Expressions of MiR210 and stat3 following global ischemia in rats and the potential relationship among MiR210, stat3 and HIF-1α

Abstract: Objective To study the expression of signal transducer and activator of transcription 3 (stat3) in hippocampus and the expression of MiR210 in cerebral cortex following global ischemia in rats, and to explore the potential relationship among MiR210, stat3 and hypoxia inducible factor-1α (HIF-1α). Methods Sixty-six SD rats were randomly divided into three groups:sham-operated group, global ischemia group (GI group), and stattic+global ischemia group (stattic+GI group). The global ischemia model was induced by modified four-vessel occlusion method. Stat3, p-stat3 and HIF-1α protein expressions in hippocampus were detected by Western blotting. MiR210 and HIF-1α mRNA expressions in cerebral cortex were detected by Real-time PCR. Results Compared with sham-operated group, stat3, p-stat3 and HIF-1α protein expressions in hippocampus and MiR210 and HIF-1α mRNA expressions in cerebral cortex of GI group significantly increased (all P<0.05). Compared with GI group, p-stat3 and HIF-1α protein expressions in hippocampus of stattic+ GI group significantly decreased, stat3 protein expression showed no statistical difference (P>0.05), and HIF-1α mRNA expression in cerebral cortex signifcantly decreased (P<0.05). Conclusion Expressions of MiR210 and stat3 have a marketable change after global ischemia reperfusion, and regulatory relationships may exist among stat3, MiR210 and HIF-1α.
