Effect of silencing Notch1 on proliferation of bladder cancer cell line T24

Abstract:

 To investigate the effect of silencing Notch1 gene on cell proliferation in vitro in bladder cancer cell line T24. MethodsThe siRNA eukaryotic expression vector of Notch1(psiRNA1)was constructed using pGenesil plasmid. psiRNA1 was transfected into bladder cancer cell line T24 by lipofectamine. Cell growth, cycle and apoptosis of T24 were determined with methabenzthiazuron (MTT) and flow cytometry (FCM) assay after transfection. The mRNA and protein expressions of Notch1 were determined by RTPCR and western blot. ResultsCompared with the control group, the growth curve of T24 cells was inhibited after transfection, which was timedependent during 60 hours. Notch1 silencing produced a cell cycle arrest at G0/G1(P<0.05). The apoptotic cell index also increased to some degree. mRNA and protein expressions of Notch1 were significantly   downregulated
compared with the controls after transfection(P<0.05).     Conclusion  Notch1 could inhibit the proliferation of bladder cancer cells by silencing expression of Notch1

, and it might act as a carcinogenic gene in bladder cancer. 
