Effect of RAD001 on cell growth of gastric carcinoma cell lines MKN28, N87: an empirical study

Abstract:

To investigate the effect of RAD001 on growth of gastric carcinoma cell lines MKN28, N87 and to investigate its possible mechanism. MethodsGastric carcinoma cell lines were treated by RAD001(20nM), then cell coulter, flow cytometry, and westernblot array were used to detect the growth, cell cycle and protein expression of tumor cells. ResultsAfter being treated by RAD001, gastric carcinoma cells were inhibited in a timedependent manner. Percentage of Go phase was increased in the two cell lines. Also, expressions of phosphatep70S6K,S6 protein, and phosphateS6 proteins were decreased while mTOR, p70S6K and phosphate mTOR had no changes. ConclusionThe present study suggested that RAD001 inhibited the growth of gastric carcinoma cells by adjustment of the cell cycle via inhibition of phosphorylation of p70S6K and S6 protein, which is due to the downregulation of mTOR activity by integrating to its FRB domain instead of changing the mTOR and phosphate mTOR protein expressions.
