慢性肾功能衰竭对大鼠阴茎海绵体缝隙连接蛋白43表达的影响

Effect of chronic renal failure on connexin 43 expression in the penis of rats

摘要: 目的:探讨慢性肾功能衰竭(chronic renal failure, CRF)对大鼠阴茎海绵体缝隙连接蛋白43表达的影响。方法:应用SD雄性大鼠36只,同期行5/6肾脏切除术,建立CRF动物模型。随机分为假手术组(NCRF组,n=6)、CRF模型组(CRF组,n=30),分别于12周注射阿朴吗啡(APO)80?μg/(kg·d)后观察大鼠阴茎勃起情况,应用Western blot方法检测大鼠阴茎海绵体缝隙连接蛋白43(connexin 43,CX43)表达。结果:NCRF组死亡1例,CRF模型组死亡5例。CRF组阴茎勃起率为28%(7/25), NCRF组阴茎勃起率100%(5/5),两组比较差异有统计学意义(P<0.05)。CRF组阴茎勃起次数为(1.0±0.0)次, NCRF组阴茎勃起次数(2.2±0.8)次,两组比较差异有统计学意义(P<0.05)。CRF组大鼠阴茎海绵体组织CX43(0.18±0.08)低于NCRF组(0.53±0.27),两组比较差异有统计学意义 (P<0.01) 。结论:CRF明显降低大鼠阴茎勃起功能。CRF降低大鼠阴茎勃起功能与大鼠阴茎海绵体CX43表达减少密切相关。

Abstract: Objective: To investigate the effect of chronic renal failure (CRF) on connexin 43 expression in the penis of rats. Methods: CRF models were established by a resection of 5/6 kidney from male sprague-dawley (SD) rats in one stage. Rats of the control group (NCRF group, n=6) and the experimental group (CRF group, n=30) were subjected to a low dose (80μg/kg·d) of apomorphine (APO) at the 12th week after resection. The correspondent penile erection was determined. Expression of connexin 43(cx43) in the rat corpus cavernosum was also determined by the Western blot method. Results: One rat died in the NCRF group and five in the CRF group. The penile erection ratio of the CRF group was 7/25 and of the NCRF group was 5/5, which presented a significant difference(P<0.05). The average penile erection frequency of the CRF group was(1.0±0.0)and of the NCRF group was(2.2±0.8), which was also significantly different (P<0.05). Expression of cx43 in the rat corpus cavernosum of the CRF group was lower than that of the NCRF group [(0.21±0.07 ) vs(0.40±0.08)] (P<0.01). Conclusion: CRF significantly reduces the erectile function of the rats, CRF-related ED bears a correlation with the decrease in expression of cx43 in the rat corpus cavernosum.
