Effects of TMPyP4-PDT on human cervical cancer cells

Abstract:

Objective   To investigate the effect of photodynamic therapy mediated by meso-5,10,15,20-Tetrakis(N-methyl4-pyridyl) porphine (TMPyP4) on human cervical cancer HeLa cells in vitro. Methods   The proliferative activity of HeLa cells was assessed by Cell Counting Kit-8 (CCK8) assay, and the modality of TMPyP4-PDT treated cells were observed with light microscope. The apoptosis was examined by Annexin-V-FITC/PI double dye kit, and the expressions of c-myc mRNA and hTERC mRNA of the cell lines were tested by RT-PCR. Results   At the certain range of the concentration, the inhibition rate and induced cell apoptosis rate mediated by TMPyP4-PDT increased with the increase of the dosage and the light energy. The necrosis and apoptosis cells were observed with light microscope. The expressions of c-myc and hTERC mRNA were decreased. Conclusion   TMPyP4-PDT has a distinctive killing effect on HeLa cells, which shows a concentration-dependent manner and an energy-dependent manner. The possible mechanism may be related with down-regulating the expressions of c-myc mRNA and hTERC mRNA.
