Expression and significance of cyclin D1, cyclin E and p27 in transitional cell carcinoma of the renal pelvis

Abstract:

Objective  To investigate  expression and clinical significance of cell cycle regulatory factor D1 (cyclin D1), cell cycle regulatory factor E (cyclin E) and p27 in transitional cell carcinoma of the renal pelvis. Methods  Expression levels of cyclin D1, cyclin E and p27 in surgical specimens from 63 patients were examined by immunohistochemistry. Pathological grading showed a G1 tumor in 12 cases, G2 in 40 and G3 in 11; TNM staging showed a Ta tumor in 4 cases, T1 in 9, T2 in 24, T3 in 17 and T4 in 9. Eleven  specimens obtained from normal renal pelvises served as the controls. Results  Cyclin D1 was weakly expressed and cyclin E was not expressed while p27 was strongly positive in normal pelvic tissue. In transitional cell carcinoma,  expressions of cyclin D1 and cyclin E were significantly increased (P<0.05), while  expression of p27 was significantly decreased (P<0.05). With the increasing of grades and stages in transitional cell carcinoma of renal pelvis,  expression rates of cyclin D1 and cyclin E increased while expression rate of p27 decreased (P<0.05), and p27 expression was negatively related with cyclin D1 and cyclin E expression respectively(P<0.05). Conclusion  Cyclin D1 and cyclin E over-expressions and p27 deactivation are found in transitional cell carcinoma of the renal pelvis. Combined detections of cyclin D1, cyclin E and p27 may help reveal the malignancy of transitional cell carcinoma and predict the prognosis.
