DNA damage of vascular endothelial cells caused by the serum of  patients with hypertension diseases

Abstract:

To explore the mechanism of  endothelial vascular cell(EVC) damage caused by the serum in patients with hypertension disease and to explore its relation to the genesis and development of essential hypertension (EH). MethodsEVCs were divided into the EH group, the healthy group and the blank group, and each group was respectively subjected to 15% of the serum of patients with hypertension disease, 15% of the serum of healthy persons, and 100% DMEM for 48?h. Injury of DNA was determined by single cell gel electrophoresis, content of MDA in damaged cells was measured by thlobarbiturie acid and activity of SOD was measured by the xanthosine oxidase method. ResultsThe data were analyzed by SPSS11.0. Compared with the healthy group and the blank group, cells of the EH group had remarkable DNA damage, increased MDA and decreased SOD activities. ConclusionDamage factors in the serum of EH patients may cause DNA damage of EVC, and the damage may be related to the occurrence and development of EH.
