Correlation between cell apoptosis and obliterate bronchiolitis in rats following allogeneic lung transplantation

Abstract: To explore the correlation between apoptosis and obliterate bronchiolitis (OB) after allograft lung transplantation in rats. MethodsHeterotopic trachea transplantation was performed on Wistar rats with OB after allograft lung transplantation. Animal models were randomly divided into the control group, the transplantation group and the immunosuppressant treatment group. All allograftic trachea were taken out on the 7th, 14th and 28th d after transplantation. TUNEL was used to detect apoptosis, and an EnVison immunohistochemical method was used to detect expressions of Caspase3 and Bax in the grafts. Results① Animal models showed the same histological changes as OB after allograft lung transplantation. ② TUNEL test showed that apoptosis in the OB model and the immunosuppressant treatment group had less apoptosis than that in the transplantation group(P<0.05). ③ HIS showed that expressions of Caspase3 and Bax were increased in OB models and were less in the immunosuppressant treatment group than in the transplantation group, especially in the later period(P<0.05). ConclusionOB after allograft lung transplantation is related to apoptosis. Suppressing apoptosis maybe a treatment method for OB after allograft lung transplantation.
