Study of combination of allicin and amphotericin B inhibitinghyphae formation of Candida albicans in vitro

Abstract:

Objective  To explore combination of allicin and amphotericin B inhibiting effect of filament formation against Candida albicans in vitro, and detect cytotoxicity variation under combination of two drugs. Methods  The effect of combination of two drugs against Candida albicans was determined by the checkerboard microdilution method, and the fractional inhibitory concentration index(FICI)was calculated. The filament formation was induced in 10% calf serum-1640.The impact of combination of allicin and amphotericin B on the rate of hyphae and filament formation of Candida albicans was determined.The H8 cytotoxicity variation under combination of two drugs was detected through cellular appearance and MTT. Results  Drugs combined together acted synergistically against Candida albicans, and allicin could enhance the ability of amphotericin B to inhibit filament formation.The effect of cytotoxicity was reduced remarkably under combination of two drugs. Conclusion  Allicin largely enhances the anti-Candida albicans activity of amphotericin B, and it is a best selection in clinical applications for reducing dose and adverse effect of amphotericin B.
