Killing action of the adenovirus vector carrying HSV-TK on synovial cells of pigmented villonodular synovitis

Abstract:

Objective〓〖WTBZ〗To explore the killing effects of the adenovirus vector carrying HSV-TK on synovial cells of pigmented villonodular synovitis(PVNS) in vitro. 〖WTHZ〗Methods〓〖WTBZ〗Byadenovirus vector carrying the LacZ gene was used in the synovial cells of the PVNS transfection, and the transfection efficiency was measured by X-gal staining. The expression of HSV-TK was assayed by PCR. The cytotoxicity and bystander effect were determined by MTT. The apoptosis and necrosis cells were determined by FCM. 〖WTHZ〗Results〓〖WTBZ〗When the MOI was 120, approximate 100% cells were infected. The HSV-TK/GCV system could kill proliferative synovial cells and the bystander effect definitely existed in this killing action. Also apoptosis and necrosis cells were found by FCM. 〖WTHZ〗Conclusion〓〖WTBZ〗The HSV-TK/GCV system can kill proliferative synovial cells of PVNS and can be used to treat PVNS.
