Expression of Cyclooxygenase2 in human transitional cell bladder carcinoma

Abstract: To investigate the expression of Cyclooxygenase2(COX2) in human transitional cell carcinoma of bladder(BTCC). Methods: The expression of COX2 mRNA was examined by reverse transcriptasepolymerase chain reaction (RTPCR). The protein expression of COX2 was examined by immunohistochemical staining. Results: Upon RTPCR, 13 frozen specimens from bladder cancer were shown to express COX2 mRNA, while none of 7 specimens from normal bladder mucosa expressed COX2 mRNA. The difference was significant. The immunohistochemical results was analogous to the RTPCR results. COX2 protein was mainly localized at cytoplasm of malignant cells (positivity rate was 55.74%), but was not expressed in normal bladder mucosa. Conclusion: The expression of COX2 mRNA and protein is enhanced in human BTCC and is associated with the malignant degree of the tumor, indicating that COX2 may play a key role in the development and progression of BTCC.
