Abstract: To explore the role of paclitaxel in inhibiting the fibroblast proliferation after the glaucoma filtering surgery in a rabbit model. Methods: Sixtyfour New Zealand White rabbits were randomly divided into four groups: control groupⅠ, control group Ⅱ, experimental groupⅠ, and experimental groupⅡ. Each right eye was given standard trabeculectomy. The control group Ⅰ received no treatment during trabeculectomy. The other three groups were given 0.3?mg/ml MMC, 0.2?mg/ml paclitaxel, and 0.3?mg/ml paclitaxel respectively for 5 minutes. The filtering blebs were observed and the filtering site was examined by light microscope， and the number of positive cell nucleus were counted under microscope area after immunohistochemical staining at the 3rd， 7th，14th, and 28th day after surgery. Results: The changes of the intraocular pressure， fitering blebs， and number of Ki67 positive cell nucleus in experimental groupⅡwere significantly different compared with those in other three groups(all P＜0.05 )at the 7th， 14th，and 28th day. Experimental groupⅡ had a better effect on inhibiting fibroblast proliferation and had fewer side effects. Injured corneal epithelium and conjunctival wound leak were found in the control groupⅡ. Conclusion: Paclitaxel can inhibit the fibroblast proliferation, decrease the intraocular pressure, and keep the filtering passage unobstructed after glaucoma filtering surgery.
