Application of Suiplus semen analysis automatic detection system and manual counting using hemocytometer in mouse semen analysis

Abstract: Objective To compare the functions and differences of Suiplus semen analysis(SSA)automatic detection system and manual counting using hemocytometer in mouse semen analysis. Methods The general morphology, concentration, kinematic parameters and motility of sperm samples of adult male C57BL/6 mice were measured by the above two methods respectively. The data were statistically analyzed. Results The sperm concentration detected by the SSA system was significantly higher than that detected by manual counting(P<0.05). The results of the SSA system showed that the straight line velocity was(25.41±6.42)μm/s, the elocity in curvilinear velocity was(74.13±5.82)μm/s, the linearity was(24.63±8.43)%, and the straightness was(41.00±9.62)%. The proportion of motile sperm detected by the SSA system was significantly higher than that detected by manual counting(P<0.05).
