Expressions and relationship of p53 and Nup88 in colorectal carcinoma

Abstract:

Objective    To study expressions of p53 and Nup88 and their correlation in patients with colorectal carcinoma(CRC). Methods     Expressions of p53 and Nup88 mRNA were determined by reverse transcriptive-polymerase chain reaction (RTPCR) in HCT-116p53+/+ and HCT-116p53-/- cell lines.  Protein expressions of p53 and Nup88 were detected in 48 CRC samples and 13 normal colon samples by immunohistochemistry. Results    In the HCT-116p53-/- cell line, expression of p53 mRNA was more significantly decreased than that in HCT-116p53+/+, while there was no significant difference in expression of Nup88 mRNA between the two cell lines. Positive expressions of p53 (68.75%) and Nup88 (72.92%) proteins in CRC were notably higher than those in the normal tissues (P<0.01). However, there was no correlation between p53 and Nup88 expressions (r=0.173, P>0.05). Conclusion     Over-expressions of p53 and Nup88 play a role in the development of CRC, while there is no significant correlation between them.
