0.001).
