Effects of corticotropin releasing factor on the intestinal goblet cell secretory productions

Abstract: Objective To investigate the effects of corticotropin releasing factor(CRF)on the intestinal goblet cell secretory productions. Methods LS174T cells were treated with different concentrations of CRF to determine the optimum concentration. After that, LS174T cells were treated with 10-10mol/L CRF. Real-time qPCR, Western blotting, immunofluorescent staining and ELISA technique were used to examine the expression levels of intestinal goblet cell secretory productions. Results Real-time qPCR results showed that CRF decreased the mRNA expressions of mucin2(MUC2), fucosyltransferase-2(FUT2)and trefoil factor-3(TFF3)in LS174T cells at 6 h(P<0.05), compared with the PBS control group. Immunofluorescence and ELISA results showed that CRF reduced the protein expression of MUC2(P<0.05)at 6 h. Western blotting results showed that FUT2 and TFF3 protein levels were decreased(P<0.05)in LS174T cells treated with CRF for 24 h. Conclusion CRF could damage the intestinal mucosal barrier by altering the expression of the LS174T cell secretory productions.
