    Effects of the overexpression of type 4 angiotensin receptor on the formation of atherosclerotic lesions

    Objective To explore the effects of type 4 angiotensin receptor(AT4R)on the formation of atherosclerotic lesions and to elaborate the possible mechanism. Methods A total of 60 apolipoprotein E-deficient mice fed with high-fat diet for 4 weeks were randomly divided into 3 groups: control group(receiving infusion of PBS), Ad-EGFP group(receiving infusion of adenoviral vectors with green fluorescence protein)and Ad-AT4R group(receiving infusion of adenoviral vectors with AT4R). High-fat diet lasted for another 4 weeks and then all mice were sacrificed to collect blood samples and whole aorta. The levels of blood glucose and blood lipid were measured. The aortic plaques were stained by HE and oil red O staining method to measure the percentage of positive atherosclerotic lesion area. Results There was no significant difference in blood glucose and serum lipid levels among the 3 groups. However, compared to the control group and the Ad-EGFP group, the Ad-AT4R group showed lower positive atherosclerotic lesion area stained by HE and oil red(P<0.05). Conclusion Overexpression of AT4R may inhibit the formation of atherosclerotic lesions independent of regulation of blood glucose and serum lipid. Thus AT4R may provide protection against atherosclerosis.
    miR-1/133 participates in the expression of ion channel genes of myocardial cells in myocarditis mice

    Objective To explore the involvement of miR-1/133 in regulating the expression of potassium and calcium ion channel in mouse with acute viral myocarditis. Methods After the mice models of acute viral myocarditis were established, they were divided into 4 groups: control group, viral myocarditis group, myocarditis+miR-1/133 mimics group, and myocarditis+miR-1/133 NC group, with 10 mice in each group. The myocardial pathological changes were observed with HE staining. The expressions of miR-1, miR-133, Kcnd2, Irx5, Kcnj2, and α1c in myocardium were detected with qRT-PCR. The expressions of Kv4.2, Kir2.1, and Cav1.2 in myocardium were detected with Western blotting. Results HE staining showed that, in the control group, myocardial cells arranged orderly, and no inflammatory cells infiltrated myocardium matrix; in the myocarditis group and myocarditis+miR-1/133 NC group, myocardial cells swelled and arranged disorderly, and inflammatory cells infiltrated myocardium matrix; in the myocarditis+miR-1/133 mimics group, myocardial cells arranged orderly without cell edema, and few inflammatory cells infiltrated myocardium matrix. Compared with the control group, in the myocarditis group and myocarditis+miR-1/133 NC group, the expressions of miR-1, miR-133, Kcnd2, Kcnj2, Kv4.2 and Kir2.1 in the myocardium significantly decreased(P<0.01), while the expressions of Irx5, α1c and Cav1.2 increased(P<0.01). Conclusion miR-1/133 is involved in the regulation of calcium and potassium ion channel gene expression in mice with acute viral myocarditis.
    p38 MAPK signaling pathway regulates the expressions of type Ⅰ and Ⅲ collagens in human aortic smooth muscle cells

    Objective To investigate the effects of p38 MAPK on the expressions of type Ⅰ and type Ⅲ collagens in human aortic smooth muscle cells(HASMCs). Methods HASMCs were treated with inhibitors and siRNAs respectively. Inhibitor test cells were divided into 3 groups: (1) normal control group(cells were cultured in smooth muscle cell complete medium); (2) DMSO control group(cells were treated with 0.01% DMSO); (3) p38 MAPK inhibitor group(cells were treated with 0.01% p38 MAPK inhibitor(20 mg/mL). Gene silencing test cells were divided into 3 groups: (1) static control group(cells were cultured in smooth muscle cell complete medium); (2) negative control siRNA group(cells were transfected with negative control siRNAs for 48 h using MACSfectin reagent); (3) p38 MAPK siRNA group(cells were transfected with p38 MAPK siRNAs for 48 h using MACSfectin reagent). Results Compared with the normal control group, the mRNA and protein expressions of type Ⅰ and Ⅲ collagens were not obviously changed in the DMSO control group(P>0.05), while the expressions decreased significantly in the p38 MAPK inhibitor group(P<0.05). Compared with the static control group, the mRNA and protein expressions of type Ⅰ and Ⅲ collagens were not obviously changed in the negative control siRNA group(P>0.05), while the expressions decreased significantly in the p38 MAPK siRNA group(P<0.05). Conclusion p38 MAPK signaling pathway regulates the expressions of type Ⅰ and type Ⅲ collagens in HASMCs.
    Effect of local slow-release coated sirolmus on fistula neointimal hyperplasia

    Objective To establish a model of cimino-brescia arteriovenous fistula by anastomosing rabbit carotid artery with internal jugular vein and to explore the mechanism of coated sirolmus in inhibiting the fistula anastomotic intimal hyperplasia. Methods A total of 18 healthy New Zealand male rabbits were randomly divided into blank control group(the grafts accepted no management), F-127 group(0.5 mL of 20% pluronic F-127 gel was locally applied on adventitia and anastomosis site)and F-127+sirolmus group(0.5 mL 20% pluronic F-127 gel containing 0.5 mg sirolmus was locally applied on adventitia and anastomosis site)to establish models of cimino-brescia arteriovenous fistula by anastomosing rabbit carotid artery with internal jugular vein. The vein intimal hyperplasis was observed with tissue morphology method, the vascular intima thickness was measured with computer image system, and expressions of TGF-β1 and CTGF were detected with immunohistochemistry and western blotting. Results Three weeks after operation, the intima in the blank control group and F-127 group thickened obviously as compared with that in F-127+sirolmus group(P<0.05). The TGF-β1 and CTGF expression were higher in the blank control group and F-127 group than in the F-127+sirolmus group(P<0.05). There were no significant differences in the above parameters between the blank control group and F-127 group(P>0.05). Conclusion Sirolmus partial paintcoated in vascular adventitia inhibits the fistula neointimal hyperplasia via inhibition of TGF-β1 and CTGF expressions.
    Effect of liraglutide on leptin and fat content in high fat diet induced obese mice

    Objective To observe the effect of liraglutide on the distribution of leptin and fat in high-fat diet induced C57BL/6 obese mice model. Methods A total of 60 C57BL/6 mice aged 4 weeks, body weight(14.03+0.92)g were randomly divided into 2 groups: high-fat diet(40% fat)group(n=45)and normal chow diet(4% fat)group(n=15). The body weight was recorded once a week until the obese mice models were built. Then the 45 obese mice in the high-fat diet group models were randomly divided into 3 groups: liraglutide group[intraperitoneal injection of 100 μg/(kg·d), n=15 ], transferred normal diet group[intraperitoneal injection of 0.017 mL/(kg·d), n=15)] and high fat diet group(n=15). During the 12-week intervention, the body weight of all animals was recorded. The fat content and leptin were monitored by magnetic resonance imaging and enzyme-linked immunosorbent assay. Results The liraglutide group had the lowest body weight than the other 3 groups(P<0.05). The liraglutide group had reduced degree of leptin compared with transferred normal diet group and high fat diet group(P<0.05). The liraglutide group was different from the other groups in the mesenteric, epididymal, perirenal, subscapular and subcutaneous fat(P<0.05). Conclusion Liraglutide can significantly reduce the body weight and leptin degree in obese mice and has an effect on regulating lipid and fat distribution.
    Effect of interleukin-25 on inducing the airway remodeling of asthma mice by nuocyte cells

    Objective To investigate the effects of interleukin-25(IL-25)on airway remodeling of asthma mice by nuocyte cells. Methods Thirty mice were randomly divided into control mouse group, asthma mouse group, and anti-IL-25 mouse group, ten in each group. Nuocyte cells were collected from the spleens and marrows of asthma mice which were induced by ovalbumin(OVA), and diveded into control cell group, IL-25 cell group, and anti-IL-25 cell group according to the different intervention conditions. The lung tissue pathology was observed by HE staining. The expressions of interleukin-4(IL-4)and interleukin-13(IL-13)proteins in bronchoalveolar lavage fluid(BALF)and nuocyte cells supernatant were detected by ELISA. The numbers of nuocyte cells in BALF were detected by flow cytometry. The expressions of α-smooth muscle actin(α-SMA)and I collagen(collagen-I)were detected with immunohistochemistry, real-time PCR and Western blotting. Results Compared with the control mouse group, expressions of IL-4 and IL-13, numbers of nuocyte cells in BALF, expressions of α-SMA and collagen-I increased in asthma mouse group(P<0.05), while those in anti-IL-25 mouse group had no significant differences(P>0.05). Compared with the control cell group, expressions of IL-4 and IL-13 increased significantly in the cells supernatant in IL-25 cell group(P<0.05), while those in anti-IL-25 group had no significant differences(P>0.05). Conclusion IL-25 may promote the secreting of TH2 cytokines by nuocyte cells, thus leads to the airway remodeling of the asthma mice.
    Influencing factors for the graft patency of radial artery after coronary artery bypass grafting surgery

    Objective To analyze the influencing factors for the graft patency of radial artery(RA)after coronary artery bypass grafting(CABG). Methods Clinical data of 65 patients who adopted RA grafts in isolated CABG and underwent computed tomography angiography one year after operation were retrospectively analyzed, including preoperative, intraoperative and postoperative data. Multiple logistic regression analysis was performed to explore the influencing factors for RA graft patency. Results Native stenosis of target vessel ≥80%(OR=0.212, 95%CI: 0.049-0.912, P=0.037), and target vessel in the left anterior descending artery territory(OR=0.104, 95%CI: 0.012-0.921, P=0.042)were independent protective factors for RA graft patency, while non-adherence to the postoperative combination of anti-spastic medications(OR=6.825, 95%CI: 1.857-25.083, P=0.004)was an independent risk factor. Conclusion Proper choice of target vessels during the operation and combination of anti-spastic medications after operation are independent influencing factors for RA graft patency.
    A comparative study of total thoracoscopic mitral valve replacement under heart beating and heart arrest

    Objective To compare the efficacy and safety of total thoracoscopic cardiac surgery under heart beating and heart arrest. Methods In the heart beating group(n=19), extracorporeal circulation was established through femoral artery and femoral venous cannula. Mitral valve replacement was implemented under the condition of light hypothermal extracorporeal circulation and aortic cross-clamping on heart beating. In the heart arrest group(n=12), extracorporeal circulation was established through femoral artery and femoral venous cannula. Mitral valve replacement was implemented under the condition of middle hypothermal extracorporeal circulation and aortic cross-clamping with cardial arrest. The clinical parameters of the two groups were compared. Results All operations were successful without low cardiac output syndrome, severe arrhythmia, air embolism, perioperative death, and other serious complications. Heart beating group had longer operation time and aorta blocking time than heart arrest group(P<0.05), but shorter in vitro cycle time, ICU monitoring time, and postoperative hospital stay, and lower medical costs(P<0.05). After in vitro cycle started, the levels of myocardial enzymes increased; CK-MB and cTnT reached the peak value 24 h after operation and then declined, while LDH showed an uptrend. There were statistical differences between the two groups in the levels of enzymes at the same time points(P<0.05). Conclusion Total thoracoscopic mitral valve replacement under heart beating has advantages of quick recovery, lower hospitalization cost, better myocardial protective effect and more patient benefits compared with total thoracoscopic mitral valve replacement under heart arrest. The combination of the two approaches has wide applicative value in clinical practice.
    Effects of different cryoprotectants and cooling methods on the histology and immunogenicity of heart valves

    Objective To explore the proper cryoprotectants and cooling methods for storing heart valves in liquid nitrogen. Methods Each heart valve from 6 healthy donors were divided into 7 specimens: 1 was observed and tested directly as control group, and other 6 were divided into 3 cryoprotectant groups(Group A, B and C), with 2 specimens in each groups. Two cooling methods were applied, including -80 ℃ direct cooling(G1 groups)and programmed cooling(G2 groups). The histological changes were observed with hematoxylin-eosin staining, Weigert-Van Gieson(ET+VG)staining, and Masson Trichrome staining. The ultrastructural changes were observed with transmission electron microscopy(TEM). The expression of human leukocyte antigen(HLA)was detected with Western blotting and immunohistochemistry. The immunogenicity of each group was compared. Results Cryopreservation injured the organizational structure of valves, while valves stored in DMSO and chondroitin sulfate sodium showed the least lesions and obviously down-regulated expression of HLA-I. Collagen fibrils were less stained, while the orientation was more disorderly in the programmed cooling groups than in the -80 ℃direct cooling groups, but the expression of HLA-I was lower. Conclusion DMSO in combination with chondroitin sulfate sodium can best preserve the heart valves, with the lowest immunogenicity. The method of -80 ℃direct cooling is conducive for the preservation of collagen fibrils, but it produces higher immunogenicity than programmed cooling.
    Treatment timing of non-infarct-related artery in patients with multi-vessel disease and acute myocardial infarction

    Objective To explore the timing of percutaneous coronary intervention(PCI)for non-infarct-related artery(Non-IRA)in patients with acute myocardial infarction(AMI)and multi-vessel disease(MVD). Methods A total of 357 cases of AMI and MVD were divided into 5 groups according to whether and when they underwent PCI for Non-IRA: control group(n=117, who underwent PCI for culprit vessel only), MV-PCI group(n=32, who underwent PCI simultaneously for both culprit vessel and Non-IRA), 0-7d group(n=28, who underwent PCI for Non-IRA within 7 day); 8-30 d group(n=84, who underwent PCI for Non-IRA within 8-30 days), 31-60 d group(n=96, who underwent PCI for Non-IRA within 31-60 days). All patients were followed up for 2 years. Clinical characteristics, data collected during surgery and major adverse cardiac event(MACE)were analyzed. Results The incidence of in-hospital MACE was the highest in the MV-PCI group(18.8%), the lowest in the control group(3.4%), and the second lowest in the 31-60 d group(P=0.02). The incidence of follow-up MACE was the highest in the control group(59.8%)(P<0.01), and the cumulative rate of MACE was the lowest in the 31-60 d group(18.80%)(P<0.01). Conclusion PCI of Non-IRA can improve the prognosis. Emergency PCI for simultaneous culprit vessel and Non-IRA is risky. Its advisable to choose PCI for Non-IRA within 31-60 days after AMI.
    Effects of perindopril on the expression of plasma miR-423-5p and cardiac function in patients with heart failure

    Objective To explore the effects of perindopril with different doses on the expression of plasma miR-423-5p in patients with chronic heart failure(CHF)and its correlation with the cardiac function. Methods A total of 75 CHF patients were randomly divided into 3 groups: routine group(n=25, treated with routine medicine), low-dose group(n=25, treated with 4 mg/d perindopril), and high-dose group(n=25, treated with 8 mg/d perindopril). Another 25 healthy controls were enrolled in the control group. After 4 weeks of treatment, pre- and post-treatment levels of LVEDD, LVEF, NT-proBNP, and miR-432-5p of all subjects were measured. After 4 weeks of treatment, the level of LVEF of the routine, low-dose and high-dose groups remarkably increased(P<0.05), while the levels of LVEDD, NT-proBNP and miR-423-5p remarkably decreased(P<0.05). After treatment, the LVEF of the low-dose and high-dose groups was significantly higher than that of the routine group(P<0.05), while the LVEDD, NT-proBNP and miR-423-5p levels were significantly lower than those of the routine group(P<0.05). Compared with the low-dose group, the high-dose group had more significant different levels of LVEF, LVEDD, NT-proBNP, and miR-423-5p(P<0.05). Conclusion Perindopril can reduce the plasma miR-423-5p expression in CHF patients, and improve the cardiac function in a dose dependent manner.
    Clinical studies on type 2 diabetic patients treated with sitagliptin

    Objective To analyze the factors that affect the efficacy of sitagliptin in the treatment of type 2 diabetic mellitus(T2DM)patients. Methods Clinical data of 86 T2DM patients who treated with sitagliptin during Sept.2012 and June 2014 were collected. According to the levels of fasting C peptide, the patients were divided into two groups: maximum C peptide group(n=12)and minimum C peptide group(n=12). Multiple line regression analysis was performed to analyze the factors that might affect the efficacy of sitagliptin. Results After administration of sitagliptin, there was no significant difference in the decrease of fasting blood glucose(FBG)and 2 h post prandial blood glucose(2h-PBG)between the maximum and minimum C peptide groups(P>0.05). Multiple line regression analysis showed that HbA1c was an independent factor that affected the decrease of FBG; baseline 2h-PBG level was an independent factor that affected the efficacy of sitagliptin on 2h-PBG. Sitagliptin significantly decreased glucose excursion(P<0.05), and reduced episodes of hypoglycemia. Conclusion Sitagliptin decreases both FBG and 2h-PBG. The efficacy of sitagliptin may not entirely depend on C peptide levels. Sitagliptin significantly decreases blood excursion and hypoglycemia.
    Effects of estradiol on Th1 cell polarization and liver fibrosis

    Objective To explore the effects of estradiol(E2)on Th1 cell polarization and liver fibrosis. Methods A total of 47 patients with liver fibrosis were divided into 2 groups according to the level of E2: group A(elevated E2 level, n=19)and group B(normal E2 level, n=28). Another 30 healthy people were enrolled as the negative controls(group C). The T lymphocyte subsets were measured with flow cytometry. The levels of hyaluronic acid(HA), laminin(LN), procollagen type Ⅲ amino-terminal peptide(PⅢP)and type IV collagen(CⅣ)were detected with chemiluminescence. The levels of IFN-γ, IL-2, IL-4 and IL-10 were measured with ELISIA assay. Then the parameters of the three groups were compared. Results The E2 level was higher in group A than in group C and group B(P<0.001). The levels of HA, LN, PIIIP and CⅣ in group A were all higher than those in group C(P<0.001, P=0.001, P=0.006, P<0.001), and the levels of these four parameters in group B were also higher than those in group C(all P<0.001). The level of PⅢP was higher in group A than in group B(P=0.045). The percentage of CD3+ T cells in group A was lower than that in group C and group B respectively(P<0.001, P=0.001), and the percentage of CD4+ T cells was also lower than that in group C and group B(P<0.001, P=0.024), while the percentages of CD3+ T cells and CD4+ T cells in group B were all lower than those in group C(P=0.002, P<0.001); The percentage of CD8+ T cells in both group A and groupB was higher than that in group C(P=0.001, P=0.012). The level of IL-4 in group A was lower than that in group C and group B(P=0.002, P=0.016), and the level of IL-10 was also lower than that in group C and group B(P<0.001, P=0.012). Conclusion E2 played a dual role in the formation of liver fibrosis. Elevated level of E2 accelerated liver fibrosis, and affected the immune function via Th1 cell polarization.

    Objective To analyze the cervical biopsy results in patients with negative cytological but positive high-risk HR-HPV test results, in order to explore the application of Cervista HPV HR A9 group virus in the identifying of such patients. Methods A total of 1,376 women who took physical examinations in our hospital during Jan. 2011 and Jan. 2016 received Cervista HR HPV test plus ThinPrep cytology(TCT). They were then divided into 3 groups according to the results of Cervista HR HPV test: A5/6, A7, and A9 groups. Those who had negative cytological but positive high-risk HPV test results underwent colposcopy or colposcopic biopsy if necessary. The correlation between A9 group virus infection and cervical high-grade intraepithelial neoplasia was analyzed. Results The positive rate in the A5/6, A7 and A9 group was respectively 28.41%, 21.51% and 28.41%. The incidence of CINII+ was 1.28%, 3.38% and 15.82%, respectively. The incidence of CINII+ in A9 group was obviously higher than that of the other two groups, and the difference was statistically significant(P<0.001, P<0.001). The risk of CINII+ in A9 group was 12.37 times of that in A5/A6 group and 4.68 times of that in A7 group. Conclusion Cervista HPV HR A9 group virus can be used to identify patients with negative cytological but positive high-risk HPV.

    Objective To explore the correlation between lymphocyte-to-monocyte ratio(LMR)in peripheral blood and prognosis of malignant pleural mesothelioma(MPM). Methods Clinicopathological data of 85 patients with MPM were retrospectively analyzed. According to the LMR in peripheral blood, the patients were divided into two groups: high LMR group(n=27)and low LMR group(n=58). The clinicopathological data and overall survival of the two groups were compared. The correlation between the clinicopathological data and the overall survival was analyzed with Cox regression model. Results Significant differences were observed between the two groups in gender, performance status(PS)score, absolute lymphocyte count and absolute monocyte count(P<0.05). The median overall survival of patients with high LMR was significantly longer than that of patients with low LMR(27 months vs 9 months, P<0.001). Of patients with various histological types and tumor stages, the median overall survival of patients with high LMR was significantly longer than that of patients with low LMR. Multivariate analysis showed that LMR was an independent prognostic factor associated with prognosis of MPM(HR: 0.277; 95% CI: 0.142-0.538; P<0.001). Conclusion LMR in peripheral blood is a biological marker for the prognosis of MPM.

    Objective To investigate the relationship between exhaled nitric oxide(FeNO)/impulse oscillometry(IOS)and small airway function in asthmatic patients. Methods A total of 140 asthma patients were divided into small airway normal function group(n=69)and small airway dysfunction group(n=71). FeNO, eosinophil(EOS)count and total immunoglobulin E(IgE)in peripheral blood and pulmonary function, as well as IOS were measured. Results The levels of FeNO, IgE, reactance area(AX), resonant frequency(Fres)and EOS in small airway dysfunction group were significantly higher than those in small airway normal function group(all P<0.01). FeNO, AX, Fres and EOS were correlative factors of mid forced expiratory flow of percentages of predicted values(FEF25%-75%pred)(r=-0.856, P<0.001; r=-0.851, P<0.001; r=-0.398, P=0.001; r=-0.288, P=0.014)and the turns of standard errors of partial regression coefficient were as follows: FeNO>AX>Fres>EOS. The turns of predictive value in diagnosis of small airway dysfunction were as follows: FeNO combining AX and Fres>FeNO>AX>Fres>EOS. Conclusion FeNO and IOS may be highly sensitive and specific in diagnosis of small airway dysfunction, and their combination can better evaluate the function of small airway in asthmatic patients.

    Objective To explore the value and safety of electronic bronchoscope in diagnosis and treatment of dyspnea in newborns. Methods A retrospective study including 122 newborns with bronchoscopy was performed, 101 of which were suspicious congenital airway dysplasia(suspicious dysplasia group), and 21 curing pulmonary atelectasis(atelectasis group)by bronchoalveolar lavage. Results In suspicious dysplasia group, there were 52 newborns with airway dysplasia, including laryngomalacia(25/101, 24.8%), tracheomalacia(18/101, 17.8%)and vocal cord paralysis(15/101, 14.9%), and 25 of which had multiple airway dysplasias; as opposed to 49 newborns without airway dysplasia. In atelectasis group, 21 newborns with pulmonary atelectasis were cured by bronchoalveolar lavage, 3 of which accompanied by congenital bronchomalacia. Conclusion Bronchoscope has the dual function in newborn dyspnea examination and treatment with high safty.
    Stoppa approach in the treatment of anterior pelvic ring fractures complicated with posterior urethral disruption

    Objective To evaluate the feasibility and efficacy of Stoppa approach in the treatment of anterior pelvic ring fractures complicated with posterior urethral disruption. Methods Clinical data of 16 cases of anterior pelvic ring fractures complicated with posterior disruption during January 2012 and July 2015 were retrospectively analyzed. All patients were treated with emergency operation with the Stoppa approach of open reduction and internal fixation of the pelvis combined with urethral realignment by traction and primary urethra suturing or partial suturing. All patients were followed up for 6 to 36 months. The reduction of fracture and recovery of function were evaluated with Tometta and Majeed score. The urethral repair effect was evaluated with postoperative clinical examination and patients subjective feelings. Results The mean length of incision was 12 cm. The average operation time was 136 min. According to the Tometta score classification, the results were excellent in 11 cases, good in 3 cases, and fair in 2 cases. According to the Majeed score classification, the results were excellent in 12 cases, good in 3 cases, and fair in 1 case. After the urinary catheter was removed, 12 patients achieved good urination, with the mean maximal urine flow being 18.5 mL/s; 4 patients developed dysuria; 6 patients had penile erectile difficulties; and 2 patients had urinary incontinence. Conclusion The Stoppa approach is a safe approach, which can provide clear surgical field to repair the urethral disruption and fix the anterior pelvic ring fractures. It can be an ideal approach in the treatment of anterior pelvic ring fractures complicated with posterior urethral disruption.

    Objective To investigate the relationship between food intolerance and major adolescent oral ulcer. Methods Eating habits of 45 children with major adolescent oral ulcer were investigated during 2008 and 2014. The levels of IgG against 14 common food were measured by ELISA. Immunology detection was conducted. Another 20 children with minor recurrent oral ulcer were selected as controls. Results Major adolescent oral ulcer often presented in patients aged 6 to 12 years and boys were more vulnerable. The tongue tip and buccal fat pad were the preferred sites. The ulcer was big and deep. The center was in depression surrounded by pale, uplifted tissues. The patients felt severe pain. All children with major adolescent oral ulcer ate unbalanced diet. Total positive rate of serum food-intolerant IgG antibodies was 90.6% and 45% in children with major adolescent oral ulcer and children with minor recurrent oral ulcer. Among these 14 food, the positive rate of egg was the highest, and it was higher in major adolescent oral ulcer than in minor recurrent oral ulcer with statistical significance(P<0.05). The immunity of children with major adolescent oral ulcer was almost normal, and 55.6% children had CD4+ T cell lower than normal. The pathology was infiltration of chronic inflammatory cells and the thickening of vessel walls. After diet changes and drug treatment, 44 childrens ulcer was under control, the total effective rate being 97.8%. Conclusion Major adolescent oral ulcer is closely related to food intolerance. Egg and milk are the most common intolerant food. Avoiding the intolerant food and drug treatment show acceptable efficacy in the treatment of most children with major adolescent oral ulcer.
