    Effect of ectopic expressed Neurotrophic Peptide on proliferation of human prostate cancer cells

    Objective: Based on neurotrophic domain sequence (NP) of saposin C, eukaryotic expression vectors containing NP sequence and NP fused to TAT transduction domain were respectively constructed. The TAT transmembrane signal was expected to help NP go through cell membranes and have effects on prostate cancer cells. Methods: The eukaryotic expression vectors pcDNA-TAT-NP and pcDNA-NP were constructed by using gene cloning technology. After transient transfection of pcDNA-TAT-NP and pcDNA-NP into the prostate cancer cells PC3 and LNCaP, RT-PCR, MTT and flow cytometry were used to respectively detect the expression of TAT-NP and NP in the level of mRNA, prostate cancer cell growth and the cell cycle changes. Results: The eukaryotic expression vectors pcDNA-TAT-NP and pcDNA-NP were constructed and confirmed by sequencing. The result obtained from RT-PCR indicated both pcDNA-TAT-NP and pcDNA-NP were expressed in prostate cancer cells. MTT analysis showed that expressions of TAT-NP and NP had a stimulating effect on prostate cancer cell proliferation, and promoted the cells to enter S and G2/M phases. Conclusion: Ectopic expression of NP could enhance the prostate cancer cell growth and affect the cell cycle.

    Objective: To study the relationship among serum apolipoprotein A-IV (apoA-IV) concentration, obesity and leptin level. Methods: Six hundred and seventeen people from Jinan urban areas and Zibo rural areas were enrolled in this study. After the donor subjects were deprived of food for 24h, 5ml blood was drawn from the donors and was kept for 30h, and then was subjected to 3000r/min centrifugalization for 15 minutes. Then the blood serum was separated, immediately filled separately into eppendorf tubes and kept under the condition of -70℃. The concentration of leptin and apoA-IV was determined. The body weight, BMI, BF%, WHR and several other indexes were measured. Results: ApoA-IV levels were significantly and negatively correlated with body fat concentration, but were not correlated with body fat distribution. Serum apoA-IV levels were not correlated with serum leptin levels. Conclusion: Obese humans have lower serum apoA-IV levels, which are not mainly influenced by leptin.

    Objective: To investigate the antivirus spectra of singlet compound named JN219 extracted from certain strains of fungi. Methods: Fifteen different strains of viruses, including herpesviridae virus(HCMV, HSV-1, HSV-2), adenoviridae virus(Ad3), paramyxoviridae virus(RSV), reoviridae virus(RV SA11), orthomyxoviridae(influenza A, influenza B), picornaviridae(COX B1-6)and rhabdoviridae(VSV)were used for in-vitro antivirus experiments, and the antiviral activity and the cytotoxicity of JN219 were evaluated by the cytopathic effect(CPE) method. Infected cells were dyed with 1% neutral red, and the decolored and the absorptance A were read by an enzyme label meter. The cell survival rate, the median toxic concentration, the median inhibiting concentrations and therapeutic indexes were calculated by the Reed-Muench method. Antivirus stability experiments of JN219 were carried out at the same time. Results: Cytopathic effects caused by HCMV, VSV and RV SA11 were significantly inhibited. The median inhibiting concentrations (IC50) were 1.42, 0.96 and 1.22μg/ml, respectively, and the therapeutic indexes (TI) were 35.26, 52.30 and 40.82, respectively. HSV-1, HSV-2, RSV, influenza A and influenza B were partially inhibited, however COX B1-6 and Ad3 were not inhibited. The cytotoxicity of the samples appeared only in the first dilution and the antiviral activity was unstable. Conclusion: JN219 has a wide antivirus spectra.
    Effect of chronic stress on the behavior and estradiol level in different age rats

    Objective: To study the effect of chronic stress on the behavior and the estradiol level in different age rats. Methods: Thirty-six two-month-old and fifteen-month-old female Wistar rats were used in the experiments. They were randomly divided into four groups: young normal control(n=8), young stress(n=10), aged normal control(n=8) and aged stress(n=10), respectively. The stress groups were exposed to a chronic unpredictable stress for 21 days, and then they were raised normally for one week. Their behavior was determined with the method of Open field before and right after stress and one week after stress respectively. A blood sample was obtained from the vena caudalis and the serum level of estradiol was detected by radio-immunoassay.Results: Compared with the control groups, the behavioral score and the serum level of estradiol were significantly lower in the stress groups after stress(P<0.05), and they decreased more remarkably in the aged group than in the young group(P<0.01), which remained at one week after stress(P<0.05). Conclusion: Damage induced by chronic stress is different in young and aged rats, and the damages to the behavior and reproductive endocrine system are more serious in the aged rats than in the young, indicating that aging is one important influencial factor in physical stress reactivity.
    Relation among single nucleotide polymorphism in estrogen metablolizing genes CYP17, COMT and endometrial adenocarcinoma risk

    Objective: To study the relationship among estrogen metablizing genes CYP17, COMT genetic polymorphism and endometrial adenocarcinoma. Methods:CYP17 and COMT polymorphisms were detected by the methods of PCR-RFLP in a case-control study, including 80 cases of endometrial adenocarcinoma and 84 cases as controls. Results:The freque ncies of the MspAI1 polymorphisms in the cancer group and the control group were significantly different (P<0.05), and the distribution values of A1/A1 and A1/A2 genotypes in the cancer group were significantly higher than those in the control group while the frequency of A1 allele in the cancer group was also significantly higher than that in the control group. The odds ratio of A/A1 and A1/A2 genotypes was 3.152 and 2.693, respectively in endometrial adenocarcinoma, which both had a significant difference from that of A2/A2 genotype (P<0.05). There was not a significant difference in the distribution of COMT genotypes between endometrial adenocarcinoma patients and control subjects (P>0.05). Conclusions: The results suggest that A1/A1, A1/A2 genotypes and A1 allele are related to the susceptibility of endometrial adenocarcinoma and they might be one of the useful markers for predicting endometrial adenocarcinoma. The allele encoding for low activity COMT may not be a genetic risk factor for endometrial adenocarcinoma.
    Progesterone combined with octreotide inhibits the growth of human non-small cell lung cancer A549 cells

    Objective: To investigate the regulation of somatostatin recepter (SSTR) subtype mRNAs expression of non-small cell lung cancer cell line A549 by progesterone and the antiproliferation of progesterone combined with octreotide on A549 cells. Methods: The progesterone receptor was detected by the immunohistochemical method, and SSTR subtype mRNAs were determined by the reverse transcription polymerase chain reaction in cells pretreated or non-pretreated by progesterone. The cell viability was evaluated by MTT assay. Results: Progesterone receptor and SSTR1, SSTR2, SSTR4 and SSTR5 were revealed in A549 cells, but SSTR3 was not. Following 24-hour treatment by 10nmol/L progesterone, SSTR3 was detected in A549 cells. Up to 48 hours, SSTR3 was still up-regulated(P=0.012). Progesterone combined with octreotide was superior to progesterone or octreotide alone in inhibiting the growth of A549 cells(P<0.05). Conclusion: Progesterone up-regulates the expression of mRNA for SSTR subtypes in A549 cells. Progesterone enhances the ability of octreotide to inhibit the growth of A549 cells.

    Objective: To establish a fluorogenic quantitative RT-PCR method for detecting the expression of OATP-E gene in tumor cells, and to investigate the expression of organic anion transporting polypeptide-E (OATP-E) gene in breast cancer cell lines MCF-7 and MCF-7/ADR. Method: Fluorogenic quantitative RT-PCR (FQ- RT- PCR) was set up to examine the specific expression of OATP-E gene in MCF-7 and MCF-7/ADR cell lines. Results: After 10 duplicate tests with the FQ-RT-PCR method, the average level of the OATP-E mRNA was 5.47×105copies/μg RNA in MCF-7 cell line and 2.82×104copies/μg RNA in MCF-7/ADR cell line, respectively, therefore the former was 19.4 times greater than the latter. Conclusions: Using FQ-RT-PCR, we can get an accurate quantitative result of OATP-E gene expression . Moreover, OATP-E gene expression in the drug resistant breast cancer cell line MCF-7/ADR is at a lower level than that in cell line MCF-7.
    Role of 1-Hexylcarbamoyl-5-Fluorouracil on the Th1/Th2 balance in patients with ankylosing spondylitis

    Objective: To investigate the influence of sulfalazine (SSZ) and 1-Hexylcarbamoyl-5-Fluorouracil (HCFU) on T helper cytokines in patients with ankylosing spodylitis (AS). Methods: Nineteen AS patients were randomized treated with SSZ or HCFU. The RT-PCR semi-quantitative method was employed to determine the mRNA expression of Th1/Th2 cytokines (IFN-γ, IL-2, IL-4, IL-6 and TNF-α). The changes of Th1/Th2 subgroups modulated by SSZ and HCFU was studied. Results: HCFU administration was followed by a decrease expression of IFN-γ mRNA and TNF-α mRNA (33.33% vs 89.74%, 44.44% vs 94.74%, P<0.05), whereas other cytokines (IL-2,IL-4, IL-6) produced no significant changes(P>0.05). SSZ administration was followed by an increased expression of IL-4 mRNA ( 80% vs 21.05%, P<0.05) and a decrease expression of IFN-γ mRNA (P<0.05), whereas other cytokines (IL-2, IL-6, and TNF-α) produced no significant changes(P>0.05). Conclusion: HCFU and SSZ therapy for AS patients results in a shift from Th1 to Th2 subsets. HCFU can decrease the mRNA levels of TNF-α.
    Role of glucagon-like peptide-1 on proinsulin mRNA levels in diabetic mouse

    Objective: To investigate the role of intravenous glucagon-like peptide-1 on blood glucose and on pancreas's proinsulin mRNA level in diabetic mice. Methods: Alloxan (45mg/kg) was infused by a tail vein in male Kunming mice to successfully make the diabetes model. Twenty diabetic mice were divided at random into a control group (normal sodium) and an experimental group[GLP-1(7-36)NH2]. Tail blood glucose was measured respectively before infusion of normal sodium[GLP-1(7-36)NH2] and at 5 and 10 minutes after infusion. Proinsulin mRNA levels were assessed by the semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR). Results: Blood glucose was not significantly different before intravenation between the control and the experiment groups(P>0.05); blood glucose concentrations at 5 and 10 minutes after intravenation were significantly decreased in the experiment group compared with the control group(P<0.05). The levels of proinsulin mRNA were significantly higher in the experiment group compared with the control group (P<0.05). Conclusion: Intravenous injection of GLP-1(7-36)NH2 can promote proinsulin gene transcripts, increase pancreas′s proinsulin mRNA level, stimulate insulin biosynthesis and secretion and quickly decrease blood glucose.
    Effects of alveolar defect repair by Perioglas and HPPA on the orthodontic tooth movement: an experimental study

    Objective: To observe the movement of rats′ molars at the area of defective alveolar bone filled with Perioglas and Hydroxyl poly calcium sodium phosphate (HPPA). Methods: Fortyfour female Wistar rats with alveolar bone defect were randomly divided into two groups: Perioglas and HPPA groups. Orthodontic appliances were fixed to draw the maxillary first molar after 12 weeks. Distances of maxillary first molar movement were measured after 20 weeks. Results: Movement distances of the first molar were not significantly different between the Perioglas group and the normal contralateral group or between the HPPA group and the normal contralateral group(P>0.05). Conclusion: The rats′ teeth can be moved with orthodontic force at the alveolar bone filled with Perioglas and HPPA, and there were no significant differences of the movement distance of a tooth in the side filled with materials and in the contralateral side.

    Objective: To investigate the effects of estrogen on the expression of KLK6 mRNA and protein in the human ovarian cancer cell line HO8910. Methods: HO8910 cells were incubated with 17-βE2 at different concentration (1×10-7、1×10-8、1×10-9 and 1×10-10mol/L) for 72 hours. Then the expression levels of KLK6 mRNA and protein were measured by real-time fluorensence quantitive RT-PCR and flow cytometry, respectively. Cell proliferation was measured by 3- (4, 5-dimethylthiazol-z-yl)-2, 5-dipheny tetrazolium blue (MTT) colorimetric assay, and the cell cycle was detected by flow cytometry. Results: Compared with the ethanol controls, the expression of KLK6 mRNA and protein in the HO8910 cell line accompanied by different concentrations of 17-βE2 increased (P<0.01 ) and the cell proliferation increased significantly, also the cell percentage of the G0/G1 phase decreased and that of the S phase and the G2/M phase increased. Conclusion: Estrogen up-regulates the expression of KLK6 mRNA and protein and stimulates HO8910 cell proliferation.
    Effect of fenofibrate on myocardial remodeling and NF-κB expression in spontaneous hypertensive rats

    Objective: To investigate the effect of fenofibrate on nuclear factor kappa B (NF-κB) in cardiac muscle and the preventive effect of fenofibrate on myocardial remodeling. Methods: Twenty-two spontaneous hypertensive rats (SHR) were randomly divided into two groups: a spontaneous hypertensive rat (SHR) group(n=10) and a fenofibrate group(n=12). A same age Wistar-kyoto(WKY) rat group(n=10) was selected as controls. The fenofibrate group was given drugs for 8 weeks. At the beginning and ending of the experiment, echocardiographic scanning was performed in each group. At the end of the experiment, the rats were executed and the cardiac muscle was examined histologically. Real-time RT-PCR was used to detect the mRNA expression of NF-κB. Results: At the end of the experiment, compared with the WKY group,the ventricular thickness and the ratio of left ventricle weight to body weight in the SHK group were significantly increased(P<0.05). Compared with the SHR group, the above parameters in the fenofibrate group were significantly decreased(P<0.05). The expression of cardiacal NF-κB mRNA in the SHR group was significantly higher than that in the WKY group(P<0.05), but showed a down regulation in the fenofibrate group(P<0.05). By immunohistochemistry, NF-κB staining in SHR group was strong and located at fargoing cardiac muscle tissues, but was weak and located within limits in fenofibrate group. Conclusion: The expression of NF-κB increases in cardiac muscle of SHR. Fenofibrate may delay the development of myocardial remodeling by decreasing the expression of NF-κB.
    Evaluation the short-term effects of uvulopalatopharyngoplasty on the sleep apnea quality of life index

    Objective: To evaluate the significance of the sleep apnea quality of life index (SAQLI) for uvulopalatopharyngoplasty (UPPP) and quality of life in patients with obstruction sleep apnea hypopnea syndrome(OSAHS). Methods: Thirty patients with OSAHS were diagnosed by PSG and treated by UPPP. SAQLI was surveyed before and after surgery. Results: Before operation, the score of SAQLI was greatly reduced and had significant statistically correlation with the apnea hypopnea index(AHI) (r=-0.54) and lowest saturation (LSaO2) (r=0.56). After operation, the total score of SAQLI including 4 domains increased significantly (P<0.001). Quality of life was improved in 86.7% cases in general. By SAQLI, we could discriminate among those with different qualities of life. Conclusion: The amelioration of quality of life in OSAHS patients is significant in the short term after UPPP. SAQLI can be a valuable measurement in clinical diagnosis and follow-up treatment.
    Effect of nitric oxide and substance P in the pathological mechanism of allergic rhinitis

    Objective: To explore the role of nitric oxide(NO) and substance P(SP) in the pathological mechanism of allergic rhinitis. Methods: The concentration of serum NO in 40 patients with allergic rhinitis and 30 healthy controls was detected by the nitrate reductase method. The expressions of inducible nitric oxide synthase (iNOS) and SP in the inferior nasal concha mucosa were detected by the strept avidin-biotin complex (SABC) immunohistochemistry method. Results: The level of serum NO in patients with allergic rhinitis was significantly higher than that in the controls (52.42±18.73μmol/L vs 28.67±10.35μmol/L, t=6.26, P<0.01). A large number of iNOS positive cells, mainly expressed in mucosa epithelial, submucos glandular, endothelial and submucos inflammatory cells were found in the concha mucosa in allergic rhinitis patients; but few iNOS positive cells could be found in the controls (27.5±3.2 vs 4.3±1.7, per scope, t=36.03, P<0.01). The number of SP positive cells was 23.5±4.6 in allergic rhinitis patients and 5.3±3.1 in the controls(t=18.7,P<0.01). There were positive correlations between the number of iNOS positive cells and of SP positive cells (r=0.69, P<0.01). Conclusion: The results suggest that NO and SP may play an important role in the pathogenesis of allergic rhinitis. SP can promote the production of NO by iNOS, which induces the process of allergic inflammation.
    Changes of transforming growth factor-β of human detrusor after bladder outlet obstruction induced by benign prostatic hyperplasia

    Objective: To investigate the changes of human detrusor transforming growth factor -β(TGF-β) after bladder outlet obstruction (BOO) induced by benign prostatic hyperplasia (BPH). Methods: Obstructive detrusor stability was found in 33 patients and obstructive detrusor overactivity in 21 patients; and bladder tumor in 15 men (control) who were operated on in the same period. TGF-β was measured by immunohistochemical staining. Results: The TGF-βlevels of the obstructive groups were much higher than those in the control group. There were significant differences among the three groups (P<0.01). Conclusion: The TGF-β level is significantly higher after BOO, which might be correlated with denervation supersensitivity and detrusor overactivity.

    Objective: To investigate the potential toxicity of Euphorbia ebiacteolata Hayata (EEH) water extract. Methods: The study was performed on inbred Kunming mice, using the traditional toxicological method based on observing ratios of micronucleus and sperm abnormity (RMS). The mice were randomly divided into 5 groups: a negative control with distilled water, a positive control with cytoxan(50mg·kg-1), and low dose, medium, and high dose of EEH extract(77g·kg-1, 144g·kg-1 and 288g·kg-1 respectively). Results: The results showed that the high and medium doses significantly increased RMS compared with the negative controls (P<0.01, P<0.005), but a similar result was not observed in the low dose group (P>0.05). Conclusion: EEH can induce mutation and sperm abnormity in medium and high doses, whereas it cannot induce significant toxicity in low doses.

    Objective: To evaluate the correlation between the mucosal epithelial chemokine CCL28 and sIgA level in the saliva of healthy humans. Methods: Unstimulated whole saliva samples were collected. The CCL28 level was determined by ELISA.The salivary sIgA was measured by rate nephelometry assay. Results: In saliva samples of 42 healthy human beings, the mean value of salivary CCL28 was 70.70±40.91ng/ml, and of salivary sIgA was 113.86±54.90mg/L. A positive linear correlation was noted between CCL28 and sIgA in saliva (r=0.540). Conclusion: The mucosal epithelial chemokine CCL28 can promote the secretion of sIgA in saliva.
    Role of the Chinese drug saffron on the expression of leukemia inhibitory factor in the endometrium of early pregnant mice

    Objective: To explore the relationship between the saffron and the expression of leukemia inhibitory factor(LIF) in the endometrium of early pregnant mice. Methods: The mated female mice were orally treated with saffron solution in a dose of 10.5g/kg, 15.5g/kg and 20.5g/kg or physiological saline (control), on the 2nd, 3rd and 4th days after fertilization. The uteri of the mice were removed and routinely sectioned 24 hours later. The morph of the endometrium was observed with HE and the expression of LIF in the endometrium of the mice was analyzed with immunohistochemistry and Quantimet. Results: The endometrium was in congestion and disorder in the experimental groups; with increasing doses of saffron, the endometrium gradually changed. The LIF was expressed strongly in the murine endometrium of control group. The expression level of LIF in experimental groups was much lower than that in the control group(P<0.05, P<0.01)and decreased gradually with increasing doses of saffron. Conclusion: Saffron can inhibit the expression of LIF in the endometrium of pregnant mice, and therefore interferes with the implantation of embryos.

    Objective: To explore the association of cytochrome P450 1A1(CYP1A1 ) gene polymorphism and frequent risk factors of gastric cancer in Chinese Han people. Methods: One hundred and two patients with gastric cancer(GC), one hundred and ten with chronic atrophic gastritis(CAG), one hundred and three with chronic superficial gastritis(CSG), sixty-two with gastric ulcer(GC), sixty-two with duodenal ulcer (DU) and sixty-two controls were enrolled in this study. Data on life-style had been routinely collected by means of common questionnaire sheets. CYP1A1 genotypes were detected by the polymerase chain reaction of sequence-specific primers (PCR-SSP). Hp infection was detected by an enzyme-linked immunosorbent assay (ELISA). The approach of case control was used in the analyses of risk factors. Results: Risks of gastric cancer was based on age, sex, education, occupation, helicobacter pylori infection, tobacco smoke, and CYP1A1 G/G genotype. Garlic had a protective effect on gastric cancer. Multivariate analyses indicated that risks of gastric cancer: helicobacter pylori infection, CYP1A1 G/G genotype, and garlic had a protective response. There was synergistic action between helicobacter pylori infection and CYP1A1 G/G genotype, tobacco smoke and CYP1A1 G/G genotype in the development of gastric cancer. Conclusion: An individual with a CYP1A1 G/G genotype is at risk of gastric cancer. This genotype may be considered as a marker related to the susceptibility to gastric cancer. It may provide a clue to define the risk of gastric cancer and lead to diagnosis and treatment at an early stage.

    Objective: To evaluate the correlation between MiniMed continuous glucose monitoring system blood glucose (CGMSBG) and self monitoring blood glucose (SMBG) both in a sedentary group and in an exercise group, and study the accuracy of CGMS after exercise. Methods: Thirteen type 2 diabetic patients admitted to hospital were submitted to CGMS. The correlation and coincidence between CGMSBG values and SMBG values before and after exercise were assessed with the Person correlation analysis. Results: The correlation coefficient r of 223 CGMSBG values and paired sedentary SMBG values was 0.939 (P<0.01). When the CGMSBG value was below 4.4mmol/L, the correlation coefficient r was 0.524(P<0.05). When it was between 4.4 and 10mmol/L, the coefficient r was 0.811(P<0.01) . The correlation coefficient r of the CGMSBG and SMBG values after exercise was 0.960(P<0.01) and was clinically favorable with the median of -0.1mmol/L. Conclusion: The correlation and coincidence between CGMSBG values and SMBG values are favorable. The CGMSBG value and the SMBG value mostly fit when they are at the middle level, but the CGMSBG value is a bit higher than the SMBG value when at a low level and is a little lower than the SMBG value when at a high level. The CGMSBG value is higher than the SMBG value after acute exercise, but they still have good concordance.

    Objective: To investigate and compare the response of laser-assisted subepithelial keratectomy (LASEK) and laser in situ keratomileusis(LASIK) for moderate and high rule-shift astigmatism. Methods: One hundred and thirty-seven patients (206 eyes) with corneal myopic astigmatism in with rule-shift were randomly divided into LASEK and LASIK groups. All were followed up for 6 to 12months after laser ablation. At 6months after operation, the changes of corneal astigmatism and astigmatic axis were studied. Results: Six months after operation, the postoperative corneal astigmatism was (0.215±0.130)D in LASEK group and (0.572±0.195)D in LASIK group, which was reduced by (3.306±0.817)D and(2.996±0.497)D, respectively. There was a significant difference between them, while the corneal astigmastic axis was not significantly changed. Conclusion: Compared to LASIK, LASEK is a better method for improvement of moderate and high rule-shift astigmatism.

    Objective: To research the effect and the prognostic assessment of VEGF in malignant pleural effusion, as well as the usefulness of VEGF in differential diagnosis between benign and malignant effusion. Methods: Using the sandwich enzyme-linked immunosorbent assay(ELISA), we detected the level of VEGF, carcinoembryonic antigen(CEA)and adenosine deaminase(ADA)in effusion and plasma respectively in pulmonary carcinoma(n=32), tuberculous(n=34) and transudative(n=9). Results: The level of VEGF in effusion and plasma in patients with pulmonary carcinoma(345±167ng/l and 120±79ng/l)was significantly higher than that in patients with tuberculous(119±52ng/l and 53±20ng/l)and transudative(57±22ng/l and 35±12ng/l)(P<0.05); the level of VEGF in tuberculous was higher than that in transudative(P<0.05); the level of VEGF in pulmonary carcinoma patients with malignant effusion whose survival time was shorter than six months(386±185ng/l)was evidently higher than that in those who survived longer than twelve months(201±88ng/l)(P<0.05).VEGF level was positively correlated with CEA and LDH level(r=0.51,r=0.46, P<0.01)and not correlated with ADA level, glucose, protein content, the total number of WBC and classified count(P>0.05).The VEGF level in pleural effusion provided a diagnostic test for malignancy with a sensitivity of 75.6% and a specificity of 73.5%(cut-off point 221ng/L). Conclusion: VEGF plays an important role in the malignant pleural effusion accumulation and is related to prognosis. The measurement of VEGF is helpful in discriminating between benign and malignant effusion.

    Objective: To explore the role of VEGF and IL-6 in the aqueous humor in the pathogenesis of diabetic retinopathy (DR) . Methods: Aqueous humor samples were collected from diabetic patients with cataracts or without retinopathy,and aqueous humor samples from non-diabetic patients with cataracts were chosen as controls. The content of VEGF in the aqueous humor was determined by using the ELISA technique. Results: The level of VEGF in the aqueous humor samples of diabetic patients without retinopathy(NDR), background diabetic retinopathy (BDR) and proliferative diabetic retinopathy (PDR) were(220.27±25.69)pg/ml, (284.31±58.92)pg/ml and (459.07±90.90)pg/ml. The level of IL-6 in the aqueous humor from NDR, BDR and PDR were(154.09±32.76)pg/ml,( 221.81±60.52 )pg/ml and (387.95±90.22)pg/ml. The levels of VEGF and IL-6 in the aqueous humour from the control group were(135.50±25.69)pg/ml,( 78.71±20.37)pg/ml. Compared with the control, the level of VEGF and IL6 in the aqueous humor of diabetic patients increased significantly (P<0.01). The level of VEGF and IL-6 in the aqueous of the NDR, BDR and PDR also increased significantly (P<0.01). The level of VEGF significantly correlated with the level of IL-6 in the aqueous humor(r=0.995, P<0.01). The level of VEGF and IL-6 in the aqueous humor also significantly correlated with the severity and prolongation of DR(r=0.869, P<0.01; r=0.865, P<0.01). Conclusion: VEGF and IL-6 play active roles in the generation and development of DR, and the level of VEGF in the aqueous humor closely correlates with the level of IL-6 in the aqueous humour of diabetic patients.

    Objective:To study the loss of heterozygosity(LOH) at FHIT, SLIT2 and EDNRB gene at microsatellite markers D3S1287, D4S1593, and D13S160 in endometrial carcinoma. Methods: Tumor tissues and relatively normal tissues of thirty-five cases of endometrial carcinoma were examined by using PCR-denaturing PAGE-silver staining at microsatellite markers D3S1287, D4S1593, and D13S160. Results: The total LOH rate was 54.3%, and LOH rate of D3S1287, D4S1593, and D13S160 was 34.5%, 20.5%, and 19.3%, respectively. The LOH rate at FHIT, SLIT2, and EDNRB gene did not positively correlate with the stage of clinical-pathogensis in endometrial carcinoma. Conclusion: LOH exists at micro-satellite markers of FHIT, SLIT2, and EDNRB gene in endometrial carcinoma, suggesting that FHIT, SLIT2, and EDNRB gene are tumor suppressor genes(TSGs) and the inactivity of TSGs maybe one of the reasons for carcinoma.

    Objective: To investigate the relationship between the transcriptional expression of RASSF1A(Ras association domain family 1A gene) and oncogenesis and the development of endometrial carcinoma. Method: RT-PCR was used to detect the expression of RASSF1A mRNA in 50 cases of endometrial carcinoma tissues and 20 cases of normal endometrial tissues. Results: ① RASSF1A mRNA was identified in all normal endometrial tissues, but not found in 54% of endometrial carcinoma tissues. ② The loss rate of RASSF1A mRNA was related with the surgical-pathological stage, lymphatic metastasis, myometrial infiltration depth and pathological subtype (P<0.05), but was not related to patients' age or pathological grade (P>0.05). Conclusion: Loss or abnormal down-regulation of RASSF1A mRNA is a frequent event in endometrial carcinogenesis, which may play an important role in the malignant progression and prognosis of endometrial carcinoma.

    Objective: To evaluate the health-related quality of life in patients with obstructive sleep apnea hypopnea syndrome and the influencing factors. Methods: The Calgary questionnaire for health-related quality of life of patients with sleep apnea and the Epworth sleepiness scale(ESS) were used to evaluate the quality of life and excessive daytime sleepiness(EDS) in OSAHS patients and simple snoring patients. The Pearson correlation and stepwise multiple regression analysis were used to analyze the correlation between the SAQLI and body mass index (BMI), neck collar, abdomen circumference, sleep architecture, respiratory disturbances and EDS. Results: There was a difference in each aspect between OSAHS patients and simple snoring patients. The SAQLI was significantly correlated with EDS. Conclusions: The health-related quality of life in patients with OSAHS is reduced. EDS is an important influencing factor for the health-related quality of life in patients with OSAHS.
    Magnetic resonance angiography with super high field in the diagnosis of carotid artery diseases

    Objective: To assess the value of 3D CEMRA in the diagnosis of arterial diseases in the neck. Methods: 3D CEMRA was performed in 130 patients who were suspected to have arterial diseases in the neck. Some patients also took TOF MRA, PC MRA and DSA. The number of lesions detected by different methods was calculated and compared. The patients were randomly divided into 3 groups based on the different contrast injection rate: 1ml/s, 2ml/s and 3ml/s. The imags of each group were graded and compared. Results: Of the cases, 18 (13.8%) did not show any abnormal arteries and 112 (86.2%) showed 288 abnormal segments of the arteries. Among all the lesions, atherosclerosing plaque accounted for 34.0%(n=98); arterial stenosis, 26.0%(n=75); arterial occlusion, 6.9%(n=20); tenuous arteries, 15.3%(n=44); tortuous arteries, 16.0%(n=46); abberant arteries, 1.4%(n=4); and aneurysm, 0.3%(n=1). For the 93 cases that took both 3D CEMRA and TOF MRA, there was a significant difference between the two methods, except for the external carotid artery. For the 81 cases that took both 3D CEMRA and PC MRA, there was also a significant difference between them for every segment of arteries. For the 8 cases that took both 3D CEMRA and DSA, there was not a significant difference between them. For the three groups divided based on the different contrast injection rate, there was a statistical difference between the group of 1ml/s and 3ml/s and between the groups of 1ml/s and 2ml/s, but not a significant difference between the groups of 3?ml/s and 2?ml/s. Conclusion: 3D CEMRA is a reliable method in the diagnosis of arterial diseases in the neck and can mostly replace the gold standard of DSA.
    Half-hepatic blood flow occlusion for hepatolithiasis at normal atmospheric temperature

    Objective: To investigate an optimum method for left hepatolith to improve the operation response. Methods: Sixty-three cases of left hepatolith underwent operations of left-half hepatectomy, Roux-en-Y cholangio-jejunostomy and removal of stones by cholangiotomy under the condition of hemihepatic blood flow occlusion at normal atmospheric temperature. The postoperative complications, the index of liver function, the recurrent rate and the re-operation rate were evaluated. Results: No one died during the operations and the postoperative recovery rate, the improvement rate and the residual rate were 82.4%, 17.6% and 8.3%, respectively. Complications were found in 23 cases, biliary fistula in 2 cases, subphrenic infection in 4, incisional wound infection in 9, transaminase lifting in 6 and digestive tract bleeding in 2, but they were all cured by non-operative treatment. During 1-8 years of follow-up, 87.7% of the cases were rechecked by ultrasonic diagnosis B, CT and liver function test, and 6 of them had relapsed. Conclusion: The hemihepatic blood flow occlusion for hepatectomy is a feasible, reasonable and effective procedure due to good control of blood loss, retention of blood circulation in the healthy liver side, and lessening of intra-operation and post-operation liver dysfunction.
