RNAi of targeted against survivin induced apoptosis of laryngeal carcinoma cell line hep2

Abstract:

To study the inhibitory effects of RNAi expression vector suppressing expression of survivin and inducing the apoptosis of laryngeal carcinoma cell line Hep2. MethodsRNAi expression vectors targeting survivin were successfully constructed, and then were transfected into Hep2 cells with liposomes. Expression of survivin mRNA was detected and the rate of cell apoptosis was measured. ResultsThere was a high expression of survivin in Hep2 cells. The DNA sequence was in accordance with the survivin gene in the GENEBANK. The survivin expression was inhibited 68.5% by pGenesil/survivin RNAi expression vectors, and the apoptosis of cells increased. ConclusionThe RNAi expression vector can effectively inhibit survivin expression and induce the apoptosis in Hep2 cells.
