Expression of S-100 and C-kit in child primary obstructive megaureter

Abstract:

Objective vTo explore expressions of S-100 and C-kit and to analyze changes of gangliocytes and interstitial cells of cajal in biomechanical properties of primary obstructive megaureter. Methods  25 specimens of the ureteral ending and dilated segments were obtained from children with POM who were referred for surgeries (aged 2 months to 10 years, mean 2.5 years). Control group:inferior segments of the ureter specimens were taken from 15 cases of renal neoplasm and the tissues were histologically confirmed as unaffected. Samples were immunostained using multiclone antibodies specific for S-100 and C-kit. Mean optical density of S-100 staining mean number of S-100 positive gangliocytes and C-kit positive interstitial cells of cajal were counted in microscopic fields by a high-fold microscope. Results  Mean optical density of S-100 staining and mean number of S-100 positive gangliocytes per microscopic field were lower in ureteral endings compared with the control(P<0.01) and the dilated group(P<0.01). The number of C-kit positive interstitial cells of cajal was also significantly lower in ureteral endings compared with the control(P<0.01) and the dilated group(P<0.01). No statistical significance was found between the dilated group and the control group. The number of gangliocytes and interstitial cells of cajal were positively correlated among the three groups. Conclusion  S-100 positive gangliocytes and C-kit positive interstitial cells of cajal play important roles and may have synergistic actions in child primary obstructive megaureter.
