Preliminary analysis of side population phenotypes in the human gastric cancer cell line BGC-823

Abstract:

Objective〓〖WTBZ〗To explore side population (sp) phenotypes in the gastric cancer cell line BGC-823 and to observe the morph of SP cells and the relationship between SP phenotype and stem cell marker ABCG2. 〖WTHZ〗Methods〓〖WTBZ〗Adherent BGC-823 cells cultured in culture flasks were determined by fluorescence microscopy after being stained with Hoechst33342, and the percentage of SP cells was determined. The adherent cells were mounted on a cover slip and stained with Hoechst 33342, and subsequently stained with mouse anti-human antibody and fluoresence antibody labeled by FITC. The staining of Hoechst33342 and expression of ABCG2 were determined by fluorescence microscopy. 〖WTHZ〗Results〓〖WTBZ〗A SP phenotype was found in the BGC-823 cell line, and it was small and occupied about 1.8% of all tumor cells, and SP cells could express the ABCG2 phenotype. 〖WTHZ〗Conclusion〓〖WTBZ〗SP cells exist in the gastric cancer cell line BGC-823 and can express the stem cell marker ABCG2.
