Progesterone combined with octreotide inhibits the growth of human non-small cell lung cancer A549 cells

Abstract: Objective: To investigate the regulation of somatostatin recepter (SSTR) subtype mRNAs expression of non-small cell lung cancer cell line A549 by progesterone and the antiproliferation of progesterone combined with octreotide on A549 cells. Methods: The progesterone receptor was detected by the immunohistochemical method, and SSTR subtype mRNAs were determined by the reverse transcription polymerase chain reaction in cells pretreated or non-pretreated by progesterone. The cell viability was evaluated by MTT assay. Results: Progesterone receptor and SSTR1, SSTR2, SSTR4 and SSTR5 were revealed in A549 cells, but SSTR3 was not. Following 24-hour treatment by 10nmol/L progesterone, SSTR3 was detected in A549 cells. Up to 48 hours, SSTR3 was still up-regulated(P=0.012). Progesterone combined with octreotide was superior to progesterone or octreotide alone in inhibiting the growth of A549 cells(P<0.05). Conclusion: Progesterone up-regulates the expression of mRNA for SSTR subtypes in A549 cells. Progesterone enhances the ability of octreotide to inhibit the growth of A549 cells.
