米非司酮对卵巢癌细胞系SKOV3增殖和CEACAM6表达的影响

Effect of mifepristone on proliferation and expression of CEACAM6 of human ovarian cancer cell line SKOV3

摘要:

目的  观察米非司酮对卵巢癌细胞系SKOV3增殖和癌胚抗原相关黏附分子6(CEACAM6)表达的影响。方法  不同浓度米非司酮(空白对照、1×10-7、1×10-6、1×10-5、5×10-5mol/L)干预卵巢癌细胞系SKOV3 24、48、72h;四甲基偶氮唑蓝(MTT)法检测米非司酮对SKOV3的抑制作用;逆转录聚合酶链反应(RTPCR)和蛋白印记(Western blot)检测不同浓度米非司酮干预下CEACAM6 mRNA和蛋白表达水平的差异,并进行半定量分析。结果  与空白对照组比较,1×10-7和1×10-6mol/L米非司酮对SKOV3的生长没有明显的抑制作用(P>0.05);5×10-5mol/L米非司酮显著抑制SKOV3的生长(P<0.01),并降调SKOV3细胞中CEACAM6 mRNA和蛋白的表达水平(P<0.05)。结论  米非司酮可明显抑制SKOV3的体外增殖,其作用机制可能与抑制CEACAM6的表达有关。

Abstract:

Objective  To evaluate the effect of mifepristone on proliferation and expression of CEACAM6 of human ovarian cancer cell line SKOV3 in vitro. Methods  Human ovarian cancer cell line SKOV3 was cultured with different concentrations (control group, 1×10-7, 1×10-6, 1×10-5, 5×10-5mol/L) of mifepristone(MIF) in vitro; MTT was used to examine the cell growth; and the semi-quantity of CEACAM6 mRNA and protein were detected by RT-PCR and western blot. Results  Proliferation of SKOV3 cultured with 1×10-7, 1×10-6 mol/L of MIF was not significantly lower than the control group(P>0.05);5×10-5mol/L of MIF was able to significantly suppress the proliferation of SKOV3(P<0.01); Expressions of CEACAM6 mRNA and protein of SKOV3 in the control group were higher than that of 5×10-5mol/L of the mifepristone group(P<0.05). Conclusion  Mifepristone could significantly suppress the proliferation of human ovarian cancer cell line SKOV3 in vitro. The action mechanisms may be associated with the suppression of expression of CEACAM6.
