Fat-derived adipokine disorder in high cholesterol diet rats

Abstract: Objective To investigate the effects of high cholesterol diet on adipokines (leptin, adiponectin, resistin, visfatin and glypican4) levels in serum and visceral adipose tissue (VAT). Methods A total of 60 SD rats were randomly divided into normal diet group (NC, standard diet, n=30) and high cholesterol diet group (HC, high cholesterol diet, n=30). Blood samples and VAT were collected at week 0, 4, 12 and 18, respectively. The levels of fasting blood glucose and fasting serum insulin were measured, and insulin resistance values were calculated. Adipokines in serum and VAT were detected with enzyme-linked immunosorbent assays. Results At week 18, the serum visfatin and adiponectin in the HC group were significantly increased compared with those in the NC group (P<0.05); levels of leptin, visfatin, adiponectin, and glypican4 in VAT were significantly higher in the HC group than in the NC group (P<0.05); insulin resistance was also observed in HC group. Conclusion Long-term excessive cholesterol intake induces disorder of adipokine synthesis and secretion, and such disorder may be correlated to insulin resistance.
