Luteolin activates Nrf2 signalling pathway and confers protection against H2O2-induced toxicity in EA.hy926 cells

Abstract: Objective To investigate the effect of luteolin on nuclear factor erythroid-2 related factor 2 (Nrf2) signalling pathway, and evaluate the protection on H2O2-induced toxicity in human umbilical vein EA.hy926 cells. Methods Cells were divided into control, positive drug (sulforaphane) treated, H2O2 treated, luteolin with different concentrations (5,10,20,40 μmol/L) treated, and luteolin with different concentrations (5,10,20,40 μmol/L) and H2O2 cotreated groups. ARE luciferase reporter gene assay, Western blotting analysis and qRT-PCR assay were used for identification of luteolin on activiting Nrf2 signalling pathway; flow cytometry with a fluorescent probe uploading was adopted for detecting the intracellular reactive oxygen species (ROS) levels; MTT assay was carried out for evaluating the protection against H2O2-induced toxicity. Results Luteolin increased the luciferase activity dose-dependently, enhanced both the protein and mRNA levels of Nrf2 and its downstream gene heme oxygenase-1 (HO-1), quinone reductase enzyme NAD(P)H: quinone oxidoreductase 1 (NQO1) and glutamate cysteine ligase modifier subunit (GCLM), and decreased intracellular ROS levels. Co-treatment with luteolin improved cell survival compared with H2O2 treatment alone. Conclusion Luteolin activates Nrf2 signalling pathway in EA.hy926 cells, inhibits H2O2-induced cytotoxicity and confers protection against oxidative-induced injury in EA.hy926 cells.
