    Effect of CD40siRNA on CD4+CD25+ regulatory T cells in rats with experimental autoimmune myocarditis

     Objective  To investigate the therapeutic effect of CD40 siRNA on viral myocarditis and its effect on CD4+CD25+ regulatory T cells (Treg). Methods  A total of 40 male Lewis rats were randomly divided into four groups: normal control group, experimental autoimmune myocarditis (EAM) model group, CD40 siRNA treatment group and siRNA treatment group, with 10 rats in each group. On the 1st and 8th day, 0.2mL PBS buffer was injected subcutaneously into the two hind footpads of rats in the control group, while 0.2mL porcine cardiac myosin was injected subcutaneously into the two hind footpads of rats in the other three groups. On the 8th day, 25μL CD40siRNA lentiviral expression vector was injected into the tail vein of rats in the CD40siRNA treatment group, and 25μL siRNA lentiviral expression vector was injected into the tail vein of rats in the siRNA group. After all rats were sacrificed on the 21st day, myocardial pathological changes were observed with optical microscope, and CD4+CD25+Treg expression in rats’ spleen was determined with flow cytometry. Results  The myocardial histopathology integral of the CD40siRNA treatment group (11.70 ± 2.95) was significantly lower than that of the EAM model group (17.00 ± 1.76) (P<0.05). The CD4+CD25+Treg expression was significantly higher in the CD40siRNA treatment group (40.70±4.00) than in the EAM model group (12.20±1.10) (P<0.05). Conclusion  CD40siRNA can relieve myocarditis in EAM rats. The mechanism may be related to the up-regulation of CD4+CD25+Treg expression.

    Identification and application of specific antibody against human TIPE3, a new member of TIPE family

    Objective  To prepare the antibody specifically against human tumor necrosis factor-α induced protein 8 like 3(TIPE3) and investigate the feasibility and specificity of the antibody in detecting TIPE3 protein in human tissues. Methods  The specific polyclonal antibody was prepared by immunization of rabbits with TIPE3 antigen. Human TIPE3 eukaryotic expression vector was constructed. The specificity of TIPE3 antibody and the expression of TIPE3 protein in tumor cell lines were analyzed by Western blotting. TIPE3 expression in tumor tissues was detected by immunohistochemistry. Results  The polyclonal antibody specifically against human TIPE3 was successfully prepared and the full-length human TIPE3 gene was constructed into pflag-cmv2 eukaryotic expressional vector. The prepared antibody was able to detect TIPE3 protein specifically by Western blotting in 293 cells which were transfected with human recombinant TIPE3 vector and it showed no cross-interaction with TIPE2 protein. Both lung cancer-derived cells (H1975) and hepatocellular carcinoma cells (HepG2) expressed TIPE3 proteins while cervical cancer cells(Hela) did not. Immunohistochemical reactions showed that strong positive staining of TIPE3 appeared in lung cancer cells, mainly in cytoplasm and on cell membrane, but not in the normal or adjacent tissues. Conclusion  The polyclonal antibody against human TIPE3 is successfully prepared and it is confirmed that the antibody can be used in both Western blotting and immunochemical staining procedures with high specificity.

    Effects of bone morphogenetic protein-2 on the proliferation and differentiation of neural stem cells

    Objective  To explore the optimal method for the isolation of primary neural stem cells (NSCs) from mouse, and the effects of bone morphogenetic protein-2 (BMP-2) on the proliferation and differentiation of NSCs. Methods  NSCs were isolated from mouse (S129 background) embryos at E12.5 by mechanical blowing combined with nylon membrane filtration. The nestin immunofluorescent staining technique was used to identify the isolated NSCs. Then NSCs were divided into two groups: control group and BMP-2 (10ng/mL) treated group.The proliferation and differentiation of NSCs were determined by BrdU labeling and immunofluorescent staining of microtubule associated protein 2 (MAP2) and glial fibrillary acidic protein (GFAP). Results  ① The isolated NSCs were round and existed in single or pairs in different sizes observed under the inverted microscope. At day 3 to 5 after culture, the neurospheres in different sizes appeared. The nestin-positive cells accounted for more than 90 percent in the cultured neurospheres. ② The ratio of BrdU-positive cells against total NSCs treated with BMP-2 was significantly decreased in comparison with that in control group (P<0.05). ③ The percentage of MAP2-positve cells in BMP-2 treated group was lower than that in control group(P<0.05), while the GFAP-positve cells in NSCs treated with BMP-2 was higher than control group(P<0.05). Conclusion   ① Primary neural stem cells can be successfully isolated and cultured from mouse using a modified mechanical blowing-nylon membrane filtration method, which is highly steady and feasible. ② BMP-2 (10ng/mL) can inhibit the proliferation of NSCs in vitro and inhibit its neuronal differentiation, while promoting its astrocytic differentiation. 

    Experimental study of anti-CD3/anti-EGFR bispecific antibody therapeutic activity against pancreatic cancer cells

    Objective  To investigate the cytotoxicity of EGFR/CD3 bispecific antibody by chemical synthesis against pancreatic cancer cells in vitro. Methods  In previous researches, the EGFR/CD3 bispecific antibody was prepared by chemical synthesis. The cytotoxicity activity of this antibody against pancreatic cancer was analyzed by the cell combination rate assay and 51Cr assay. The cell cycle and apoptosis were analyzed by flow cytometry. Then the comparisions of the cytotoxicity activity between the bispecific antibody and EGFR mAb was conducted. Results  The biding rate of EGFR/CD3 bispecific antibody cell with effector cells PBLS and pancreatic cancer cell line SW1990 was high enough to satisfy the treatment demand.The lysis rate and apoptosis rate of cancer cells treated with immunological effector cells targeted by EGFR/CD3 bispecific antibody were higher than those of the  control groups significantly (P<0.05). The apoptosis rate of cancer cells increased. Conclusion  It is suggested that the cytotoxicity of effector cells could be enhanced by EGFR/CD3 bispecific antibody in vitro, so it has promising curative effect on pancreatic cancer.

    Role and molecular mechanism of TIPE2 in the progression of colon cancer

    Objective  To explore whether tumor necrosis factor-α-induced protein-8 like-2 (TIPE2) inhibits toll-like receptor 4 (TLR4) pathway by binding to caspase-8 to participate in the progression of colon cancer. Methods  TIPE2 expression was down-regulated in HT-29 cells by small RNA interference technology. TLR4 pathway was activated with 1μg/mL lipopolysaccharide (LPS). Caspase-8 activities were detected in the TIPE2 knockdown cells, TIPE2 knockdown & TLR4 activation cells, TLR4 activation cells and wild-type cells. Results  Real-time PCR found that TIPE2-siRNA decreased the TIPE2 expression successfully. Caspase-8 activities were higher in TIPE2 knockdown cells and TLR4 activation cells than in wild-type cells (P<0.05). Caspase-8 activities were higher in TIPE2 knockdown & TLR4 activation cells than in TLR4 activation cells (P<0.05). Conclusion  TIPE2 can inhibit caspase-8 activity and it is involved in the progression of colon cancer through its binding to caspase-8 which negatively regulates TLR4 pathway.

    Objective  To construct Escherichia coli (E.coli) expression vector of human intestinal trefoil factor (hITF), express recombinant hITF and analyze its biological activity. Methods  The hITF gene encoding mature peptide was amplified by RT-PCR, and then inserted into the expression vector pET32a. Recombinant plasmid pET32a-hITF was transformed into the Escherichia coli Origami B(DE3) and hITF was expressed by IPTG induction. hITF was purified by Ni-NTA affinity chromatography, and determined by SDS-PAGE, Western blotting, and N-terminal amino acid sequence analysis. Biological activity was assayed in an in vitro restitution model. Results  It was proved that the fragment amplified was inserted into the expression vector pET32a correctly by PCR and gene sequencing. SDS-PAGE analysis proved that the molecular weight of hITF was about 32kD, and Western blotting demonstrated that the expressive proteins had good antigenicity and specificity. The N-terminal 15 amino acid sequence was consistent with the theoretical value. In addition, hITF was proved to enhance migration activity. Conclusion  hITF Escherichia coli expression vector is successfully constructed and recombinant hITF is expressed.

    Expression and function of miR-181a in thymocytes with thymic aging

    Objective  To explore the expression of miR-181a in age-associated thymic involution, and the interaction between miR-181a and its target gene ZFP36l2 in thymus during aging. Methods  A total of 18 C57BL/6 mice were randomly divided into 3 groups, with 6 in each, including the 1-month-old group, 9-month-old group, and 15-month-old group. The thymic dissociated cells were stained by fluorescence-conjugated anti-CD45, followed by metal beads through LS column. Quantitative PCR was performed to observe changes in miR-181a and ZFP36l2 expressions in the different age groups. Transfecting luciferase reporters were transfected into HEK293 cells, and the auto-fluorescence values were measured 48 hours later. Results  The miR-181a expression showed a downward trend with aging (P<0.05), while its target gene ZFP36l2 showed an upward trend (P<0.05), with a statistically significant negative correlation between the two expressions (P=0.04). The luciferase reporter assay confirmed that miR-181a targeted the ZFP36l2 3'UTR region. Conclusion  The miR-181a, which targets ZFP36l2, plays an important role during age-associated thymic involution.

    Effect of PARP inhibitor combined with Gemcitabine or Docetaxel on the  proliferation of androgen-independent prostate cancer cell line PC3

    Objective  To explore the effect of PARP inhibitor AG014699 combined with  Gemcitabine or Docetaxel on the proliferation of androgen-independent prostate cancer cell line PC3. Methods  PC3 cells were cultured in vitro and then divided into PARP inhibitor group, Gemcitabine group, Docetaxel group, combination of PARP inhibitor and Gemcitabine group, combination of PARP inhibitor and Docetaxel group. CCK-8 method was used to assess the vitality of PC3 cells after different treatments; flow cytometry was adopted to determine apoptosis and distribution of cell cycle after PC3 cells were treated with PARP inhibitor. Results  PARP inhibitor AG014699 showed significant anti-proliferative effects on PC3 cells in a dose-and-time dependent pattern (P<0.05); Gemcitabine or Docetaxel inhibited the growth of PC3 cells in a dose dependent manner (P<0.05); compared with Gemcitabine or Docetaxel used alone, combination of PARP inhibitor and Gemcitabine or Docetaxel could significantly inhibit the growth of PC3 cells (P<0.05). Conclusion  PARP inhibitor AG014699 combined with Gemcitabine or Docetaxel has a synergistic antitumor effect on androgen-independent prostate cancer cell line PC3, which may become a promising therapy for androgen-independent prostate cancer.  

    Effects of bone marrow mesenchymal stem cells transplantation on the expression of  Toll-like receptor 4 in injured spinal cord of rats

    ObjectiveTo investigate the effect of bone marrow mesenchymal stem cells (BMSCs) transplantation on the expression of Toll-like receptor 4 (TLR4) in injured spinal cord of rats, and to explore the mechanism by which BMSCs transplantation treats spinal cord injury. Methods  A 4-week old mouse was utilized to separate BMSCs, which were cultured into third generation and then identified by immunohistochemical Methods  . A total of 90 8-week old male Wistar rats were evenly divided into three groups: group of surgical control, group of spinal cord injury and group of cell transplantation. Rats in group of spinal cord injury was used to establish Allen′s SCI model, rats in group of cell transplantation was injected with the BMSCs suspention after SCI. Then the rats were killed respectively at 24 hours, 72 hours, 7 days and 1 cm of the spinal cord blocks in the center of contusion site was removed. The expression of TLR4 in the injured area was detected with immunohistochemical method and real-time quantitative PCR (RT-qPCR). Results  The expression of TLR4 in group of spinal cord injury increased compared with that of group of surgical control (P<0.01). The expression of TLR4 in group of cell transplantation decreased compared with that of group of spinal cord injury (P<0.01). Conclusion  BMSCs transplantation may inhibit inflammation in spinal cord injury by decreasing the expression of TLR4.

    Correlation of ATF4 and GPR48 expressions in the progression of traumatic necrosis of the femoral head in rats

    Objective  To explore the correlation between ATF4 and GPR48 expressions, which might reflect the correlative regulation effects during the progression of traumatic necrosis of the femoral head in rats. Methods  A total of 36 SD rats were divided into two groups: the experimental group (n=24) which were models of traumatic necrosis of the femoral heads operated with Norman′s technique, and the control group (n=12) which received sham operation. The femoral heads were collected 1w, 2w and 4w after operation, and then analyzed with histological and immunohistochemical Methods  , RT-PCR and Western blotting to observe the progression of necrosis.  Results  From the 1st to 4th week, the necrosis gradually aggravated. In the 4th week, there were breakages on the articular cartilage with rough surface. Immunohistochemical analysis showed that the expressions of ATF4, GPR48 and PCNA in the experimental group gradually increased, but were significantly lower than those in the sham operation group (P<0.05) at the same time point. RT-PCR Results   suggested that mRNA of ATF4 and GPR48 gradually increased with time going. Western blotting displayed that the protein expressions of ATF4 and GPR48 gradually elevated in the experimental group, and ATF4 had a positive correlation with GPR48 (r=0.659, P<0.01). Conclusion  In the progression of traumatic necrosis ofthe femoral head, the expressions of ATF4 and GPR48 decline in the initial stage but may gradually increase. There is a positive correlation between them, which might be associated with the concomitant repair of the necrotic bone.

    Effects of Avastin inhabiting corneal neovascularization induced by penetrating keratoplasty in rabbit models

    Objective  To investigate the effects of Avastin on VEGF protein expression in the cornea and aqueous humor after penetrating keratoplasty. Methods  Rabbits were randomly assigned to two groups: the eyedrop group and the subconjunctival injection group after PKP according to different administration routes. Each group was then divided into the experimental group and the control group based upon whether Avastin intervention was used. Corneal neovascularization (CNV) was observed at the 3rd, 7th and 14th day after the surgery. VEGF expression in the cornea was detected by immunohistochemistry technique and VEGF protein expression in the aqueous humour was measured by ELISA. Results  Growth of corneal neovascularization and expression of VEGF in the cornea and aqueous humor of the experimental groups were significantly lower than those in the control groups (P<0.05). All the observation indexes had no significant difference between the eyedrop groups and the subconjunctival injection groups (P>0.05). Conclusion  Early subconjunctival injection  of Avastin eyedrop can partially inhibit coreal neovascularization after PKP and decrease VEGF protein expression in the cornea and aqueous humour.

    Classification and surgical planning of bilateral cerebral falcine meningiomas

    ObjectiveTo probe into the classification and surgical planning of bilateral cerebral falcine mengingiomas (BCFMs). Methods  The clinical materials of 31 patients with BCFMs treated surgically were analyzed retrospectively. A classification system of BCFMs was proposed according to the tumors’ bases, symmetry and distance between the bases of bilateral tumors and the surgical planning was determined. Results  Simpson GradeⅠ-Ⅱ resections were achieved in 27 cases and Ⅲ in 4. No perioperative death occurred. No tumor recurrence was observed during follow-up period of 3 months to 3 years after operation in all patients. Conclusion  The classification system has certain reference values to determine the surgical planning and improve the operative effect.

    Objective  To explore the imaging manifestations of fetal malformations of cortical development (MCD). Methods  Data of 280 singleton pregnancy women for fetal brain MRI and ultrasound examination during Aug. 2012 to Aug. 2013 were retrospectively reviewed.  Results  Altogether 12 cases were diagnosed of MCD, 8 of which were confirmed by MRI after ultrasound findings, and 4 of which were detected after being missed by ultrasound. The 8 cases included heterotopic gray matter (n=3), microcephaly (n=4), and hemimegalencephaly (n=1); the 4 cases were schizencephaly (n=2), tuberous sclerosis (n=1), and hypoplasia of temporal lobe (n=1). Among these cases, 10 terminated pregnancy, while 2 continued gestation. Conclusion  Ultrasound combined with MRI presents typical imaging characteristics, which is a powerful tool to diagnose fetal malformations of cortical development.

    Application of the UPOINT clinical classification in patients with chronic prostatitis

    Objective  To explore the application of a clinical phenotype system, UPOINT, in the classification of patients with chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS).  Methods  A total of 241 patients with CP/CPPS treated during Feb. to Oct. 2012 were classified using UPOINT. Patients′ symptoms severity was assessed with National Institutes of Health-Chronic Prostatitis Symptom Index (NIH-CPSI) and International Prostate Symptom Score (IPSS).  Results  Median CPSI score was 20.78±5.89; median IPSS score was 8.08±4.85. The percent of each UPOINT domain was 58.9%, 60.2%, 86.3%, 26.6%, 26.1% and 39.8% respectively. Of all patients, 9.5% were positive with only 1 domain, 29.9% with 2 domains, 27% with 3 domains, 21.6% with 4 domains, and 12% with 5 domains. The number of positive domains was closely correlated with the symptom severity assessed with NIH-CPSI and IPSS (r1=0.61, r2=0.45; P<0.01), and positively correlated with the quality of life (CPSI QoL) (r=0.48, P<0.01).  Conclusion  The UPOINT phenotype system can classify patients with CP according to clinically relevant domains, which can not only improve the diagnostic rate, but also provide patients individualized treatment and improve their quality of life.

    Association between low T3 syndrome and arterial stiffness in continuous ambulatory peritoneal dialysis patients

    Objective  To investigate the association between low T3 syndrome and arterial stiffness in continuous ambulatory peritoneal dialysis (CAPD) patients. Methods  129 CAPD patients were divided into the low T3 syndrome group and euthyroid function group. All patients received a series of examinations every 3 months, including blood biochemical indices and pulse wave velocity(PWV)for arterial stiffness. The data were analyzed using an independent t-test to compare PWV, blood biochemical indices and metabolic status between low T3 syndrome and euthyroid patients. Multiple regression analysis was performed to identify risk factors for PWV. Results  Both age and proportion of female patients were significantly higher in low T3 syndrome group compared with the euthyroid function group (P<0.001; P=0.044 ), although no significant differences in hemoglobin, albumin, metabolic status or other values were found. However, PWV was much higher in low T3 syndrome group than that in euthyroid function group (P<0.001). PWV was significently correlated with the diabetes (P<0.001), age (P=0.001), systolic pressure (P=0.038), diastolic pressure (P=0.045), euthyroid function (P<0.001), plasma albumin (P=0.007) and KT/V(remnant renal function) (P=0.018). Multiple regression analysis revealed that the level of free triiodothyronine(FT3), diabetic status and age were the independent risk factors of PWV. They accounted for 57.0% of the total variance. Conclusion  TheCAPD with low T3 syndrome is correlated with PWV, and the CAPD patients with low T3 syndrome show increased arterial stiffness.

     Objective  To investigate the association between G(-248)A single nucleotide polymorphism (SNP) in pro-apoptosis gene Bax promoter and the clinicopathological features of breast cancer. Methods  The 3 genotypes (AA, AG, GG) of Bax G(-248)A SNP in 113 patients with breast cancer were analyzed using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), and the distributions of the 3 genotype frequency were compared. Results  The distribution of genotype AG+AA was notably lower than that of GG in tumor size T>2cm, clinical stage II+III, positive lymph node metastasis and C-erbB2 protein expression, with statistically significant difference (P<0.05). There were no differences between the distributions of AG+AA genotype and GG genotype in pathological types, histological grade, estrogen receptor (ER) and progesterone (PR). Conclusion  The AG+AA genotype may be associated with smaller breast cancer, lower clinical stage, lower auxiliary lymph node metastasis rate, and negative C-erbB2 protein expression.

    Objective  To evaluate the treatment of caesarean scar pregnancy. Methods  Data of 102 patients with caesarean scar pregnancy treated during Jan. 2006 to Feb. 2012 were retrospectively reviewed with clinical cohort study. The patients were divided into 3 groups, including muscle and intrauterine cavity injection of methotrexate (MTX, Group A), MTX therapy followed by ultrasound-guided suction curettage (Group B) and uterine arteries embolization followed by low vacuum aspiration (Group C). Results  The mean resolution time of serum β-hCG, time taken for CSP mass to disappear, time of vaginal bleeding, hospital stay and costs were statistically different in the three groups (P<0.05). The success rate for Group A, B and C were 92.86%, 88.52% and 84.62%, respectively, with no statistically significant difference (P>0.05). Conclusion  All three therapies yield high success rate, while muscle and intrauterine cavity injection of methotrexate is effective, cheap and non-invasive.

     Objective  To explore gene polymorphisms of 5,10-methylenetetrahydrofolate reductase (MTHFR) and 5-methyltetrahydrofolate-homocysteine methyltransferase reductase (MTRR) in women from Chinese Han population in Yantai city. Methods  A total of 2 670 Han women were recruited in the study. Their oral epithelial cells were collected, followed by the extraction of genome DNA, in order to detect gene polymorphisms of MTHFR and MTRR using fluorescence quantitative PCR method. The Results   were compared with those in other cities of China. Results  The frequency of MTHFR 677CC, 677CT and 677TT of Han women in Yantai city were 18.6%, 49.2% and 32.2%, respectively. The frequency of MTHFR 677TT among women in Yantai city was obviously different from Weifang, Zibo, Zhengzhou, Zhenjiang, Wuhan, Deyang, Kunming, Guangzhou and Qionghai (P<0.05). The frequency of MTHFR 1298AA, 1298AC and 1298CC of Han women in Yantai city were 72.5%, 25.7% and 1.8%, respectively.The frequency of MTHFR 1298CC in Yantai was different from Weifang, Zibo, Linyi, Zhenjiang, Wuhan, Deyang, Kunming, Guangzhou and Qionghai (P<0.05). The frequency of MTRR 66AA, 66AG and 66GG of Han women in Yantai city were 54.6%, 38.1% and 7.2%, respectively. The frequency of MTRR 66GG in Yantai was only different from Zibo, Linyi and Qionghai(P<0.05). Conclusion  The MTHFR, MTRR polymorphism distribution of Han women in Yantai city is different from other regions, showing region-specific characters.

    Significance and influential factors of Fibroscan detection  in the progress of patients with chronic hepatitis B disease

    Objective  To explore the evaluation effect and analyze the influential factors of Fibroscan (FS) detection in the progress of patients with chronic hepatitis B disease. Methods  Six hundred and sixty patients with chronic hepatitis B disease were detected with Fibroscan and their clinical data were collected, and 96 of them had liver biopsy Results  . Influences of various clinical indicators on FS values were analyzed. The 96 cases were grouped based on their fibrosis stage in biopsy, the receiver-operating characteristic curves (ROC) of FS values were drawn and the areas under the curve (AUC) were calculated. Results  Multiple clinical indexes, such as age, gender, PLT, PTA, ALT, TBIL, the inner diameter of portal vein and thickness of spleen were associated with FS value, and their differences between different levels had statistical significance(P<0.05). Multiple linear stepwise regression analysis showed that ALT, PTA, thickness of spleen, the inner diameter of portal vein, PLT and TBIL were correlated with FS value. According to the absolute value of the standard regression coefficient, the influence of these indicators on FS value decreased gradually. The AUCs of FS detection for 18 cases of mild (S1), 39 cases of moderate (S2), 12 cases of severe (S3) degree liver fibrosis and 27 cases of cirrhosis (S4) were 0.724, 0.832, 0.836 and 0.798, respectively, and the cut-off values for diagnosis of various degree of fibrosis and cirrhosis were 9.8, 10.5, 12.3 and 14.8 kPa, respectively. Conclusion  Fibroscan detection has a high coincidence rate with liver pathological fibrosis stage. It is a very effective and noninvasive examination in assessing disease progression of patients with chronic hepatitis B disease.

    Risk factors of recurrence and survival time for single small liver cancer after percutaneous microwave coagulation therapy

    Objective  To investigate the risk factors of intrahepatic recurrence and survival time after percutaneous microwave coagulation therapy (PMCT) for single small liver cancer patients with hepatitis B virus related cirrhosis. Methods  130 patients′ medical records for 3 years were analyzed retrospectively. The patients with intrahepatic recurrences were divided into the early (≤1 year) and late (2-3 years) recurrence groups, and the patients without recurrence were defined as no recurrence group. The χ2  test and Logistic analysis were used to evaluate the univariate and multivariate analyses associated with intrahepatic recurrence, respectively. Cox proportional hazard model was used to evaluate the univariate and multivariate analyses associated with the survival time. Results  ① 81 patients out of 130 cases were diagnosed with postoperative intrahepatic recurrence (65.85%), in which 50 were early recurrence (39.06%) and 31 were late recurrence (25.20%). ② The irregular boundary of tumor (P=0.010) and distance to the large vascular≤1cm (P<0.001) were related to the early recurrence, and the serum HBV-DNA titer (≥105 copies/mL) was related to the late recurrence (P<0.001). The irregular boundary of tumor (P<0.001), distance to the large vascular ≤1cm (P=0.007) and recurrence time  ≤ 12 months (P=0.008) were related to the survival time. Conclusion  For the single small liver cancer patients with hepatitis B virus related cirrhosis, the irregular boundary of tumor and small diatance to the large vascular are independent risk factors of early recurrence; serum HBV-DNA titer is the risk factor of late recurrence; the irregular boundary of tumor, diatance to the large vascular and recurrence time are the independent risk factors of the survival time.

    Objective  To validate the association of ankylosing spondylitis (AS) with ERAP1 gene polymorphism with Meta-analysis. Methods  Relevant studies on AS and ERAP1 were retrieved and a Meta-analysis was conducted, and then the statistics were analyzed with Reveman 5.0 software. Results  A total of 17 studies with 11411 AS patients and 16511 controls were included. Meta-analysis showed significant differences in minor allele frequencies of rs27044, rs30187, rs10050860, rs2287987, rs17482078, rs27434 and rs27037 between the two groups (P< 0.05). In addition, significant differences of genotype frequencies of these SNPs were also found between the AS cases and controls (P<0.05). Conclusion  ERAP1 gene polymorphism is related to the genetic susceptibility to AS.

    Effect of academic stress on CCL28 and sIgA in saliva of undergraduates1

     Objective  To investigate the effect of academic stress on oral mucosal immune function by semi-quantitatively detecting and comparing the levels of mucosa-associated epithelia chemokine (CCL28/MEC) and secretory immunoglobin A (sIgA) in saliva before and after certain exams. Methods  A total of 37 undergraduates (17 sophomore girls, 10 junior boys and 10 junior girls, the latter two were defined as the male group and the female group ) were enrolled to collect non-stimulated completed saliva before and after certain exams. The changes of CCL28 and sIgA in the samples were detected with ELISA. Results  The OD values of sIgA and CCL28 after exams significantly decreased (P<0.05) in all 37 students. The change of CCL28 (P<0.05) in the saliva spanning the exams was more significant in the female group than in the male group, whereas the change of sIgA level (P<0.05) showed no obvious difference. After the same exam, the change of CCL28 in saliva was more obvious than the change of sIgA in the female group (P<0.05), without significant difference in the male group (P>0.05). Conclusion  Academic stress plays an important role in the secretion of CCL28 and sIgA in saliva, and females are more susceptible to such stress.

    Root resorption associated with self-ligating versus conventional ligating orthodontic bracket systems: a systematic review

    Objective  To evaluate root resorption associated with self-ligating and conventional ligating bracket systems in orthodontic treatment. Methods  Electronic databases such as CNKI, Wan Fang Data, VIP, CBM, the Cochrane Library, Medline, PubMed and EMbase, were searched from the day of they were established to June 30, 2013, in order to collect randomized controlled trials (RCTs) about the root resorption associated with self-ligating versus conventional ligating brackets. The quality of the included studies was evaluated with two reviewers independently, and Meta-analysis was performed by using RevMan 5.2.1 software. Results  Three RCTs including 464 teeth were identified. The Results   of Meta-analysis showed no significant differences between the two groups in maxillary incisors (MD = 0.20, 95%CI -1.45 to 1.05, P=0.75), maxillary lateral incisors (MD=0.53, 95%CI -0.50, 1.56, P=0.31), mandibular incisors (MD=-0.01, 95%CI  -0.22, 0.20, P=0.92) and mandibular lateral incisors (MD=-0.31, 95%CI -1.33, 0.71, P=0.55). Conclusion  As the type of orthodontic appliance may not be the major factor influencing root absorption, and suitable brackets can be chosen by orthodontic dentists according to the merits and demerits of self-ligating and traditional ligating brackets and deformity of patients’ teeth.

    Objective  To analyze the relationships between lifestyle and common chronic diseases of centenarians in longevity areas of Shandong Province, China. Methods  1340 people aged over 60 were selected as research subjects from  five longevity areas (Pingyi, Chengyang, Shanxian, Rushan and Feixian) of Shandong Province, including 896 people(aged 60-79), 189 people (aged 80-99) and 255 centenarians(aged ≥100). Their socio-demographic characteristics, family environment, economic conditions, lifestyle and the prevalence of chronic diseases were collected. Results  The total prevalence of chronic diseases in centenarians was 68.63% (175/255), among them 47.45% (121/255) suffered from one disease, 17.25% (44/255) suffered from two and 3.92% (10/255) suffered from three or more diseases; Compared with the oldest-old and the young-old, the centenarians had statistical differences (P<0.05) from the prevalence of movement system diseases, eye diseases, hearing disorders and digestive diseases. The Logistic regression showed that the influencing factors of chronic diseases included age, drinking, smoking and temper.Conclusion  Low rates of smoking and drinking and gentle temper are important characteristics of centenarians and there are differences among different population on prevalence of chronic diseases. We should carry out different strategies according to different conditions so as to improve the health of the elderly people.
