Effect of Reserpine on VMAT2CHO cells to resist  toxicity of Dopamine and Rotenone

Abstract:

To study the effect of Reserpine on VMAT2 transgenic CHO(VMAT2CHO) cells to resist Dopamine and Rotenone toxicity. MethodsMTT assay was used to detect viability of VMAT2CHO with different concentrations of Rotenone and Dopamine, and FCM was used to determine reactive oxygen species of the cytoplasm with different concentrations of Dopamine. ResultsThe result indicated that viability of VMAT2CHO was decreased with an increase of concentrations of rotenone. ROS was increased with an increase of concentrations of Dopamine. Reserpine could raise toxicity of Dopamine, which decreased the viability of VMAT2CHO and ROS was increased by the same concentration of Dopamine. ConclusionEndogenous toxin triggered by inhibition of VMAT2 may have its effect through oxidative stresses. Endogenous toxicity triggered by Dopamine may raise toxicity of Rotenone.
