Effects of resveratrol on brain inflammation and the level of Aβ1-42 in hippocampus from ovariectomized obese rats fed with a high-fat diet

Abstract:

Objective   To explore the effects of resveratrol on the neuroinflammation and the level of Aβ1-42 in hippocampus from ovariectomized obese rats fed with a highfat diet. MethodsThirty female Wistar rats aged 3 months were randomly divided into three groups: the sham operation fed with low-fat diet (SHAM) group; the ovariectomy fed with high-fat diet(OH) group; the ovariectomy plus highfat diet treated with resveratrol [40mg/(kg·d)-1] (OHR) group. After three months of administration, the rats blood was collected from arteria cruralis to detect the serum concentration of total TC, TG, LDLC, HDLC and E2; the expression of IL6 and TNFα mRNA in hippocampus was determined by realtime PCR; the levels of IL6, TNFα and Aβ142 protein in hippocampal tissues were observed by ELISA. ResultsThe level of serum E2 was significantly lower in the OH group than that in the SHAM group(P<0.01). However, rats in the OHR group had a higher level of serum E2 than rats in the OH group (P<0.05); The levels of serum TC and LDLC in the OH group were higher than those in the SHAM group (P<0.01), and rats in the OHR group had a lower level of serum TC(P<0.01); Expression of IL6, TNFα mRNA and IL6, TNFα, Aβ142 proteins in the OH group significantly increased compared with that in the SHAM group (P<0.01). Nevertheless, rats in the OHR group showed significantly lower expression of IL6, TNFα mRNA and IL6, TNFα, Aβ142 proteins rats in the OH group (P<0.05). ConclusionResveratrol can attenuate the accumulation of Aβ142 in hippocampus from ovariectomized obese rats fed with a highfat diet. The mechanism may be associated with its increasing the level of E2, decreasing the content of TC and ameliorating the obesityrelated brain inflammatory responses.
