The expression of spinal Reelin following chronic constriction injury of the sciatic nerve in rats

Abstract: Objective To detect the expression of Reelin in the lumbar spinal cord of a rat model of neuropathic pain induced by chronic constriction injury on sciatic never (CCI). Methods 54 male SD rats were randomly divided into sham operation group (Sham group, n=12) and CCI model group (CCI group, n=42). Paw withdrawal mechanical threshold (PWMT) and paw withdrawal thermal latency (PWTL) were tested on day 1 before operation and day 1, 4, 7, 10, 14, 17, 21 and 28 after operation. Western blot and RT-PCR were used to detect the protein and mRNA level of Reelin in rat spinal dorsal horn. Results Both PWMT and PWTL have no difference in rats between sham group and CCI group before operation. After sciatic never ligation, both PWMT and PWTL were significantly decreased(P<0.01)while sham operation have no effects on PWMT and PWTL (P>0.05). Compared with sham group, the expression of Reelin mRNA Were significantly decreased in CCI rats(P<0.05),so were the expression of Reelin protein(P<0.05). Conclusion Reelin was decreased in CCI rat spinal dorsal horn.
