Effects of high exposure of endogenous reproductive hormones on perimenopausal and postmenopausal blood pressure in rats

Abstract: Objective To explore the effects of high-dose endogenous estrogen(17-β E2)and progesterone(P)of the reproductive phase on the blood pressure of perimenopausal and postmenopausal rats and to observe the role of angiotensin II type-1 receptor(AT1R). Methods Bilateral ovariectomy was performed during the diestrus phase of estrus cycle to induce rat models of artificial estrous cycle by 17-β E2 and P. The models were divided into 3 groups: high-dose estrogen plus low-dose progesterone group(n=20, HELP), high-dose progesterone plus low-dose estrogen group(n=20, HPLE), and ovariectomized group(n=20, OVX). The experimental animals were given hormones for 8 weeks, decreasing dosages for 5 weeks and withdrawal hormones for 6 weeks to imitate the reproductive phase, perimenopausal phase and postmenopausal phase respectively. The changes of blood pressure, serum E2 and P, and protein and mRNA expressions of AT1R in the abdominal aorta were monitored. Results Compared with the OVX group, the HELP and HPLE groups had higher systolic blood pressure(SBP)in all phases(P<0.05); the HELP group had higher diastolic blood pressure(DBP)in the perimenopausal and postmenopausal phases(P<0.05), and higher protein and mRNA expressions of AT1R in the reproductive and perimenopausal phases(P<0.05). The protein expression of AT1R was positively correlated with serum concentration of E2 and SBP(P<0.01).
