To investigate the expression of Tribbles (TRBs) in human monocytedriven macrophages and to analyze the regulative role of oxidized low density lipoprotein (ox-LDL) on TRB1, TRB2 and TRB3. MethodsPrimary human monocyte-driven macrophages were cultured in vitro. Macrophages were incubated with ox-LDL and low density lipoprotein (LDL) at various concentrations for various times. The level of TRBs mRNA was determined by quantification real-time RT-PCR. ResultsAll three TRBs genes (TRB1, TRB2 and TRB3) were expressed in human monocyte-driven macrophages, among them, TRB3 was predominant, and TRB1 and TRB2 were little expressed. OxLDL increased the levels of TRB3 andTRB2 mRNA, while it decreased the expression of TRB1 in a dose-and time-dependent manner. Conclusions All three TRBs are expressed in human monocytedriven macrophages and can be regulated by ox-LDL, so TRBs, especially TRB3 may participate in the course of macrophage apoptosis.

To explore the effects of dehydroepiandrosterone (DHEA) on the development of experimental autoimmune neuritis (EAN) and to explore its mechanism. Methods Thirty-six Lewis rats were randomly divided into the DHEA 0.5mg treatment group, the DHEA 2mg treatment group and the control group (n=12). The treatment groups were subcutaneously injected every day with DHEA and the control group was subcutaneously injected with the same volume of DHEA dissolvent from day 5 post-immunization (p.i.) to day 28 p.i. with bovine peripheral myelin(BPM)in Freund′s complete adjuvant (CFA). The effects were assessed in terms of appearance of clinical signs, clinical score, histopathology and IFN-γ and TNF-α positive cells in sciatic nerve sections, and T-cell proliferation and inflammatory cytokines (IFN-γ,TNF-α and IL-10) synthesis by draining lymph nodes and spleen cells. ResultsRats receiving DHEA in the two different therapeutic regimens displayed a significant delay in onset (P<0.05 and P<0.05), a decreased inflammatory cell infiltration into the PNS (P<0.05,P<0.05) and decreased numbers of IFN-γ and TNF-α expression cells in the PNS(P<0.05 and P<0.05 ), and reduced BPMstimulated T cell proliferation(P<0.05 and P<0.05) and IFN-γ and TNF-αsecretion in the draining lymph node and spleen (P<0.05, P<0.05) compared to the control group. Only the DHEA 2mg treatment group had the significant differences of supernatant IL-10 were found among the different treatment and control groups(P>0.05).ConclusionAdministration of exogenous DHEA ameliorates the severity of EAN, presumably by suppressing the proliferation of auto-reactive T-cells and moderating the over-expression of pro-inflammatory cytokines. DHEA 2mg therapy is more effective than DHEA 0.5mg therapy.

To investigate factors affecting calcium content in aortic valved homograft(AVH) tissues in rats. Methods90 Wistar rats were randomly allocated into 3 groups: two allogene groups and one isogenenic group. Allogene group: SD→Wistar, one group received AVH of cryopreservation, the other group received AVH preconditioned with dendritic cell monoclonal antibodies(DCmAb )diluted by 1:5 and cryopreserved, and then was treated with DcmAb. Isogenenic group: Wistar→Wistar. AVH with myocardial cuff was heterotopically allografted onto the abdominal aorta of rats. Rats were postoperatively sacrificed in batches at 2, 4, 8, 12 and 16 weeks. Blood samples were obtained for detection of the TCRα, β and CD28 expressions, and the AVH specimens were obtained for detection of the changes of endotheliocyte and smooth muscle cells with a light and electron microscope, and the CD54 expression by immunohistochemistry. Also, the calcium contents of the AVH tissues after transplantation were determined. Results① Compared with the isogenic group, the expression of TCR-α, β and CD28 in the allogene group was significantly increased at each time point(P<0.01). Compared with the cryopreservation group, the expression of TCR-α, β and CD28 in the DCmAb treatment group was significantly decreased(P<0.01). ② The calcium contents of AVH tissues in the allogene group gradually increased at 4 weeks, which was significantly different from that in the isogenenic group over 5 different periods (P>0.05). In the cryopreservation group, the calcium contents of the AVH tissues were 2856±79 and 3587±168 at 4 and 8 weeks post-operatively; were 2176±210, 3089±176 in the DCmAb treatment group; and were 860±60, 870±50 in the isogenic group. Conclusion Immunosuppressive treatment has obviously effect in lightening the immune rejection response and delaying the AVH calcification course.

To investigate the role of peroxisome proliferator-activated receptor γ (PPARγ) in the process of diabetes myocardial fibrosis in rats. MethodTwentyseven male Wistar rats were randomly divided into two groups: the DM group (n=15), which were induced by a single intraperitoneal injection of STZ (65mg/kg body wt) and the control group (n=12). All rats were fed normally for 5 months and eleven DM rats survived (n=11). The radiate escape pestilence method was used to determine the level of plasma angiotensin II (Ang II). HE staining and Masson staining were used to test collagen. Also, immunohistochemistry was used to determine the protein expressions of PPARγ and Ang II. ResultsThe plasma AngⅡ level was higher in the DM group. Cardiac collagen deposition was obviously enhanced in the DM groups compared with the control groups. Expression of PPARγ in the DM group was higher than that in the control group, especially in the capillary endothelium and the myocardial cells. The expression of AngⅡ in the cytoplasm of myocardial cells in the DM group was higher than that in the control group. Conclusion The DM animal model was successfully established. Expression of PPARγ in myocardial cells and capillary endothelium significantly increases in diabetes rats. PPARγ may play an important role in the process of diabetes myocardial fibrosis.

To explore the effects of pioglitazone on the plasma ghrelin levels and the ghrelin mRNA expression in the stomach of type 2 diabetic rats. MethodsWistar rats were fed with a high fat diet for 8 weeks and then treated with a low dose of STZ(30mg/kg) by intraperitoneal injection to establish type 2 diabetic rat models, then they were divided into three groups: the control group(NC, n=10), the diabetes group(DM, n=12) and the pioglitazone treatment group(DM-PIO, n=12). Rats in the DM-PIO group were treated with pioglitazone(20mg·kg-1·d-1) by intra-gastric administration. All the animals were sacrificed 8 weeks later. The blood glucose, lipids, insulin, leptin and ghrelin were determined, and the ghrelin mRNA expression in the stomach was determined by RT-PCR. ResultsThe DM and DM-PIO groups had significant reduced plasma levels of ghrelin and mRNA expression levels in the stomach compared with the NC rats (P<0.05 for each group). Plasma ghrelin and mRNA expression levels were higher in the DM-PIO rats than in the DM rats, which was not significant (P>0.05). By the correlation analysis, plasma ghrelin levels were negatively correlated with body weight (P<0.05), HOMAIR (P<0.01) and leptin P<0.05) in diabetic rats. ConclusionThe plasma ghrelin levels and the expression of ghrelin in diabetic rats are significantly reduced. Pioglitazone can not change the plasma levels of ghrelin and its expression in the stomach.

To explore the changes of transcranial magnetic stimulation motor evoked potentials(TMSMEP) and spinal cord pathology in rabbit spinal cord injury, and to explore their correlation. MethodsA digital weight drop apparatus induced spinal cord lesion was performed to mimic human SCI. Based on the drop height, 32 rabbits were randomly divided into four groups: the control, 2cm, 4cm and 6cm injured groups. TMSMEP was used to assess neurologic function. The MEP was recorded at 5min, 15min, 30min, 60min, 6h, 24h, 3d, 7d post-injury and before spinal cord injury in injury groups. Tarlov behaviour score was performed before and after surgery at 1, 3, 7 days. Light and electron microscope were used to observe the pathological changes. Results Spinal cord injury resulted in a gradual decrement in the peak latency and significant increment in the peak amplitude. Simultaneously, histological studies showed that the drop causes a ischemia lesion, which was found in human spinal cord injury. ConclusionTMSMEP has a significant association with spinal cord pathological changes and will be a noninvasive examination for spinal cord function.

To investigate whether N-acetylcysteine has protective effects on oxygen-glucose deprivation injured cortical astrocytes in rats. Methods The fourth passage of the cortical astrocytes of primary cultured neonate rats was used. Na2S2O4 and sugar free Earle′s solution were used to establish the chemical oxygen glucose deprivation (OGD) model, which followed by changing the culture into complete mediums to mimic ischemia/reperfusion injury. The 3-(4,5-dimethylthialzol-2-yl)-2-5 -diphenyl-tetrazolium bromide (MTT) method and the lactate dehydrogenase (LDH) leakage method were used for evaluations of cell viability and cell lysis, and the Colorimetric method was used to determine changes of cerebral energy metabolism. Results Compared with the sham-group, the cultured neonate rat cortical astrocytes were subjected to one hour of OGD followed by 24 hours of reperfusion, then the cell viability was decreased and the cell lysis was significantly increased, and the cellular energy metabolism was disordered. Pre-treatment with N-acetylcysteine can dose-dependently reverse these injuries. Conclusion N-acetylcysteine exhibits protective effects on OGD/reoxygenation injuries in rat cortical astrocytes.

To study the expression of Syk and its correlations with clinical pathology and angiogenesis in pancreatic adenocarcinoma (PAC). Methods Forty PAC, 9 chronic pancreatitis and 8 normal pancreatic tissues were collected. The expressions of Syk protein in these tissues were assayed by immunohistochemical staining, and also expressions of Syk mRNA in PAC were investigated by realtime PCR. MVD marked by CD34 was detected by morphometric analysis, and the relationship between MVD and clinical pathology of PAC was analyzed. ResultsThe positivity rate of Syk protein in PAC, chronic pancreatitis and normal pancreatic tissues was 27.5%, 77.8% and 100%, respectively. There were 29 samples with negative Syk protein, and among them the Syk levels had no relations to the cancer position, but had relations to the TNM classification, tumor differentiation and lymphotic metastasis(P<0.05). The MVD was higher in positive Syk proteins than in negative ones (P=0.000). ConclusionsThe expression of Syk in pancreatic cancer is significantly lower than that in chronic pancreatitis and normal pancreatic tissues and is related to the development of pancreatic cancer. MVD has significant relationship with differentiation and histology types of PAC.

To develop a closedloop feedback controlled volatile anesthesia system with index (BIS) as the control variable and to clinically validate it. MethodsIn the first stage, a closedloop feedback controlled volatile anesthesia system was set up. A volatile anesthetic physiological pharmacokinetics model was built in the system software, whose controlled algorithm was based on the proportional-integral-derivative principle. System software, anesthesia deep monitor, syringe pump and anesthesia machine made up the whole administrate system. In the second stage, enflurane, isoflurane or sevoflurane was used in clinical operations to verify the performance of the administrate system for patients with general surgeries, single side lung venting or obesity. ResultsThe close-loop feedback administrate system can maintain BIS around the target value and retained hemodynamics stabilization. This system reduced the inducement and analepsia time. All patients administrated by this system had not encountered awareness during general anesthesia. ConclusionThe system is able to provide clinically adequate anesthesia using the commonly used volatile anesthetics even when patients′ pharmacokinetics parameters have evident changes.

To explore the force system of different precision attachments in mandible distalextension edentulous by a finiteelement model. MethodsThe threedimensional finite element stress analysis method was used. Results The effects of attachments of ASC-52 were reduced in abutment when the molar was forced by 200N. When the molar was forced by 200N, the effection of the attachment ASC-52 was much more reduced than the attachment of the revax system. When the premolar was forced by 200N, the effection on attachments was reduced either liable or lingual. When the double molars were forced by 200?N, the stress intruding was added in mandibles and abutments. ConclusionsThe labial effection of abutments is smaller than lingual in the attachment of the revax system or ASC-52. The effection of the ASC-52 attachment is reduced in the abutment when the molars are forced.

MethodsAged patients with hypertension were divided into two groups: the control group(n=177)(UAER<30?μg/min) and the MA group(UAER 30-200μg/min) (n=179). Incidence of retinopathy, peripheral neuropathy, and lower extremity arterial sclerosis occlusion were determined, also the levels of plasma vonWillebrand factor (vWF) and endothelin-1 (ET-1) were determined. ResultThe incidences of retinopathy, peripheral neuropathy and lower extremity arterial sclerosis occlusion were significantly higher in the MA group than those in the control group (P<0.05). The plasma vWF level was markedly increased in the MA group (202.07±73.30)% as compared with that in the control group (118.95±46.72)% (P<0.01), also the plasma ET-1 level in the MA group (79.13±14.04) μg·L-1 was significantly higher than that in the control group (50.60±12.39) μg·L-1. The plasma vWF level was significantly correlated with UAER (r=0.72,P<0.01) and the plasma level of ET-1 was closely associated with vWF (r=0.69,P<0.01). ConclusionAged patients with hypertension with MA have more remarkable endothelial dysfunction than those without MA. The increase of UAER is correlated with the degree of endothelial dysfunction.

To explore the effects of remifentanil combined with propofol or etomidate in providing conscious sedation for awake craniotomy in eloquent cortex surgery. MethodsThirty patients undergoning glioma removal in the cerebral cortical functional area were randomly divided into two groups: the etomidate group and the propofol group (n=15). The status of eloquent areas mapping, respiratory variables and hemodynamic stability were assessed, also the recall time and complications were documented. Results There were no differences in status of eloquent areas mapping or hemodynamic and respiratory variables between the two groups. Respiratory complications occurred in 1 patient in the propofol group, but there were significant differences in recall time between the two groups (etomidate, 321?s; propofol, 410?s). ConclusionEtomidate and propofol are good in the status of eloquent areas mapping, but etomidate is superior to propofol in recall time.

To explore the therapeutic effects of Yinxingdamo combined with Lovastatin in treatment of acute cerebral infarction. Methods 127 patients with acute cerebral infarction were randomly divided into three groups. The Chuanxiongqin group was subjected to Chuanxiongqin injection 200mg+Ns250mL, ivdrip, Qd, for 14 days; the Yinxingdamo group was subjected to Yinxingdamo injection 20mL+NS250mL, ivdrip, Qd, for 14 days; the combination group was subjected to the Yinxingdamo injection 20mL+NS250mL, ivdrip, Qd, and Lovastatin 40?mg, PO, QN. 14 days later, Yinxingdamo was disused and Lovastatin was continued. The therapeutic effects were evaluated after 30 days. ResultsThe response rate of the combination group was higher than that of the Yinxingdamo group(P<0.05) and the Chuanxiongqin group(P<0.01). The degree of NFD and the levels of serum lipids and whole blood viscosity, and the plasma viscosity were significantly decreased(P<0.01). However, the HDLC level was significantly increased(P<0.01). ConclusionLovastatin combined with Yinxingdamo has obvious therapeutic effects on acute cerebral infarction, and it can improve the neurological function and decrease the levels of serum lipids and blood viscosity.

To study the value of laserinduced autofluorescence on the diagnosis of gastric cancer and its precancerosis. MethodsEndoscopic laserinduced fluorescence diagnosis spectra were taken in 198 patients with upper gastrointestinal symptom. Results198 patients were diagnosed by endoscopy and biopsy, including 20 gastric cancer, 22 gastric precancerosis,156 gastrelcoma and chronic superficial gastritis. Characteristic peaksof autofluorescence in cancerous tissue were observed in 16 out of 20 patients with gastric cancer and 19 out of 22patients with gastric precancerosis. The coincidence was 80.0% and 86.4% respectively. While 141 had no such peaks out of 156 patients with gastrelcoma and chronic superficial gastritis. ConclusionLaserinduced autofluorescence spectra have higher sensitivity and specificity in the diagnosis of gastric cancer and its precancerosis, and can be used as an effective means of supporting inspections.

To investigate the present situation of nosocomial infection(NI) and the use of antibiotics and to provide scientific data for prevention and control of hospital infection. MethodsForms were filled with case data obtained from clinical investigations and references to inpatient medical records of all patients admitted to Qilu Hospital on 22th August 2007. ResultsAnalysis revealed that the prevalence rate of NI was 5.68% and the case prevalence rate was 5.93%, and the highest NI rate was found in the general ICU (36.36%). The respiratory tract was the main infected site (58.95%). The antibiotic adoption rate was 51.37%, and 59.30% of the antibiotics were singly used. Nine among 31 strains were identified as Escherichia Coli, 8 Pseudomonas aeruginosa, 2 Klebsiella pneumonia and 2 fungi. ConclusionsThe present NI rate survey indicates that targeted surveillance should be carried out in high risk departments, such as ICU, Department of Burns, Hemodia1ysis and Hematology, and it also helps doctors to properly use antibiotics to reduce the NI rate in the hospital.
