Inhibition of migration in ovarian cancer cell SKOV3    by gene-silence of kinesin KIF4A

Abstract:

Objective  To study the effect  of silencing expression of kinesin KIF4A on migration of ovarian cancer cell SKOV3. Methods  Construct shRNA expression vector targeting KIF4A, and then transfect the plasmid into human ovarian cancer cell SKOV3. Generate the cell lines that stably expressed KIF4A shRNA and confirm KIF4A gene-silence in cells by Western Blotting assay. Employ the Transwell assay to evaluate the impact on migration in SKOV3 when silencing KIF4A. Results  KIF4A shRNA stably expressed cell lines were generated and Transwell assay showed that the migrated cells with expressing KIF4A shRNA we threefold faster than the Negative Control cells. Conclusion  Silencing KIF4A in human ovarian cancer cell SKOV3 increases its migration capability.
