Effect of  glutathione on proliferation of KLE and 
HEC1b endometrial cancer cells

Abstract:

To investigate the effects of glutathione on proliferation of typeⅠand Ⅱ endometrium cancer cells and to explore its related mechanism. Method① TypeⅠand Ⅱ endometrium cancer cells were challenged by GSH, and proliferation of cells was observed with MTT assay. ② TypeⅠor Ⅱ endometrium cancer cells were treated with 5?mmol/L GSH and the cellular ROS, GSH, and GSSG levels and SOD, CAT and GPx activities were determined. ResultsWithout treatment with GSH, typeⅠ endometrium cancer cells, compared with type Ⅱ endometrium cancer cells, had a lower ROS level and CAT activity and higher GSH level, GSH/GSSG ratio, and SOD and GPx activities. In the condition of treating the cells with reductive GSH, proliferation was promoted in typeⅠendometrium cancer cells while it was inhibited in typeⅡendometrium cancer cells. The ROS level was decreased in both typeⅠand Ⅱ endometrium cancer cells. The GSH level, GSH/GSSG ratio, and SOD, CAT and GPx activities were reduced in type Ⅰ endometrium cancer cells while the GSH and GSSG levels, and CAT and GPx activities were decreased and the GSH/GSSG ratio was markedly increased in type Ⅱ endometrium cancer cells. ConclusionGlutathione definitely differently affects proliferation of typeⅠand Ⅱ endometrium cancer cells, which is related to the ROS level and activities of antioxidative systems in two type of endometrium cancer cells.
