人类脐带间充质干细胞抑制肝内胆管癌细胞生长的机制

Mechanism of human umbilical cord mesenchymal stem cells suppressing cholangiocarcinoma cell growth

摘要: 目的 探讨人类脐带间充质干细胞(hUC-MSCs)对肝内胆管癌细胞生长的影响,深入了解其分子机制。方法 采用hUC-MSCs条件培养基作用于肝内胆管癌HCCC-9810细胞,通过Transwell 共培养实验、MTT比色实验、DNA断裂实验等方法检测细胞增殖和细胞凋亡,通过Western blotting及免疫荧光染色方法检测相关凋亡通路中蛋白质表达水平。结果 细胞增殖实验证实,间充质干细胞条件培养基对HCCC-9810细胞的增殖抑制表现出时间和剂量依赖性,且抑制作用是通过诱导细胞凋亡介导,增殖抑制率由12.87%升至50.98%,凋亡率由10.1%升至49.67%。免疫印迹实验结果显示,p-AKT、p-GSK-3β表达被下调。结论 人类脐带间充质干细胞条件培养基可抑制肝内胆管癌细胞的生长,其分子机制与抑制PI3K/Akt信号通路有关。

Abstract: Objective To explore the effect of human umbilical cord mesenchymal stem cells on cholangiocarcinoma HCCC-9810 cells and its molecular mechanism. Methods HCCC-9810 cells were treated with conditioned media from umbilical cord mesenchymal stem cells. Cell proliferation and apoptosis were detected with co-culture systems, MTT assay and DNA fragmentation assay; the protein expressions were determined with Western blotting and immunoflorescence staining. Results Conditioned media from hUC-MSCs could inhibit proliferation and induce apoptosis of tumor cells in a dose- and time-dependent manner. The proliferation inhibition rate increased from 12.87% to 50.98%, whereas the apoptosis rate increased from 10.1% to 49.67%. Immunoblot analysis showed that the expressions of p-Akt and p-GSK-3β were down-regulated. Conclusion The hUC-MSCs conditioned media may suppress the growth of HCCC-9810 human cholangiocarcinoma cell line via PI3K/Akt pathway.
