Expression and function of the FLT3 gene in patients with myelodysplastic syndromes

Abstract:

Objective  To investigate FLT3 expression and FLT3 length mutation (FLT3-LM) in patients with myelodysplastic syndromes(MDS)and their clinical implication. Methods  Mono-nucleus cells were extracted from the bone marrow of 38 patients with MDS and 10 with non-malignant blood diseases. PCR was used to examine FLT3 gene expression and FLT3-LM. Results   11 FLT3 gene expression was found in 11(28.9%) of the 38 patients with MDS. FLT3-LM were detected in 2(18.2%) of 11 the MDS patients. No FLT3-LM was found in the 10 controls(P<0.05). There were no significant differences in clinicohematological  features including age, hemoglobin level, platelet count, WBC count and PB blasts percentage between the ITD(+) and ITD(-) groups. Patients with FLT3/ITD(+) had a significantly higher percentage of BM blasts (P=0.102). Conclusion  The  FLT3 mutation represents an abnormality in MDS patients. FLT3LM could be helpful to the diagnosis, prognosis and target therapy of MDS.
