Expression of differential genes in placental vascular disease  and normal placenta through gene arrays

Abstract:

Objective  To further approach a new clue for the mechanism of placental vascular diseases(PVD) by performing a gene expression profiling study in placenvascular disease through gene array.Methods  We performed a gene expressionprofiling study of placental tissues from women with and without PVD by using 22k Human Genome Oligo Array and screened the different expressive genes between PVD and controls. Then real-time quantitative polymerase chain reaction (PCR) and Western-blot were used to verify  expression of some different genes. Results  By a comparative differential gene expression analysis between the two groups, we found 10 genes to be significantly altered in their expression levels in PVD. Among them 9 genes were up-regulated and 1 gene was down-regulated in PVD. Theverification test result was coincident with the gene array result. Conclusion  Placental vascular disease is an inflammatory disease characterized by activation of endothelial cells and immune response in the vessel wall. These genes can participate in endothelial activation and inflammatory response through activation of the transcription factors, which may result in the occurrence and development of PVD.
