Determination of urinary tribendimidine metabolite-terephthalic acid by HPLC

Abstract: To develop a HPLC method for determination of tribendimidine metabolite terephthalic acid (TPA) concentration in human urine. MethodsDiluted urine was separated on the Ultimate XB-C18 column (4.6mm×200mm, 5μm), eluted with methanol and 1% glacial acetic acid (35∶65, V/V) at 0.8mL/min and 30℃, and detected at 240nm with ferulic acid as an internal standard (IS). ResultsA good linearity was obtained from 1 to 400μg/mL (r=0.9954). The lower limit of quantitation (LLOQ) was 1μg/mL. The relative recoveries were (106.0±4.4)%, (107.4±4.2)% and (108.5±2.4)% at 3, 25 and 200μg/mL, and the intra- and inter-batch relative standard deviations (RSD) were less than 8% for all quality control samples. ConclusionThe developed HPLC method is proved to meet the requirements of biological sample analyses, and is appropriate for determination of urinary tribenmidine metaboliteTPA in human urine.
