Abstract: To To investigate the effect of androgen on GDF9 expression both in vivo and in vitro. Methods： In vivo: In situ hybridization technique was used to measure the expression of GDF9 mRNA in mouse ovaries following once 2.5mg testosterone (experiment group) or NS (control group) every one day, 3 times, respectively. In vitro: Immunohistochemisty was used to detect the expression of GDF9 mRNA in mouse follicles 72 hours after cultured with (experiment group) or without （control group）testosterone. Results： In vivo: The expression of GDF9 mRNA was lower in experiment group than in control group (P＜0.05). In vitro: The expression of GDF9 protein in experiment group was significantly lower than that in control group (P＜0.05). Conclusion： Androgen may decrease the expression of GDF9 mRNA and protein, which is likely associated with the follicular development arrest.
