血管性痴呆大鼠小脑顶核形态学变化及活化型Caspase-3、PARP-1的表达

Morphological changes and expressions of cleaved Caspase-3, PARP-1 in cerebellar fastigial nucleus of rats with vascular dementia

摘要: 目的 观察血管性痴呆(VD)大鼠小脑顶核形态学变化及活化型Caspase-3、PARP-1的表达。方法 选用健康SD大鼠32只,随机分为假手术组(8只)及VD组(24只)。采用Morris水迷宫,在假手术组术后1周,VD组术后1、2、4周时对大鼠做行为学观察和测试,通过尼氏染色观察各组大鼠小脑顶核形态学的变化,用免疫组化法检测小脑顶核活化型Caspase-3、PARP-1的表达。结果 VD组大鼠各时间点水迷宫逃避潜伏期较假手术组均明显延长(P<0.01);尼氏染色可见VD组大鼠小脑顶核神经元细胞数较假手术组减少(P<0.01);VD组大鼠小脑顶核活化型Caspase-3、PARP-1的表达与假手术组比较明显增加(P<0.01)。结论 VD大鼠小脑顶核发生了损伤,活化型Caspase-3、PARP-1在此过程中发挥重要作用。

Abstract: Objective To observe the morphological changes and expressions of cleaved Caspase-3 and PARP-1 in the cerebellar fastigial nucleus of rats with vascular dementia (VD). Methods Thirty-two healthy Sprague-Dawley (SD) rats were randomly divided into two groups, sham group(n=8) and VD group(n=24). Morris water maze test was performed to evaluate the behavior of rats in sham group at week 1 and VD group at three time points(week 1, 2 and 4) after injury. The morphological changes of cerebellar fastigial nucleus were examined by Nissl staining. The expressions of cleaved caspase-3 and PARP-1 in the cerebellar fastigial nucleus were detected by immunohistochemical method. Results Compared with sham group, water maze escape latencies of VD group at three time points increased significantly (all P<0.01). Compared with sham group, the number of neurons in the cerebellar fastigial nucleus significantly decreased in VD groups (P<0.01). Compared with sham group, the expressions of cleaved Caspase-3 and PARP-1 in VD group significantly increased in the cerebellar fastigial nucleus (P<0.01). Conclusion The cerebellar fastigial nucleus is damaged in VD rats, while the cleaved caspase-3 and PARP-1 play an important role in this process.
