Evaluation of CD4+CD25+FoxP3+ regulatory T cells and FoxP3 mRNA expression in 20 NMOSD patients

Abstract: Objective To assess the percentage of CD4+CD25+FoxP3+ regulatory T(Treg)cells and the mRNA expression of forkhead box protein 3(FoxP3)in patients with neuromyelitis optica spectrum disorders(NMOSD). Methods The case-control study compared 20 NMOSD patients with 20 healthy controls. The mononuclear cells in peripheral blood were collected. The percentage of CD4+CD25+FoxP3+ Treg cells was measured with flow cytometry using specific monoclonal antibodies recognizing CD4, CD25 and FoxP3 markers. FoxP3 gene expression was determined with real-time polymerase chain reaction. The expressions of transforming growth factor β1(TGF-β1), interferon γ(IFN-γ), and interleukin 10(IL-10)were determined with enzyme-linked immunosorbent assay. Results The percentage of CD4+CD25+FoxP3+ Treg cells and the mRNA expression of FoxP3 were significantly decreased in NMOSD patients compared with those in the controls(P<0.05). NMOSD patients had significantly increased IFN-γ(P<0.05)but decreased IL-10(P<0.05)levels, while there was no significant difference in the level of TGF-β1. Conclusion NMOSD may be associated with an abrogated function of circulating CD4+CD25+FoxP3+ Treg cells and a decreased level of FoxP3 gene expression.
