创伤性应激对大鼠海马BDNF表达的影响

Effect of post-traumatic stress on hippocampal BDNF expression in rats

摘要: 目的 探讨精神创伤后应激大鼠海马组织中脑源性神经营养因子(BDNF)的水平变化及其意义。 方法 将Wistar大鼠随机分为正常组与应激组(应激造模后1 d组、7 d组、14 d组),每组8只。应激组建立SPS&S模型,采用旷场实验、Morris水迷宫等方法检验各组大鼠造模前后的行为学指标变化并评估造模效果,采用酶联免疫吸附法(ELISA)测定各组大鼠海马组织中 BDNF蛋白的水平变化。 结果 各组两两比较,正常组海马组织中BDNF水平高于14 d组,差异有统计学意义(Z=-3.363,P=0.001),与1 d组和7 d组无统计学差异;应激各组之间两两比较无统计学差异。 结论 正常组海马BDNF含量高于应激组(14 d组),应激后BDNF的水平有所下降,但其变化过程是缓慢的,BDNF可能成为精神创伤后应激的生物学标志之一。

Abstract: Objective To investigate changes of brain-derived neurotrophic factor(BDNF)in the hippocampus of rats suffering from post-traumatic stress, and to explore the significance of such changes. Methods A total of 32 Wistar rats were randomly divided into control group and post-traumatic stress group, which was subdivided into 3 groups:1 d group(1 day after modeling, n=8), 7 d group(7 days after modeling, n=8), and 14 d group(14 days after modeling, n=8). Rats in the stress groups were evaluated with open-field test and Morris water maze, and changes in the ethological indexes before and after modeling were assessed, followed by determination of BDNF level in hippocampus of the each group of rats by ELISA. Results BDNF level of the 14 d group was significantly lower than that of the control(Z=-3.363, P=0.001), but there was no difference between the 1 d group and control, 7 d group and control, and 1 d group and 7 d group. Conclusion BDNF level decreases in rats suffering from post-traumatic stress, but such change is rather slow. BDNF is a potential marker for post-traumatic stress.
