热休克蛋白27在脂多糖介导的牙龈成纤维细胞损伤中的表达

Expression of Hsp27 in lipopolysaccharide-induced injury of human gingival fibroblasts

摘要: 目的 观察在不同浓度脂多糖(LPS)干预下, 热休克蛋白27(Hsp27)在人牙龈成纤维细胞(HGFs)损伤过程中的表达。方法 取原代培养并鉴定3~8代的HGFs, 采用细胞划痕实验检测不同浓度LPS对HGFs体外迁移的影响;以LPS梯度浓度干预, 采用Western blotting方法检测Hsp27在HGFs中的表达。结果 LPS浓度越高细胞迁移能力越弱;Hsp27在正常HGFs中呈弱阳性表达, 在LPS干预后的HGFs中呈强阳性表达, 且随干预浓度增高表达量有逐渐增高的趋势, 与细胞迁移能力呈反比。结论 Hsp27在LPS介导的HGFs损伤中表达量升高, 提示Hsp27在LPS介导的HGFs损伤过程中具有重要作用。

Abstract: Objective To observe the expression of heat shock protein 27 (Hsp27) in injury of human gingival fibroblasts (HGFs) induced by lipopolysaccharide (LPS) with different concentrations. Methods The 3rd-8th generations of cultured HGFs were treated with serially diluted LPS. Would healing assay was used to determine the migration of HGFs and Western blotting was used to determine the expression of Hsp27 in them. Results The migration capability of HGFs was weakened with the increase of LPS concentration. Expression of Hsp27 was negative in normal HGFs, but was positive in the LPS-intervened HGFs with a concentration-dependent manner. Conclusion Hsp27 concentration increases in LPS-induced injury of HGFs, which suggests that Hsp27 plays an important role during the occurrence of LPS-induced injury of HGFs.
